Abstract: Rice cells derived from PI 353705 (similar to Assam 5) were isolated from anthers cultured on Blaydes medium containing IAA, 2,4-D, kinetin, yeast extract, and coconut milk. Isolated aggregates of cells were plated on a modified Blaydes medium containing 10−3 M S-aminoethyl-l-cysteine (S-AEC). This level ofS-AEC inhibits nonselected wild type cells. Cells or aggregates of cells resistant to this analog of lysine were subcultured three times in the presence of 2×10−3 M S-AEC. The selected cells were then placed on a Murashige-Skoog (MS) regenerating medium containing 1 mg/l each of IAA and kinetin. Ten plants were recovered from 34 selected cell lines, three plants grew to maturity, and two produced seeds. Seeds from plants regenerated from cells in culture had higher lysine than the original field controls and had increased levels of free alanine, arginine, and asparagine. The in vitro selection produced plants with higher protein than the field controls. Plant breeders have begun to evaluate the genotype recovered from in vitro selection.

Abstract: Single applications of either 6-benzyladenine or zeatin to inhibited axillary buds of intact, light-grown seedlings of Pisum sativum L. cv Black-eyed Susan, resulted in the formation of rapidly elongating, leafy shoots. Similar treatment with kinetin or isopentenyladenine caused only limited but outgrowth which stopped 6 days after application.

Abstract: Multiple shoots were induced from shoot tips and nodal segments of a 12-year-old tree of Cinnamomum camphora on Woody Plant Medium (WPM) supplemented with BA and kinetin. The nodal segments from the in vitro developed plantlets could be induced again to produce a large number of harvestable shoots. Harvested shoots were rooted in vitro in WPM supplemented with activated charcoal (AC) and IBA. The plantlets were transferred to thermocol cups after which they were replanted into polybags and then to field. These plants survived with over 90% success under field conditions and exhibited vigorous growth. This system could be utilized for large-scale multiplication of C. camphora by tissue culture.

Abstract: Various factors such as browning, season, media type and plant growth regulators influence the micropropagation of female trees of Myrica esculenta . These factors have successfully been addressed after addition of some media additives, collection of the explant at right season, standardizing the media type and use of plant growth regulators at varying concentrations. Polyvinylpyrollidone (PVP - 0.5%) was found effective for successful partial removal of phenolic compounds and obtaining maximum percent survival of explants. The explant collection season played an important role in reducing phenol induced browning and winter season was found best for explant establishment. The maximum number of shoots (4-5/explant) was obtained in Woody Plant Medium (WPM) supplemented with 10µM kinetin and 0.1µM naphthalene acetic acid (NAA). Of all the media types tried, Woody Plant Medium was found to be the best. Kinetin was found superior to benzyl amino purine and N 6 (γ,γ-dimethylallyamino)purine (2iP) for explant establishment and multiplication. NAA induced rooting to 45.8% of explants in 1/2 strength Woody Plant Medium. However, ex vitro survival percentage was low.
