TL;DR: Six strains of those Leishmania species less sensitive to the azole drugs could be subcultured indefinitely at reduced growth rates in the presence of a ketoconazole concentration causing the same extraordinary alterations in sterol composition, suggesting that the bulk membrane functions of sterols in leishmanias can be served by 14 alpha-methyl sterols and cholesterol, albeit imperfectly, while traces of 14alpha-des methyl sterols are needed for uncharacterized metabolic functions.

Abstract: The effects of fenpropimorph on sterol biosynthesis and growth of Saccharomyces cerevisiae were examined to pinpoint the mode of action of fungicides that inhibit ergosterol biosynthesis. Taking advantage of sterol auxotrophy and sterol permeability in mutant strains, we show that growth inhibition is strongly correlated with inhibition of sterol biosynthesis. We confirm that in vivo and at low concentrations, fenpropimorph inhibits delta 8----delta 7-sterol isomerase, and in addition, when it is used at higher concentrations, it inhibits delta 14-sterol reductase. We show also that the fungistatic effect of fenpropimorph is not due to the accumulation of abnormal sterols in treated cells but is linked to the specific inhibition of ergosterol biosynthesis, leading to the arrest of cell proliferation in the unbudded G1 phase of the cell cycle.

Abstract: Previously, we reported neutron diffraction studies on the depth of cholesterol in phosphatidylcholine (PC) bilayers with varying amounts of acyl chain unsaturation [Harroun, T. A., et al. (2006) Biochemistry 45, 1227−1233]. The center of mass of the 2,2,3,4,4,6-D6 deuterated sites on the sterol label was found to reside 16 A from the middle of the bilayer in 1-palmitoyl-2-oleoylphosphatidylcholine (16:0-18:1PC), 1,2-dioleoylphosphatidylcholine (18:1-18:1PC), and 1-stearoyl-2-arachidonylphosphatidylcholine (18:0-20:4PC). This location places cholesterol’s hydroxyl group close to the membrane surface, indicative of the molecule in its commonly understood “upright” orientation. However, for dipolyunsaturated 20:4-20:4PC membranes the label, thus the hydroxyl group, was found sequestered in the center of the bilayer.
