Abstract: The effects of the combined administration of theophylline and ouabain were examined in isolated right ventricular strips from the rabbit heart. Separately, each drug produced positive inotropism. If ouabain was added 10 minutes after theophylline, a decrease in dF/dt was observed. The presence of theophylline augmented the increase in baseline tension induced by ouabain, although theophylline, by itself, did not augment baseline tension at the concentrations tested. There was a significant correlation between the decrease in dF/dt and the rise in baseline tension. Theophylline did not enhance the ability of ouabain to induce arrhythmia. The actions of caffeine, theobromine and digoxin were also examined and gave similar results.

Abstract: Tea Caffeine synthase (TCS) is one of the key enzymes involved in caffieine biosynthsis in tea plant (Camellia sinensis),which catalyses conversions of 7-methylxanthine to theobromine and theobromine to caffeine. Inhibition of TCS gene expression can leads to breeding low caffeine tea cultivars. Two cDNA fragments of TCS gene were amplified by RT-PCR, and ligated into T-vector. The two TCS gene fragments were inserted into RNAi vector pFGC5941 in reverse direction after double digestion with two pairs of restriction endonucleases. The insertion of two fragments, namely pFGC5941-TCS02 and pFGC5941-TCS03, into the RNAi vector were confirmed by PCR and DNA sequencing.

Abstract: NEW MATERIAL:A xanthine derivative of formula I [one of R 1 WR 3 is HC≡C(CH 2 ) n -, (n is 2W5), and the other two are methyl]. EXAMPLE: 1-(5'-Hexynyl)theobromine. USE: Intermediate of medicines. It has peripheral vessel dilating activity and erythrocyte deformation promoting activity, low toxicity and high solubilty, and is useful as an intermediate of a pharmaceutically valuable xanthine derivative (e.g. pentoxifylline). PROCESS: The compound of formula I is prepared by reacting an easily available compound of formula II (one of R 4 WR 6 is H and the other two are methyl) with an alkyne derivative of formula HC≡C(CH 2 ) n X (X is halogen, etc.).
