Abstract: Acinetobacter radioresistens KA53, isolated by enrichment culture, was found to produce an extracellular, nondialyzable emulsifying agent (referred to as alasan) when grown on ethanol medium in a batch-fed reactor. The crude emulsifier was concentrated from the cell-free culture fluid by ammonium sulfate precipitation to yield 2.2 g of emulsifier per liter. Alasan stabilized a variety of oil-in-water emulsions, including n-alkanes with chain lengths of 10 or higher, alkyl aromatics, liquid paraffin, soybean and coconut oils, and crude oil. Alasan was 2.5 to 3.0 times more active after being heated at 100 degrees C under neutral or alkaline conditions. Emulsifying activity was observed over the entire pH range studied (pH 3.3 to 9.2), with a clear maximum at pH 5.0. Magnesium ions stimulated the activity both below (pH 3.3 to 4.5) and above (pH 5.5 to 9.3) the pH optimum. Alasan activity was higher in 20 mM citrate than in 20 mM acetate or Tris-HCl buffer. Preliminary chemical characterization of alasan indicated that it is a complex of an anionic, high-molecular-weight, alanine-containing heteropolysaccharide and protein.

Abstract: 1. Single Malpighian tubules of the stick insect have been studied as preparations isolated in drops of haemolymph under liquid paraffin. Measurements of osmotic pressure have been made on haemolymph and urine. 2. The urine is more or less isotonic with the haemolymph over a wide (45-145%) range of osmotic pressure. But in a majority of cases the urine is slightly but significantly hypotonic to the haemolymph. 3. From these and other observations it is concluded that water must be actively transported across the wall of the tubule. The implications of this conclusion are discussed. 4. The normal rate of urine production is of the order of 0.07 x 1o-3 cu.mm./sq.mm./min. The rate increases with increasing dilution of the haemolymph and vice versa. 5. Urine production normally ceases, probably as a result of distension of the tubule, when the pressure in the lumen rises to about 20 cm. H2O.

Abstract: Laser trapping of a metal particle in water or a water droplet in liquid paraffin, which cannot be attained by irradiation of a TEM00 mode focused laser beam, was experimentally confirmed based on a scanning laser trapping technique. Although a metal particle or a water droplet experiences repulsive radiation force from a laser beam (1064 nm, focused into a ∼1 μm spot), scanning of the laser beam circularly around the particle was successful to optically trap and tweezer the particle. Water and ethylene glycol droplets dispersed in liquid paraffin were also shown to be manipulated independently by scanning double laser‐beam trapping.
