Abstract: This study compared the synergistic effects of elevated pCO2 and temperature on the early life history stages of two ecologically and economically important oysters: the Sydney rock oyster, Saccostrea glomerata and the Pacific oyster, Crassostrea gigas. Gametes, embryos, larvae and spat were exposed to four pCO2 (375, 600, 750, 1,000 µatm) and four temperature (18, 22, 26, 30°C) levels. At elevated pCO2 and suboptimal temperatures, there was a reduction in the fertilization success of gametes, a reduction in the development of embryos and size of larvae and spat and an increase in abnormal morphology of larvae. These effects varied between species and fertilization treatments with S. glomerata having greater sensitivity than C. gigas. In the absence of adaptation, C. gigas may become the more dominant species along the south-eastern coast of Australia, recruiting into estuaries currently dominated by the native S. glomerata.

Abstract: In Korea, soils adjacent to abandoned mines are commonly contaminated by heavy metals present in mine tailings. Further, the disposal of oyster shell waste by oyster farm industries has been associated with serious environmental problems. In this study, we attempted to remediate cadmium (Cd)- and lead (Pb)-contaminated soils typical of those commonly found adjacent to abandoned mines using oyster shell waste as a soil stabilizer. Natural oyster shell powder (NOSP) and calcined oyster shell powder (COSP) were applied as soil amendments to immobilize Cd and Pb. The primary components of NOSP and COSP are calcium carbonate (CaCO3) and calcium oxide (CaO), respectively. X-ray diffraction, X-ray fluorescence and scanning electron microscope analyses conducted in this study revealed that the calcination of NOSP at 770°C converted the less reactive CaCO3 to the more reactive CaO. The calcination process also decreased the sodium content in COSP, indicating that it was advantageous to use COSP as a liming material in agricultural soil. After 30 days of incubation, we found that the 0.1 N HCl-extractable Cd and Pb contents in soil decreased significantly as a result of an increase in the soil pH and the formation of metal hydroxides. COSP was more effective in immobilizing Cd and Pb in the contaminated soil than NOSP. Overall, the results of this study suggest that oyster shell waste can be recycled into an effective soil ameliorant.

Abstract: Bivalve molluscan shellfish, such as oysters, filter large volumes of water as part of their feeding activities and are able to accumulate and concentrate different types of pathogens, particularly noroviruses, from fecal human pollution. Based on our previous observation of a specific binding of the Norwalk strain (prototype norovirus genogroup I) to the oyster digestive tract through an A-like carbohydrate structure indistinguishable from human blood group A antigen and on the large diversity between strains in terms of carbohydrate-binding specificities, we evaluated the different ligands implicated in attachment to oysters tissues of strains representative of two main genogroups of human norovirus. The GI.1 and GII.4 strains differed in that the latter recognized a sialic acid-containing ligand, present in all tissues, in addition to the A-like ligand of the digestive tract shared with the GI.1 strain. Furthermore, bioaccumulation experiments using wild-type or mutant GI.1 Viruslike particles showed accumulation in hemocytes largely, but not exclusively, based on interaction with the A-like ligand. Moreover, a seasonal effect on the expression of these ligands was detected, most visibly for the GI.1 strain, with a peak in late winter and spring, a period when GI strains are regularly involved in oyster-related outbreaks. These observations may explain some of the distinct epidemiological features of strains from different genogroups.

Abstract: A 4-year bacteriological survey (2003-2007) of four molluscs cultivated in France and faced with mortality episodes was performed by the French shellfish pathology network. The more abundant bacteria isolated during 92 mortality episodes, occurring mainly in Pacific oyster Crassostrea gigas, were identified by genotyping methods. It allowed us both to confirm the representativeness of Vibrio splendidus and Vibrio aestuarianus bacterial strains and to identify both a large number of Vibrio harveyi-related strains mainly detected during 2007 oyster mortality outbreaks and to a lesser extent bacterial strains identified as Shewanella colwelliana. Because metalloprotease has been reported to constitute a virulence factor in a few Vibrio strains pathogenic for C. gigas, several bacterial strains isolated in this study were screened to evaluate their pathogenicity in C. gigas spat by experimental infection and their ability to produce metalloprotease-like activity in the culture supernatant fluids. A high level (84%) of concordant results between azocaseinase activities and virulence of strains was obtained in this study. Because bacterial metalloprotease activities appeared as a common feature of pathogenic bacteria strains associated with mortality events of C. gigas reared in France, this phenotypic test could be useful for the evaluation of virulence in bacterial strains associated with such mortality episodes.

Abstract: Oyster extracts have been reported to have many bioactive peptides. But the function of oyster peptides produced by proteolysis is still unknown. In this study, the oligopeptide-enriched hydrolysates from oyster (Crassostrea gigas) were produced using the protease from Bacillus sp. SM98011 at laboratory level, and scaled up to pilot (100 L) and plant (1,000 L) levels with the same conditions. And the antitumor activity and immunostimulating effects of the oyster hydrolysates in BALB/c mice were investigated. The growth of transplantable sarcoma-S180 was obviously inhibited in a dose-dependent manner in BALB/c mice given the oyster hydrolysates. Mice receiving 0.25, 0.5 and 1 mg/g of body weight by oral gavage had 6.8%, 30.6% and 48% less tumor growth, respectively. Concurrently, the weight coefficients of the thymus and the spleen, the activity of natural killer (NK) cells, the spleen proliferation of lymphocytes and the phagocytic rate of macrophages in S180-bearing mice significantly increased after administration of the oyster hydrolysates. These results demonstrated that oyster hydrolysates produced strong immunostimulating effects in mice, which might result in its antitumor activity. The antitumor and immunostimulating effects of oyster hydrolysates prepared in this study reveal its potential for tumor therapy and as a dietary supplement with immunostimulatory activity.

Abstract: Aims: To assess the presence of human adenovirus (HAdV), hepatitis A (HAV) virus and rotavirus A (RV-A) in environmental samples from the Southern region of Brazil and to provide viral contamination data for further epidemiological studies and governmental actions. Methods and Results: Water samples from various sources (seawater, lagoon brackish water, urban wastewater, drinking water sources-with and without chlorination and water derived from a polluted creek) and oysters of two growing areas were analysed by enzymatic amplification (nested PCR and RT-PCR), quantification of HAdV genome (qPCR) and viral viability assay by integrated cell culture-PCR (ICC-PCR). From June 2007 to May 2008 in a total of 84 water samples, 54 (64·2%) were positive for HAdV, 16 (19%) for RV-A and 7 (8·3%) for HAV. Viability assays showed nonpositive samples for HAV; though, infectious viruses were confirmed for RV-A (12·5%) and HAdV (88·8%). Oyster samples by PCR were positive for HAdV (87·5%) and RV-A (8·3%), but none for HAV. Quantitative PCR in oysters showed means loads in genomic copies (gc) of 9·1 × 104 gc g−1 (oyster farm south) and 1·5 × 105 gc g−1 (oyster farm north) and in waters ranging from 2·16 × 106 (lagoon water) to 1·33 × 107 gc l−1 (untreated drinking water). Conclusions: This study has shown a widespread distribution of the analysed viruses in this particular region with high loads of HAdV in the environment which suggests the relevance of evaluating these viruses as positive indicators of viral contamination of water. Significance and Impact of the Study: The environmental approach in this study provides data concerning the prevalence, viability and quantification of enteric viruses in environmental waters and oysters in the South region of Brazil and has indicated that their presence might pose a risk to population in contact with the environmental samples searched.

To understand the species diversity and its evolutionary history, we collected five Crassostrea species from Asia and sequenced their complete mitochondrial (mt) genomes in addition to two newly released Asian oysters (C. iredalei and Saccostrea mordax) for a comprehensive analysis.

Abstract: Biogenic reefs formed by dense aggregations of the eastern oyster Crassostrea virginica are a dominant feature in most estuarine systems along the Atlantic and Gulf of Mexico. Oyster reefs are complex in their structural nature and have long been recognized for their potential habitat value. However, relatively few studies have characterized nekton abundance in this complex habitat type, and live high-relief oyster beds have been particularly difficult to sample with conventional gear. We used a quantitative sampling device to compare nekton use among high-relief live oyster reef, vegetated marsh edge Spartina alterniflora, and nonvegetated bottom habitat types. During 1 yr of seasonal sampling we collected 3791 fishes and 12 386 crustaceans representing 38 and 21 different species, respectively. Density and biomass of most fishes and crustaceans were significantly higher in oyster reef than over nonvegetated bottom. For benthic crustaceans, oyster reef supported a higher density and biomass than vegetated marsh edge. Nektonic crustaceans were generally more abun- dant in marsh edge than on oyster reef. Species composition and richness varied among habitat types and season; however, richness was highest in oyster reef, followed by marsh edge, and lowest on nonvegetated bottom, except during seasonal low densities during winter. Species composition and size differences were observed among habitat types. Our results show that oyster reef supports a high density, biomass, and richness of estuarine nekton in relation to typically examined estuarine habitat types and has the potential to be an essential habitat. Identifying and quantifying the role of oyster reefs will be critical to implementing effective management for essential fish habitat.

Abstract: To resolve the taxonomic status of a small cupped oyster from southern China, we collected and sequenced oysters from 20 sites in China along with sister species from other parts of the world. A total of 187 oysters were sequenced for 2 fragments belonging to the mitochondrial 16S rRNA and cytochrome oxidase I (COI) genes. Sequence analyses of both gene fragments indicate that the small cupped oyster from southern China is the same species as Crassostrea angulata from Portugal. C. angulata has a wide distribution in southern China, ranging from Zhejiang to Hainan, with the Yangtze River separating it from Crassostrea gigas in northern China. The wide distribution and high gene diversity in China support the suggestion of a historical transfer of C. angulata from China to Europe. Sequence divergence between C. angulata and C. gigas, 1.05–1.32% in 16S and 2.22–3.37% in COI, is higher than that between the most divergent populations of a Crassostrea species (C. virginica, 0.51% in 16S and 2.04–2.22%...

Abstract: Noroviruses have been recognized to be the predominant agents of nonbacterial gastroenteritis outbreaks in humans, and their transmission via contaminated shellfish consumption has been demonstrated. Norovirus laboratory experiments, volunteer challenge studies, and community gastroenteritis outbreak investigations have identified human genetic susceptibility factors related to histo-blood group antigen expression. Following a banquet in Brittany, France, in February 2008, gastroenteritis cases were linked to oyster consumption. This study identified an association of the norovirus illnesses with histo-blood group expression, and oyster contamination with norovirus was confirmed by qualitative and quantitative analyses. The secretor phenotype was associated with illness, especially for the non-A subgroup. The study showed that, in addition to accidental climatic events that may lead to oyster contamination, illegal shellfish collection and trading are also risk factors associated with outbreaks.

Abstract: Aims: To determine whether the infestation by the protozoan paramyxean parasite, Marteilia sydneyi, changes the bacterial community of the digestive gland of Sydney rock oysters, Saccostrea glomerata. Methods and Results: Six 16S rDNA clone libraries were established from three M. sydneyi-infected and three un-infected oysters. Restriction enzyme analysis followed by sequencing representative clones revealed a total of 23 different operational taxonomic units (OTUs) in un-infected oysters, comprising the major phyla: Firmicutes, Proteobacteria, Cyanobacteria and Spirocheates, where the clone distribution was 44, 36, 7 and 5%, respectively. Close to half of the OTUs are not closely related to any other hitherto determined sequence. In contrast, S. glomerata infected by M. sydneyi had only one OTU present in the digestive gland. Phylogenetic analysis of the 16S rDNA sequence reveals that this dominant OTU, belonging to the α-Proteobacteria, is closely related to a Rickettsiales-like prokaryote (RLP). Conclusions: The microbiota of the digestive gland of Sydney rock oysters is changed by infection by M. sydneyi, becoming dominated by a RLP, and generally less diverse. The bacterial community of un-infected S. glomerata differs from previous studies in that we identified the dominant taxa as Firmicutes and α-Proteobacteria, rather than heterotrophic γ-Proteobacteria. Significance and Impact of the Study: This is the first culture-independent study of the microbiota of the digestive glands of edible oysters to the species level. The commercial viability of the Sydney rock oyster industry in Australia is currently threatened by Queensland Unknown disease and the changes in the bacterial community of S. glomerata corresponding with infection by M. sydneyi sheds further light on the link between parasite infection and mortality in this economically damaging disease.

Abstract: Oysters from a harvesting area responsible for outbreaks of gastroenteritis were relaid at a clean seawater site and subsequently depurated in tanks of purified seawater at elevated temperatures. This combined treatment reduced norovirus levels to those detected prior to the outbreak. On the basis of norovirus monitoring the sale of treated oysters was permitted although the harvest area remained closed for direct sale of oysters. No reports of illness have been associated with the consumption of treated oysters.

Abstract: We report on the invasion of Brazil by the Pacific oyster Crassostrea gigas, and discuss the likely routes of invasion. Because this phenotypically diverse oyster sometimes resembles the native species C. brasiliana and C. rhizophorae, its invasion went unnoticed until it was detected through the analysis of DNA sequences for ribosomal 16S and the ribosomal second internal transcribed spacer. C. gigas was found amongst the native species in oyster banks up to 100 km south of oyster farms in South Brazil. Under most circumstances, water temperatures in the coastal southerly Brazil current would be too high to allow for the establishment of stable populations of C. gigas, but the production of spat in oyster farm laboratories has probably selected for resistance to warmer temperatures, which would promote invasion by C. gigas.

Abstract: Coastal ecosystems rely upon oyster reefs to filter water, provide protection from storms, and build habitat for other species. From a chemistry perspective, few details are available to illustrate how these shellfish construct such extensive reef systems. Experiments presented here show that oysters generate a biomineralized adhesive material for aggregating into large communities. This cement is an organic−inorganic hybrid and differs from the surrounding shells by displaying an alternate CaCO3 crystal form, a cross-linked organic matrix, and an elevated protein content. Emerging themes and unique aspects are both revealed when comparing oyster cement to the adhesives of other marine organisms. The presence of cross-linked proteins provides an analogy to mussel and barnacle adhesives whereas the high inorganic content is exclusive to oysters. With a description of oyster cement in hand we gain strategies for developing synthetic composite materials as well as a better understanding of the components nee...
