Abstract: Zygotic embryos excised from immature female cones of Araucaria angustifolia were cultured to induce embryonal-suspensor masses. The conditions for the establishment and multiplication of embryogenic cultures and formation of early somatic embryos were investigated. Cultures consisted of embryogenic and filamentous cells, the latter similar to the suspensor cells. Higher induction rates of embryonal-suspensor cells were obtained with 2,4-D (45 μM), casein acid hydrolysate (500 mg.L -1 ), BAP and Kin (11 μM each), depending on the developmental stage of the explants. Repetitive cycles of cell division were achieved both by cell plating and suspension cultures in LP basal medium supplemented with 7 μM 2,4-D plus 2 μM BAP and Kinetin. The growth dynamic showed a 3.5 fold increase in packed cell volume within 30 days. The progression to proembryonal complex was observed when embryonal- suspensor masses were transferred to basal LP medium supplemented with 1% PEG 8.000 regardless of ABA presence. Somatic proembryos did not develop to cotyledonary stage in any of the maturation treatments used. Abstract: Sorghum is a wonder crop from physiological point of view. It is the most important cereal crop, after rice and wheat. The number of reports describing the use of transgenic Sorghum for basic studies in Biotechnology is still limited when compared to other crops. In one hand, the success of the transformation techniques is mainly dependent upon the availability of optimal protocols for highly efficient tissue culture techniques. On the other hand, regeneration in Sorghum is difficult. Hence, in this study an efficient and reproducible method for in vitro development of embryogenic callus and regeneration in Sorghum bicolor was developed. Immature embryo explants of Sorghum bicolor were cultured on MS nutrient medium supplemented with different concentrations and combinations of auxins and cytokinins. Embryogenic callus was initiated on MS medium supplemented with 2 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) and 0.5 mg/l Kinetin (KN). Addition of KN to MS medium containing 2,4-D resulted in a significant enhancement on embryogenesis of the embryos. Amino acids also supported an improved frequency of embryogenesis. Therefore induction of a high frequency of somatic embryogenesis in immature embryos on MS medium is possible. Development of embryogenic callus, induction of somatic embryo and subsequent shoot regeneration was proficient at a concentration of 2 mg/l BAP. The regenerated shoots readily rooted on half strength MS medium supplemented with 1 mg/l naphthalene acetic acid (NAA). The regenerated plantlets were successfully transferred to soil and subsequently plants produced seeds. There was no difference between the acclimatized plants in comparison with in vivo plants in the respect of morphological characters.

Abstract: Fenugreek ( Trigonella foenum-graecum L.) is an important vegetable, spice and medicinal legume plant used as fresh and dried leaves and seeds in many parts of the world. Fenugreek seeds contain polysaccharide galactomannan and different saponins and have anti-microbial activity. The study presents an efficient shoot regeneration protocols from 8 - 10 days old in vitro grown cotyledon node explants cultured on Murashige and Skoog (MS) medium supplemented with 0.05 - 0.80 mg/l kinetin, 0.25 - 1.0 mg/l 6 benzyladenine (BA) with 0 and 0.20 mg/l α naphtalene acetic acid (NAA) and 0.05 - 0.80 mg/l thidiazuran (TDZ) with 0 and 0.10 mg/l indole 3 butyric acid (IBA). All culture mediums were solidified with 0.22% gelrite. Maximum shoot regeneration and number of shoots per explant were recorded on MS medium supplemented with TDZ with or without IBA. Maximum of 22.21 shoots per explant were recorded on MS medium containing 0.40 mg/l TDZ. Presence of auxins in the culture medium positively increased the mean shoot length. Regenerated shoots were transferred to rooting media containing 0.1 - 1.0 mg/l IBA or NAA. Key words : Fenugreek , cotyledon node, hypocotyl, shoot regeneration.
