Abstract: Background: Ziziphus jujubaMill var. spinosa seeds (ZSS) is one of the most popular traditional Chinese herbs. It shows several pharmacological effects, such as anti-anxiety, antidepressant, neuroprotection, and cardiotonic, and prevents insomnia. The primary biologically active flavonoids derived from ZSS are 6´´´-feruloyl spinosin, 6´´´-p-coumaroyl spinosin, spinosin, swertisin, and isovitexin. Objectives: The primary objective of this study was to investigate the mechanism of degradation of co-cultured flavonoids, namely 6´´´-feruloyl spinosin, 6´´´-p-coumaroyl spinosin, spinosin, swertisin, and isovitexin, in the intestinal flora of rats under in vitroconditions. Materials and Methods: In this study, we co-cultured the five flavonoids in the intestinal flora of rats under in vitro conditions and determined the degradation of these five flavonoids via high-performance liquid chromatography with tandem mass spectrometry (MS/MS). Results: The degradation rate of the 6´´´-feruloyl spinosin, 6´´´-p-coumaroyl spinosin, and swertisin was affected by the concentration of the sample and conforms to the first-order kinetic model. According to our results, the 6´´´-feruloyl spinosin and 6´´´-p-coumaroyl spinosin may be degraded to spinosin and swertisin, and spinosin continued to decompose into swertisin. These results were consistent with their individual experiments. Moreover, by comparing the structures of isovitexin and swertisin, isovitexin was first found to be the product of the seventh demethylation reaction of the flavonoid core structure, demonstrating that isovitexin is a metabolite of ZSS flavonoids following spinosin and swertisin. Conclusion: Taken together, the results of this study explain the metabolic and interrelation of these five main flavonoids in vitro.

Abstract: The invention provides a method for extraction and separation of isovitexin The method comprises the following steps of subjecting Desmodium styracifolium to reflux extraction by utilizing ethyl alcohol so as to obtain a Desmodium styracifolium extract; and subjecting the Desmodium styracifolium extract to multistage chromatographic separation by utilizing an alcohol system, wherein the step of multistage chromatographic separation comprises the following sub-steps of subjecting the Desmodium styracifolium extract to a first chromatographic separation by utilizing a first alcohol system so as to obtain a first separation part, subjecting the first separation part to a second chromatographic separation by utilizing a second alcohol system so as to obtain a second separation part, and subjecting the second separation part to a third chromatographic separation by utilizing a third alcohol system so as to obtain the isovitexin The invention also provides a system for implementing the above-mentioned method By utilizing the method and/or the system for extraction and separation of the isovitexin from the Desmodium styracifolium, process operation is simple; production time is short; product yield is high; purity is high; and the isovitexin with a purity reaching to 997% can be obtained

Abstract: Streptocarpus x hybridus is a very popular houseplant with colourful flowers arousing thus the interest of plant breeders. Its potential value is even higher since different chemical compounds could be identified and used for food and pharmaceutical industries. Therefore, the objectives of this study were: to determine genetic relationships between twelve F1 Streptocarpus hybrids and their genitors; to determine the total phenolic and flavonoid content; to evaluate the antioxidant activity of the analysed individuals. Although 12 out of the 24 RAPD primers detected polymorphism among the individuals, the UPGMA dendrogram did not show a very clear grouping pattern for flower colour. In contrast, the phytochemical dendrogram shed more light on the dissimilarities among Streptocarpus flowers in terms of their polyphenolic content. The total phenolic and flavonoid content of the flower extracts determined by the Folin-Ciocalteau and aluminium chloride colorimetric methods ranged from 680.02 to 2360.57 mg of GAE/100 g FW and from 12.52 to 78.36 mg QE/100 g FW. The radical scavenging activity against ABTS ranged from 27.17 to 130.35 µM Trolox/g FW. Due to its ornamental value, H12 was screened by HPLC-PDA-ESI-MS to identify and quantify the chemical compounds involved in flower colour. Among the six compounds that were identified and quantified (i.e. cyanidin-3-O-galactoside, cyanidin-3-O-xyloside, cyanidin-3-O-glucoside, cyanidin-3-O-arabinoside, peonidin-3-O-glucoside and apigenin-6-C-glucoside), isovitexin reached the highest concentration 4183 µg rutin/100 g FW. These preliminary results, showing high total polyphenolic content and ABTS antioxidant activity, suggest that Streptocarpus flowers could be a valuable source of antioxidants.

Abstract: From the methanolic extract of the fruits of Gleditschia australis Hemsl., a new flavonoid derivative 3"-O-menthiafoloylisovitexin (1) and a new carbohydrate ester of cinnamic acid 1-O-E-cinnamoyl-[2-O-E-cinnamoyl-alpha-L-rhamnopyranosyl-(1-->6)-beta-D-glucopyranoside] (2) have been isolated along with four known compounds, 1-O-E-cinnamoyl-[3-O-E-cinnamoyl-alpha-L-rhamnopyranosyl-(1-->6)-beta-D-glucopyranoside], isovitexin, luteolin, and quercetin. Their structures were elucidated on the basis of physical and spectroscopic evidence.
