Abstract: Prolactin has numerous biological actions in the brain, and transgenic mice are increasingly being used to investigate these actions. The present study aimed to provide a detailed mapping of the prolactin-responsive neurons in the female mouse forebrain by describing the distribution of prolactin receptor mRNA by in situ hybridization, and measuring prolactin-induced phosphorylation of signal transducer and activation of transcription 5 (pSTAT5) by immunohistochemistry. For in situ hybridization, a probe designed to detect both long and short receptor isoforms showed mRNA expression in a heterogeneous manner within the forebrain. Strong expression was observed in the rostral hypothalamus, particularly in periventricular regions, as well as in the arcuate and ventromedial nuclei of the mediobasal hypothalamus. There was also significant expression in some nonhypothalamic regions, notably high expression in the choroid plexus, and lower levels of expression in the medial amygdala, bed nucleus of the stria terminalis, and lateral septum. Prolactin-induced pSTAT5, detected by immunohistochemistry, provided a functional index of prolactin receptor activation in neurons. Prolactin-induced pSTAT5 was only observed in areas containing prolactin receptor mRNA, and was particularly prominent in the rostral and mediobasal hypothalamus. Most other areas that contained prolactin receptor mRNA also showed positive signal for prolactin-induced pSTAT5. The major exceptions were paraventricular nucleus and median preoptic nucleus, in which prolactin receptor mRNA was observed, but no induction of pSTAT5 by prolactin. The data provide key neuroanatomical information facilitating the use of the mouse model for furthering our understanding of prolactin actions in the brain.

Abstract: Concentrations of LH, FSH, prolactin, estrogen, and progesterone were measured in blood samples collected at various intervals during and immediately following lactation to determine the role of the ovary in controlling gonadotropin and prolactin secretion and initiation of estrous cycles in the pig. Ten primiparous sows were ovariectomized (ovex, n = 5) or left intact (n 5) 2 to 4 days after farrowing. Serum progesterone and estrogens in samples collected daily during lactation were not different between intact and ovex sows. Serum FSH increased immediately after ovariectomy and remained elevatedduring lactation,but LH concentrations were similar between ovex and intact sows. Both LH and FSH were higher during the last 2 weeks of lactation than during the first 3 weeks. On Days 10 and 20 of lactation, all sows received (i.v.) two challenges of gonadotropin releasing hormone (GnRH; 400 �g each) at 135 mm intervals. Peak change (maximum value minus baseline) in FSH after GnRH was higher in ovex than in intact sows on both Days 10 and 20. Peak change in FSH in ovex rows was higher on Day 20 than on Da� 10, but peak change in FSH in intact rows was similar between Days 10 and 20. Peak change in LH after GnRFI was similar between ovex and intact sows. Peak change in LH after the second GnRH challenge on Day 10 was higher than that after each of the other three challenges. Although serum prolactin concentrations in selected samples collected on Days 10 and 20 were not significantly different between treatment groups, intact sows had consistently higher concentrations compared with ovex sows. Separation of four sows from their litters for 4 h during the last week of lactation resulted in a decline in serum prolactin, but prolactin increased fivefold within 15 mm and tenfold during the second hour after piglets were replaced. Abstract: The effects of single sc injections of various doses of estradiol benzoate (Eb) and testosterone propionate (Tp) on plasma levels of LH, FSH and prolactin have been reinvestigated in castrate rats of both sexes. Our results indicate that Eb or Tp inhibited release of LH in castrate rats of both sexes. Males were more sensitive to the inhibitor effect of the low dose of Tp than females. The steroids also lowered plasma FSH levels in spayed females; however, in castrated males Eb stimulated FSH release and a decrease in FSH followed the injection of only the highest dose (2 mg) of Tp. Eb and Tp had a synergistic effect in inhibiting FSH release in the male. Both steroids stimulated release of prolactin in castrate males and females (no sex difference in release of prolactin in response to Eb was observed; however, males appeared more sensitive to Tp treatment).
