Abstract: A callus induction and in vitro plantlet regeneration system for the endangered state flower of Uttaranchal (Saussurea obvallata) was optimized by studying the influence of explant type (root, hypocotyl, cotyledon and leaf), age and different concentrations of plant growth regulators. Explants from 10 to 15-day-old seedlings showed maximum callus induction. Callus formation and shoot differentiation was initiated on Murashige-Skoog (MS) medium containing 6-benzyladenine (BA) and α-naphthalene acetic acid (NAA) in all explant types. The best results were obtained using leaf explants: 100% callusing was achieved in MS medium supplemented with 2.5 μM BA and 1.0 μM NAA, and 100% differentiation along with a multiplication rate of 12 shoots per explant with a combination of 5.0 μM BA and 1.0 μM NAA. However, the results reflected the existence of high inter-explant variability in response to growth regulators. In vitro rooting of shoots was achieved at an efficiency of 100% in one-half strength MS medium supplemented with 2.5 μM indole-3-butyric acid. Application of this protocol has potential for mass multiplication of the target species in a limited time period.

Abstract: Morphogenetic potential of root, leaf, node and internode expiants of 3 cultivated Piper species was investigated to develop a reliable plant regeneration protocol. P. longum (pipli) was the most responsive followed by P. betle (betel vine) and P. nigrum (black pepper). In P. longum the highest number of shoot buds was produced on root expiants followed by node, internode and leaf expiants. In P. betle and P. nigrum adventitious shoot buds differentiated only from internodal and nodal ring regions, respectively. Histological examination in P. longum showed that adventitious shoot buds originate directly from the cortical cells of the root and the internode without an intervening callus phase. Benzyladenine was superior to kinetin for shoot induction and its optimum concentrations for P. longum, P. betle and P. nigrum were 1–2, 10 and 10 μM, respectively. Shoot elongation and rooting were achieved in B5 medium containing 0.5 μM benzyladenine and 1 μM indoleacetic acid, respectively. Regenerated plants were established in soil.

Abstract: Hypocotyl explants of Catharanthus roseus produced hairy roots when cultured on Murashige and Skoog (MS) basal medium after infection by Agrobacterium rhizogenes. Explants gave rise to adventitious shoots at a frequency of up to 80% when cultured on MS medium supplemented with 31.1 μM 6-benzyladenine and 5.4 μM α-naphthaleneacetic acid. There was a significant difference in the frequency of adventitious shoot formation for each hairy-root line derived from a different cultivar. Plants derived from hairy roots exhibited prolific rooting and had shortened internodes. Approximately half of the plants had wrinkled leaves and an abundant root mass with extensive lateral branching, but otherwise appeared morphologically normal. Plants with hairy roots that were derived from the cultivar Cooler Apricot developed flowers with petals that were white in the proximal region, whereas the wild-type flower petals are red. PCR and Southern blot analyses revealed that plants derived from hairy roots retained the Ri TL-DNA.
