Abstract: The invention discloses a cuttage reproduction method of polygonum multiflorum. According to the cuttage reproduction method, lignified twigs of polygonum multiflorum which healthy grows for half a year to two years are taken as cutting slips, the cutting slips are specially processed, fertilizers are reasonably screened, and a proper cottage method is utilized, so that the survival rate of the cut polygonum multiflorum exceeds 95%, and high-quality seedlings can be provided for the field production of the polygonum multiflorum; and furthermore, the whole process is simple, the operation is simple, the breeding cycle is short, and the polygonum multiflorum can be transplanted after 30-45 days. The cuttage reproduction method is completely applicable to field cultivation.

Abstract: Objective To establish and optimize the rapid propagation system of Polygonum multiflorum, as well as explore method for induction and identification of autotetraploid. Method Propagation medium was optimized by orthogonal test. The buds were immersed in colchicine solution with different concentrations for different time to select induction conditions for autotetraploid of P. multiflorum. Result The most appropriate propagation medium was MS medium supplemented with 1.0 mg x L(-1) 6-BA, 0.3 mg x L(-1) NAA, and 0.4 mg x L(-1) PP333. That the buds were soaked in 0.2% colchicine solution for 30 h, or soaked in 0.3% colchicine solution for 18 h, was optimal condition to induce autopolyploid of P. multiflorum with induction rate as high as 16.7%. Conclusion Rapid propagation of P. multiflorum could be achieved by tissue culture. Furthermore, colchicine was an effective inducer of polyploidy, and 25 tetraploid lines were obtained through chromosome identification. The experiment laid a foundation for the wild resource conservation, superior varieties breeding of P. multiflorum.

Abstract: The invention discloses an establishment method of a rapid polygonum capitatum in-vitro propagation system. The polygonum capitatum is a polygonaceous polygonum plant, is used in miao minority in guizhou province, can be used as medicine wholly and can be commonly used for treating urinary system infection, rheumatalgia and other symptoms. The wild resource of polygonum capitatum is distributed rarely and dispersedly, and is in shortage severely due to excessive industrial consumption and deterioration of the ecological environment, and therefore, large-scale production is seriously hindered. According to the method, the vane of the polygonum capitatum is used as an explant, the callus is subjected to induction culture, multiplication culture, differentiation, rooting culture, acclimatization and transplantation to acquire the polygonum capitatum in-vitro regeneration plant, the rapid propagation technology system for polygonum capitatum tissue culture is established and scale development is promoted.
