Abstract: Cotyledons of mature seeds of Vigna radiata were found to be variable in their response to N 6-benzytadenine, kinet in and zeatin. The two cotyledons of a seed were designated as CE and C; where CE referred to the cotyledon that remained closely attached to the embryonal axis, and the other more loosely attached cotyledon was referred to as C. Shoots formed from the proximal end of both explants in all nine cultivars studied. Shoot regeneration was faster and regeneration efficiency was higher in CE explants than in C explants in these cultivars. BA was found to be the most suitable cytokinin for both multiple shoot induction and regeneration.

Abstract: In anther cultures of Datura innoxia the addition of growth hormones, such as auxins, gibberellins, and cytokinins, to the culture medium enhanced the production of pollen embryoids. Cytokinins appeared to be the most effective and, among the four cytokinins tested, zeatin and kinetin gave the best results. Generally speaking, combinations of hormones did not improve the response over that of an individual hormone. The number of embryoids per anther varied in the same medium and did not strictly correlate with the percentage of respond ing anthers.

Abstract: The effects of abscisic acid and kinetin on RNA synthesis in senescing radish leaf disks were investigated using the improved resolution afforded by polyacrylamide gel electrophoresis. Kinetin stimulated and abscisic acid inhibited incorporation of radioactivity into cytoplasmic ribosomal RNA and soluble RNA. Chloroplast ribosomal RNA synthesis appeared to be confined to the period of leaf expansion and was not detected in fully mature leaves. The effects of kinetin in retarding and of abscisic acid in accelerating leaf senescence were not altered by the inhibition of cytoplasmic ribosomal RNA synthesis with 5-fluorouracil. Following inhibition of cytoplasmic ribosomal RNA synthesis with 5-fluorouracil, kinetin stimulated and abscisic acid inhibited incorporation of radioactivity into polydisperse RNA. These results are discussed in relation to the possible mode of action of kinetin and abscisic acid in senescing leaf tissue.

Abstract: To improve plant regeneration from oat (Avena sativa L.) anther culture, the effects of induction medium supplements and culture conditions were studied. Significantly better plant regeneration rates were obtained with cultivars Lisbeth (naked type) and Aslak when a medium containing W14 salts and vitamins, supplemented with 2,4-d, BAP, Ethephon, l-cysteine and myo-inositol, was used for induction in the dark compared with a medium containing only 2,4-d and kinetin. Genotypes reacted differently on the light during the induction phase. Use of dim light significantly decreased the green plant regeneration rates in cv. Lisbeth, while in cv. Aslak the difference was not so clear. Up to 30 green plants per 100 anthers were recovered from Aslak × Lisbeth progeny and in total, over 500 oat regenerants were produced. With these numbers, acceptable rates of DH-production for cultivar breeding and genetic study purposes are approached. The agronomic performance of some DH lines was compared with that of the plants derived from commercial seeds of the same cultivars in the field experiment. A few differences were found, but generally DH lines yielded the same or more as the commercial cultivars.

Abstract: The present study investigated the effect of various concentrations and combinations of plant growth regulators on callus induction, shoot proliferation and root regeneration of three potato (Solanum tuberosum L.) cultivars by using potato nodal and leaf explants under long-day conditions. The minimum days for callus induction (8.33 d), the maximum callus formation percentage (87.50%), the largest callus diameter (1.90 cm) and the maximum callus weight (2.04 g) were recorded on 1.0 × Murashige and Skoog (MS) medium containing 3.0 mg L−1 benzyl amino purine (BAP) + 2.0 mg L−1 naphthalene acedic acid (NAA). Nodal explants demonstrated a better performance for callus induction compared to leaf segments.
