Abstract: Maize (Zea mays L.) growth at low soil P levels is affected both by inherent physiological factors as well as interactions with soil microbes. The objectives of this study were (i) to quantify differences among maize inbred lines for growth at low P and response to mycorrhizal fungi, and (ii) to identify quantitative trait loci (QTL) controlling these traits in a B73 x Mo17 recombinant inbred population. Shoot dry weight and root volume were measured in the greenhouse after 6 wk of growth in a factorial experiment of 28 inbred maize lines using treatments of low vs. high P and mycorrhizal vs. nonmycorrhizal treatments. Shoot dry weight for the low P treatment in the absence of mycorrhizae ranged from 0.56 to 3.15 g. Mycorrhizal responsiveness based on shoot dry weight ranged from 106 to 800%. Shoot dry weight in the low P-nonmycorrhizal treatment was highly negatively correlated with mycorrhizal responsiveness. Plants grown at high P in the presence of mycorrhizae accumulated only 88% of the biomass of plants grown at high P in the absence of mycorrhizae, indicating that mycorrhizae can reduce plant growth when not contributing to the symbiosis. Percentage of root colonization was not correlated with mycorrhizal responsiveness. B73 and Mo17 were among the extremes for growth at low P and mycorrhizal responsiveness, and a B73 X Mo17 population of 197 recombinant inbred lines was used to detect QTL for growth at low P and mycorrhizal responsiveness. Three QTL were identified which controlled growth at low P in the absence of mycorrhizae based on shoot weight and one QTL which controlled mycorrhizal responsiveness. This study indicates that there is substantial variation among maize lines for growth at low P and response to mycorrhizal fungi. This variation could be harnessed to develop cultivars for regions of the world with P deficiency and for reduced-input production systems.

Abstract: Emergence and survival of honey mesquite (Prosopis glandulosa var.glandulosa Torr.) seedlings was quantified on sites with contrasting grazing histories: long-term continuous grazing (LTG) and long-term protection (LTP) from grazing by cattle. On each site, different levels of heroaceous defoliation were imposed at monthly intervals (no defoliation=ND, moderate=MD and heavy=HD). The two weeks following seed dissemination appeared to be the most critical toProsopis establishment on LTP-ND plots. Openings in the herbaceous layer created by moderate defoliation of grasses on the LTP site increased germination and/or survival 7-to 8-fold during this period. However, increasing the degree of defoliation from moderate to heavy did not stimulate additional emergence on either the LTP or LTG site. Emergence from scarified seed placed in cattle dung (17 to 30%) was lower than that of bare seed placements in various microhabitats (43–60%). However, deposition of scarifiedProsopis seed in dung in conjunction with graminoid defoliation may be the most likely combination of events when livestock are present. Emergence from seeds transported into grasslands by other fauna likely would be low, unless seeds were deposited in areas where grasses had been defoliated.Prosopis survival was comparably high in dung and bare seed placements after one growing season. survival of seedlings present two weeks after seed dissemination ranged from 74 to 97% at the end of the second growing season. Seedling survival and shoot development (biomass, leaf area and height) were similar on LTP and LTG sites, regardless of the level of herbaceous defoliation or seed placement. In addition, the magnitude and patterns of net photosynthesis, stomatal conductance and xylem water potential were comparable among one-year-old seedtings on ND, MD and HD plots, even though differences in herbaceous species composition and above- and below-ground biomass between these treatments were substantial. Such data suggest competition for soil resources between grasses andProsopis may be minimal early in the life cycle ofProsopis. High rates ofProsopis emergence and establishment on LTP-MD plots are counter to the widespread assumption that long-term and/or heavy grazing is requisite forProsopis encroachment into grasslands. Results are discussed with regard to factors contributing to the recent, widespread invasion of this woody legume into grasslands of southwestern North America.

Abstract: Conditions affecting Agrobacterium-mediated transformation of soybean [Glycine max (L.) Merr.], including seed vigor of explant source, selection system, and cocultivation conditions, were investigated. A negative correlation between seed sterilization duration and seed vigor, and a positive correlation between seed vigor and regenerability of explants were observed in the study, suggesting that use of high vigor seed and minimum seed sterilization duration can further improve transformation efficiency. Selection schemes using glufosinate or bialaphos as selective agents in vitro were assessed. Glufosinate selection enhanced soybean transformation as compared to bialaphos. The use of 6 mg L-1 glufosinate during shoot induction and shoot elongation stages yielded higher final transformation efficiency ranging from 2.0% to 6.3% while bialaphos at 4 to 6 mg L-1 gave 0% to 2.1% efficiency. Including cysteine and DTT during cocultivation increased the transformation efficiency from 0.2–0.9% to 0.6–2.9%. This treatment also improved T-DNA transfer as indicated by enhanced transient GUS expression. Shoot regeneration and Agrobacterium infection were attained in twelve soybean cultivars belonging to maturity groups I-VI. These cultivars maybe amenable to genetic transformation and may provide a valuable tool in soybean improvement programs.

Abstract: In seed plants, leaves are born on radial shoots, but unlike shoots, they are determinate dorsiventral organs made of flat lamina. YABBY genes are found only in seed plants and in all cases studied are expressed primarily in lateral organs and in a polar manner. Despite their simple expression, Arabidopsis thaliana plants lacking all YABBY gene activities have a wide range of morphological defects in all lateral organs as well as the shoot apical meristem (SAM). Here, we show that leaves lacking all YABBY activities are initiated as dorsiventral appendages but fail to properly activate lamina programs. In particular, the activation of most CINCINNATA-class TCP genes does not commence, SAM-specific programs are reactivated, and a marginal leaf domain is not established. Altered distribution of auxin signaling and the auxin efflux carrier PIN1, highly reduced venation, initiation of multiple cotyledons, and gradual loss of the SAM accompany these defects. We suggest that YABBY functions were recruited to mold modified shoot systems into flat plant appendages by translating organ polarity into lamina-specific programs that include marginal auxin flow and activation of a maturation schedule directing determinate growth.
