Abstract: Nanosized crosslinked polymer networks, named as nanogels, are playing an increasingly important role in a diverse range of applications by virtue of their porous structures, large surface area, good biocompatibility and responsiveness to internal and/or external chemico-physical stimuli. Recently, a variety of carbon nanomaterials, such as carbon quantum dots, graphene/graphene oxide nanosheets, fullerenes, carbon nanotubes, and nanodiamonds, have been embedded into responsive polymer nanogels, in order to integrate the unique electro-optical properties of carbon nanomaterials with the merits of nanogels into a single hybrid nanogel system for improvement of their applications in nanomedicine. A vast number of studies have been pursued to explore the applications of carbon-based hybrid nanogels in biomedical areas for biosensing, bioimaging, and smart drug carriers with combinatorial therapies and/or theranostic ability. New synthetic methods and structures have been developed to prepare carbon-based hybrid nanogels with versatile properties and functions. In this review, we summarize the latest developments and applications and address the future perspectives of these carbon-based hybrid nanogels in the biomedical field.

Abstract: Superparamagnetic iron oxide nanoparticles (SPIONs) have been receiving great attention lately due to their various biomedical applications, such as in MR imaging and image guided drug delivery. However, their systemic administration still remains a challenge. In this study, the ability of biodegradable thermosensitive polymeric micelles to stably encapsulate hydrophobic oleic-acid-coated SPIONs (diameter 5−10 nm) was investigated, to result in a system fulfilling the requirements for systemic administration. The micelles were composed of amphiphilic, thermosensitive, and biodegradable block copolymers of poly(ethylene glycol)-b-poly[N-(2-hydroxypropyl) methacrylamide dilactate] (mPEG-b-p(HPMAm-Lac2)). The encapsulation was performed by addition of one volume of SPIONs in THF to nine volumes of a cold aqueous mPEG-b-p(HPMAm-Lac2) solution (0 °C; below the cloud point of the polymer), followed by rapid heating of the resulting mixture to 50 °C, to induce micelle formation (“rapid heating” procedure). Abstract: Nanooncology, the application of nanobiotechnology to the management of cancer, is currently the most important chapter of nanomedicine. Nanobiotechnology has refined and extended the limits of molecular diagnosis of cancer, for example, through the use of gold nanoparticles and quantum dots. Nanobiotechnology has also improved the discovery of cancer biomarkers, one such example being the sensitive detection of multiple protein biomarkers by nanobiosensors. Magnetic nanoparticles can capture circulating tumor cells in the bloodstream followed by rapid photoacoustic detection. Nanoparticles enable targeted drug delivery in cancer that increases efficacy and decreases adverse effects through reducing the dosage of anticancer drugs administered. Nanoparticulate anticancer drugs can cross some of the biological barriers and achieve therapeutic concentrations in tumor and spare the surrounding normal tissues from toxic effects. Nanoparticle constructs facilitate the delivery of various forms of energy for noninvasive thermal destruction of surgically inaccessible malignant tumors. Nanoparticle-based optical imaging of tumors as well as contrast agents to enhance detection of tumors by magnetic resonance imaging can be combined with delivery of therapeutic agents for cancer. Monoclonal antibody nanoparticle complexes are under investigation for diagnosis as well as targeted delivery of cancer therapy. Nanoparticle-based chemotherapeutic agents are already on the market, and several are in clinical trials. Personalization of cancer therapies is based on a better understanding of the disease at the molecular level, which is facilitated by nanobiotechnology. Nanobiotechnology will facilitate the combination of diagnostics with therapeutics, which is an important feature of a personalized medicine approach to cancer.
