Abstract: A 58-point genetic map was constructed with RFLP, RAPD, isozyme, morphological, and disease-resistance markers spanning 766 cM on ten linkage groups for a cross within the cultivated cucumber (Cucumis sativus var. sativus). Relatively few DNA polymorphisms were detected, agreeing with previous studies documenting a narrow genetic base for cucumber. Most RFLPs within the cultivated cucumber appear to be changes at restriction-enzyme sites. Sixty-four percent of RAPD markers that fit expected ratios at P<0.001 were unlinked, possibly due to poor amplification and the inefficiency of dominant markers to detect linkage in an F2 family. A 70-point linkage map, spanning 480 cM on ten linkage groups, was constructed with RFLP, isozyme, morphological, and diseaseresistance markers for a cross between the cultivated cucumber and the wild or feral C. sativus var. hardwickii. Unlinked markers and more linkage groups than chromosome pairs indicated that both maps were not saturated. Twentyone markers doubly segregated in both families and regions of colinearity were identified.

Abstract: Species of the Heterodera avenae complex, including populations of H. arenaria, H. aucklandica, H. australis, H. avenae, H. filipjevi, H. mani, H. pratensis and H. ustinovi, obtained from different regions of the world were analysed with PCR-RFLP and sequencing of the ITS-rDNA, RAPD and light microscopy. Phylogenetic relationships between species and populations of the H. avenae complex as inferred from analyses of 70 sequences of the ITS region and of 237 RAPD markers revealed that the cereal cyst nematode H. avenae is a paraphyletic taxon. The taxonomic status of the Australian cereal cyst nematode H. australis based on sequences of the ITS-rDNA and RAPD data is confirmed. Morphometrical and ITS-rDNA sequence analyses revealed that the Chinese cereal cyst nematode is different from other H. avenae populations infecting cereals and is related to H. pratensis. Bidera riparia Kazachenko, 1993 is transferred to the genus Heterodera as H. riparia (Kazachenko, 1993) comb. n. As a consequence, H. riparia Subbotin, Sturhan, Waeyenberge & Moens, 1997 becomes a junior secondary homonym and is renamed as H. ripae nom. nov. Morphological, morphometrical characters and RFLP profiles for identification of the nine species presently placed in the H. avenae species complex are given.

Abstract: In the present study, two polymerase chain reaction (PCR)-based methods namely, randomly amplified polymophic DNA (RAPD) and amplification fragment length polymorphism (AFLP) were employed to assess genetic variations, which may appeared, in tissue culture-derived date palm (Phoenix dactylifera) offshoots. Analysis of RAPD banding patterns generated by PCR amplification using 37 random primers gave no evidences for somaclonal variations and the percentage of polymorphic bands in a total of 259 scored bands was zero. Meanwhile, analysis of AFLP banding patterns generated using 13 primer combinations pointed to minor genetic variations in the AFLP banding patterns. The percentage of genetic variations (polymorphism) in tissue culture-derived date palm offshoots belonging to cultivars Sakkoty, Gandila and Bertamoda was 2.6, 0.79 and 1 %, respectively, as revealed by AFLP analysis. The low percentage of genetic variations confirms the genetic stability of tissue culture-derived dry date palm cultivars.

Abstract: The genotypic and phenotypic diversity among isolates of the Lactobacillus curvatus/Lactobacillus graminis/Lactobacillus sakei group was evaluated by comparing RAPD data and results of biochemical tests, such as hydrolysis of arginine, D-lactate production, melibiose and xylose fermentation, and the presence of haem-dependent catalase. Analyses were applied to five type strains and to a collection of 165 isolates previously assigned to L. sakei or L. curvatus. Phenotypic and RAPD data were compared with each other and with previous DNA-DNA hybridization data. The phenotypic and genotypic separation between L. sakei, L. curvatus and L. graminis was clear, and new insights into the detailed structure within L. sakei and L. curvatus were obtained. Individual strains could be typed by RAPD and, after the elimination of similar or identical isolates, two sub-groups in both L. curvatus and L. sakei were defined. The presence or absence of catalase activity further distinguished the two L. curvatus sub-groups. By cloning and sequencing specific RAPD products, pairs of PCR primers were developed that can be used to specifically detect L. curvatus, L. sakei and each of the L. sakei sub-groups.

Abstract: With more than 100 accessions, the CBNMP olive collection includes a major part of the French germplasm. We used molecular markers to characterise all accessions and to study genetic relationships between cultivars. Firstly, 497 olive trees were genotyped using 32 RAPD markers. We identified 114 RAPD profiles and detected several cases of mislabelling, synonymy and homonymy. Secondly, for each RAPD profile, one tree was analysed using mtDNA RFLPs to determine the cytoplasmic lineage of each cultivar and using five nuclear SSR loci. French germplasm displayed ME1, MOM and MCK mitotypes with ME1 prevailing (84%). Based on SSR markers, we revealed a slight differentiation between French cultivars growing in the West and the East side of the Rhone Valley. This study allowed us to construct a molecular data-base for the reference collection and to analyse genetic diversity for further prospecting, and for introducing new olive accessions.
