Abstract: A highly sensitive and specific “rocket” immunoassay was used to measure the content of an endo-type β-1,3-glucanase (EC 3.2.1.39) in tissues of Nicotiana tabacum L. cv. Havana 425. We show that the accumulation of β-1,3-glucanase in cultured pith-parenchyma tissue is blocked by combinations of the auxin, α-naphthaleneacetic acid (NAA), and the cytokinin, kinetin. When tissues pre-incubated for 7 d on complete medium containing 2.0 mg·l-1 NAA and 0.3 mg·l-1 kinetin are transferred onto medium without hormones or with either hormone added separately, the β-1,3-glucanase content expressed per mg soluble protein increases approx. ten fold over a 7-d period. Under these inductive conditions, up to approx. 5% of the soluble protein is β-1,3-glucanase. The induction is inhibited by >90% when tissues are cultured over the same period on medium containing both hormones. This β-1,3-glucanase is developmentally regulated in the intact plant. It is a major component of the soluble protien in the lower leaves and roots but is not detectable in leaves near the top of the plant.

Abstract: After 2 weeks in the dark, leafy shoots were initiated on subcultured callus tissue of triploid quaking aspen on Wolter's medium without auxin and with 6-benzylaminopurine at 0.05, 0.10., 0.15, or 0.20 milligram per liter substituted for kinetin. Shoots transferred to Wolter's medium under light (323 milliphots) grew large roots and were isolated as plantlets.

Abstract: Multiple shoot cultures of Withania somnifera were established from single shoot tip explants and their potential for the production of two principle withanolides, withaferin A and withanolide D was investigated. Shoot tips grown on MS medium supplemented with BA (1 mg l-1) induced 10.0 ± 1.15 microshoots per explants and shoot cultures accumulated both withanolides (withaferin A = 0.04 %, withanolide D = 0.06 %). Supplementation of MSSM (solid) agar medium with 4 % sucrose enhanced accumulation of both withaferin A (0.16 %) and withanolide D (0.08 %). Reduction of the agar concentration to 0.16 % increased the number of microshoots induced per explant to 25.5. MSSM liquid medium containing 10 % coconut milk favoured a maximum increase in biomass (27 fold); number of microshoots induced (37.6 ± 1.45) as well as accumulation of withaferin A (0.14 %).
