Abstract: Bionanotechnology involves the use of biomolecules to control both the structure and property of nanomaterials. One of the most studied examples is DNA-directed assembly of inorganic nanoparticles such as gold nanoparticles (AuNPs). However, systematic studies on DNA-linked soft nanoparticles, such as liposomes, are still lacking. We herein report the programmable assembly and systematic characterization of DNA-linked liposomes as a function of liposome size, charge, fluidity, composition, DNA spacer, linker DNA sequence, and salt concentration for direct comparison to DNA-directed assembly of AuNPs. Similar to the assemblies of AuNPs, sharp melting transitions were observed for liposomes where the first derivative of the melting curve full width at half-maximum (fwhm) is equal to or less than 1 °C for all of the tested liposomes, allowing sequence specific DNA detection. Abstract: Nanoparticles serve as fundamental building blocks for nanobiotechnology, especially in several applications in the development of novel (bio)sensing systems. Nanoparticles can be used for modification of the surfaces of (bio)sensing transducers or as optical or electroactive labels to improve different aspects of performance, for example sensitivity, detection limit, multidetection capability, and response stability. Nanoparticles can be integrated into the transducer materials on an individual basis or inside other matrices to ensure the immobilization of recognition biomolecules and/or receptors which are the principal components of the (bio)sensing systems. Incorporation of nanoparticles into optical and electrochemical (bio)sensing systems, including their use in microfluidic based systems has the advantages of enabling the design of robust, easy to use, portable, and cost-effective devices.

Abstract: The ability to tune the size, shape, and composition of nanomaterials at length scales <10 nm remains a challenging task. Such capabilities are required to fully realize the application of nanotechnology for catalysis, energy storage, and biomedical technologies. Conversely, nature employs biomacromolecules such as proteins and peptides as highly specific nanoparticle ligands that demonstrate exacting precision over the particle morphology through controlling the biotic/abiotic interface. Here we demonstrate the ability to finely tune the size, surface structure, and functionality of single-crystal Pd nanoparticles between 2 and 3 nm using materials directing peptides. This was achieved by selectively altering the peptide sequence to change the binding motif, which in turn modifies the surface structure of the particles.
