Abstract: The transport of proteins through skin can be facilitated potentially by using terpenes as chemical enhancers. However, we do not know about the effects of these enhancers on the stability and biological activity of proteins which is crucial for the development of safe and efficient formulations. Therefore, this project investigated the effects of terpene-based skin penetration enhancers which are reported as nontoxic to the skin (e.g., limonene, p-cymene, geraniol, farnesol, eugenol, menthol, terpineol, carveol, carvone, fenchone, and verbenone), on the conformational stability and biological activity of a model protein lysozyme. Terpene (5% v/v) was added to lysozyme solution and kept for 24 h (the time normally a transdermal patch remains) for investigating conformational stability profiles and biological activity. Fourier transform infrared spectrophotometer was used to analyze different secondary structures, e.g., α-helix, β-sheet, β-turn, and random coil. Conformational changes were also monitored by differential scanning calorimeter by determining midpoint transition temperature (Tm) and calorimetric enthalpy (ΔH). Biological activity of lysozyme was determined by measuring decrease in A450 when it was added to a suspension of Micrococcus lysodeikticus. The results of this study indicate that terpenes 9, 10, and 11 (carvone, l-fenchone, and l-verbenone) decreased conformational stability and biological activity of lysozyme significantly (p < 0.05) less than other terpenes used in this study. It is concluded that smaller terpenes containing ketones with low lipophilicity (log Kow ∼2.00) would be optimal for preserving conformational stability and biological activity of lysozyme in a transdermal formulation containing terpene as permeation enhancer.

Abstract: The aim of this work was to characterize and compare the profiles of volatile terpenes in four potential superfruits. These profiles were determined using headspace solid-phase microextraction coupled with comprehensive two-dimensional gas chromatography linked to time-of-flight mass spectrometry. The proposed technique allowed the separation and identification of 79 terpenes present in cape gooseberry, crabapple, cherry silver berry, and scarlet hawthorn. The preliminary compound identification was based on the analysis of deconvoluted mass spectra and a comparison of the calculated linear retention indices with their values reported in the scientific literature. The compound identification was performed using the available standards. Also, a semi-quantitative total ion chromatogram-based analysis was performed. The richest terpene profile was identified in cape gooseberry (62 terpenes), where the terpene fraction constituted about 14% of total volatile fraction.

Abstract: The current text provides a comprehensive introduction to essential oils, their biosynthesis, naming, analysis, and chemistry. Importantly, this text quickly brings the reader up to a level of competence in the authentication of essential oils and their components. It gives detailed descriptions of enantiomers and other forms of stereoisomers relevant to the study of natural volatiles and essential oils. The text also describes GC-MS work and provides tips on rapid calculation of arithmetic indices, how to interpret suggested names from the NIST mass spectral library, and what additional efforts are required to validate essential oils and defeat sophisticated adulteration tactics. In brief, essential oils are mixtures of volatile organic compounds that were driven out of the raw plant material in distillation, condensed into an oil that is strongly aroma emitting, and collected in a vessel as the top layer (uncommonly bottom layer) of two phase separated liquids: oil and water. Essential oils commonly include components derived from two biosynthetic groups, being terpenes (monoterpenes, sesquiterpenes and their derivatives) and phenylpropanoids (aromatic ring with a propene tail). The current text provides details of how terpenes and phenylpropanoids are further categorised according to their parent skeleton, then recognised by the character of oxidation, which may be from oxygen, nitrogen, or sulphur, or the presence/absence of a double bond. The essential oil’s science niche is an epicentre of individuals from diverse backgrounds, such as aromatherapy, pharmacy, synthetic and analytical chemistry, or the hobbyist. To make the science more accessible to the curious student or researcher, it was necessary to write this fundamentals-level introduction to the chemistry of essential oils (i.e., organic chemistry in the context of essential oils), which is herein presented as a comprehensive and accessible overview. Lastly, the current review constitutes the only resource that highlights common errors and explains in simplistic detail how to correctly interpret GC-MS data then accurately present the respective chemical information to the wider scientific audience. Therefore, detailed study of the contents herein will equip the individual with prerequisite knowledge necessary to effectively analyse an essential oil and make qualified judgement on its authenticity.

Abstract: Essential oil terpenes are secondary metabolites produced in different biological pathways as pollinator attraction signals, but also as defense mechanisms against insect pests, herbivores, microorganisms, competing plants, etc. In this context, we have evaluated a total of 24 terpenes commonly found in Mediterranean aromatic plants, including 8 monoterpene hydrocarbons, 9 oxygenated monoterpenes, 2 esterified monoterpenes, 3 sesquiterpene hydrocarbons and 2 oxygenated sesquiterpenes in order to determine their antifeedant effects on the Colorado potato beetle (Leptinotarsa decemlineata Say), a major pest of potato crops. Terpene hydrocarbons showed low antifeedant activity, whereas the oxygenated sesquiterpene (-)-α-bisabolol with an inhibition activity of 96.3% was the most active, followed by carvacrol (90.9%), (+)-Terpinen-4-ol (87.1%) and thymol (81.5%). Other terpenes like (1S)-(-)-verbenone (72.9%), (+/-)-camphor (63.4%) and linalyl acetate (60.7%) showed moderate activity. Subsequently, terpenes showing antifeedant activity against this insect were also tested in allelopathic assays to determine potential damage to the crops. Results showed phytotoxic effects for (1S)-(-)-verbenone on Lactuca sativa germination, and for carvacrol on both the seed germination and the leaf and root growth of Lolium perenne. In conclusion, (-)-α-bisabolol is the best option to develop natural antifeedant formulations against L. decemlineata on the basis of its high antifeedant and low phytotoxic activities.
