Abstract: Our goal in this research was to obtain lead oxide nano-powders by sol–gel method. In this method, lead oxide nano-powders were synthesized through the reaction of citric acid (C6H7O8·H2O) solution and lead acetate [Pb(C2H3O2)2] solution as stabilizer and precursor, respectively. The effect of different parameters including calcination temperature, (molar ratio of citric acid to lead acetate) and drying conditions were investigated. The prepared lead oxide nano-powders were characterized by FT-IR spectroscopy, X-ray diffraction, thermogravimetric analysis and scanning electron microscopy. The prepared PbO samples consist of the particles in the range of 50–120 nm or the thick plate like structures with thickness of 53 nm depending on the drying conditions.

Abstract: Lead (Pb) enhances the production of reactive oxygen species and depletes the antioxidant molecules that cause tissue damage. In the current study, we investigated the protective effect of Indigofera oblongifolia (hasr in Arabic) against lead acetate-induced reproductive toxicity in rats. Exposure of rats to lead acetate (PbAc; 20 mg/kg body weight; intraperitoneal injection) induced a significant change in both of body weight loss and the relative testis weight. Furthermore, a significant increase in lipid peroxidation and nitric oxide and a marked depletion of glutathione were evident in the testis of the PbAc group compared to the control group. Also, PbAc significantly reduced the activity of antioxidant enzymes. Pre-administration of I. oblongifolia leaves extract (IOLE; 100 mg/kg body weight) to the PbAc-treated rats restored most of the parameters mentioned above to near-normal levels. Additionally, pretreatment of animals with IOLE accompanied with a significant decrease in the toxic effects of PbAc as shown by caspase-3 and Bax expressions and prevented the histological injury in the testis. On the basis of the above results, I. oblongifolia appeared to be a promising agent for protection against lead-induced oxidative damage and apoptosis in the testis of rat.

Abstract: Sprague‐Dawley rats were injected sc with lead acetate in suspension, or with sodium acetate (controls) on the d 9 of pregnancy and every 3–4 d thereafter until the pups were 13 or 21 d of age. At termination, testicular homogenates or isolated Sertoli cells were used to study steroidogenesis and gonadotropin binding. Lead had no significant effect on the mother's water and food consumption, on the pup's body or testis weights, on the number of pups and the time of birth, and on the seminiferous tubule diameter. Homogenates of testes of the lead‐treated group converted significantly less (p <0.01) labeled progesterone (14Cor 3H) to 5α‐pregnane‐3,20‐dione, 3α‐hydroxy‐5α‐pregnan‐20‐onet 17β‐hydroxy‐5α‐androstan‐3‐one (DHT), 3(α,β))‐hydroxy‐5α‐androstan‐17‐one, testosterone/17α‐hydroxyprogesterone, and androstenedione. Sertoli cells from lead‐treated animals converted significantly less (p < 0.01) progesterone to 5α‐pregnane‐3α,20α‐diol, 3α‐hydroxy‐5α‐pregnan‐20‐one, DHT, and 20α‐hydroxy‐4‐pregnen‐3‐one. The...
