Abstract: Background:Optical coherence tomography can be used to assess retinal degeneration in multiple sclerosis (MS). Thinning of the retinal nerve fibre layer and macular thickness have been well characterized, but newer devices allow quantification of all retinal layers.Objectives:The objective of this study was to evaluate the thickness of the paramacular retina, peripapillary retinal nerve fibre layer, and deeper paramacular layers in MS patient subgroups, using state-of-the-art optical coherence tomography.Methods:Using a Heidelberg Engineering Spectralis device, we performed paramacular volumetric retinal scans and circular peripapillary fibre-layer scans, manually segmenting different retinal layers into single horizontal foveal scans in 95 patients with definite MS (42 relapsing–remitting, 41 secondary progressive, 12 primary progressive), plus 91 age- and sex-matched controls.Results:Even without a history of optic neuritis, all MS subgroups had significant thinning of the peripapillary retinal nerve fi...

Abstract: In recent years, superluminescent diodes (SLDs) have gained increasing importance as light sources for partial coherence interferometry and optical coherence tomography. The requirements of SLDs are high spatial coherence, low temporal coherence, and, for some applications, high power. Since there might be a trade-off between these properties we built an instrument for measuring the spatial coherence of SLDs. This instrument consists of a hybrid bulk optic-fibre optic Mach-Zehnder interferometer. The special advantages of the instrument are the ability to measure the spatial coherence between arbitrary points within the light beam and to observe directly the measurement points within the beam which facilitates the alignment. We used this instrument to evaluate the spatial coherence of several commercially available SLDs. As expected, a single mode fibre pigtailed SLD shows the best spatial coherence. Abstract: The frequency-sampling method is widely used to accommodate nonlinear laser tuning in swept-wavelength interferometric techniques such as optical frequency domain reflectometry (OFDR) and swept-wavelength optical coherence tomography (OCT). In this paper we analyze the frequency-sampling method and identify two sources of sampling errors. One source of error is the limit of an underlying approximation for long interferometer path mismatches and fast laser tuning rates. A second source of error is transmission delays in data acquisition hardware. We show that the measurement system can be configured such that the two error sources cancel to second order. We present experimental verification of sampling error correction using a general swept-wavelength interferometer with a significantly nonlinear laser sweep.

Abstract: We have been investigating ultrahigh resolution optical coherence tomography using supercontinuum. Low noise, Gaussian like supercontinua were generated at five wavelength regions. Ultrahigh resolution cross-sectional and three dimensional imaging were demonstrated for biomedical tissues and materials.
