Abstract: Cyanobacteria are pathogenic prokaryotes and known for producing a high variety of cyclic hepatotoxic peptides in fresh and brackish water. Prominent members of these toxins are microcystin LR (MC LR) and nodularin (Nod), which are under suspicion to cause cancer. Various analytical methods have been reported for the detection of these cyclopeptides, and these are mainly based on liquid chromatography combined with mass spectrometric techniques. Here, we introduce a new approach based on the direct coupling of high-performance thin-layer chromatography (HPTLC) with infrared matrix-assisted laser desorption/ionization orthogonal time-of-flight mass spectrometry (IR-MALDI-o-TOF MS) using the liquid matrix glycerol. The analysis of the cyclopeptides involves the application of three complementary methods: (i) HPTLC separation of MC LR and Nod, (ii) their detection and quantification by UV spectroscopy at λ = 232 nm, and (iii) direct identification of separated analytes on the HPTLC plate by IR-MALDI-o-TOF MS...

Abstract: Objective: The objective was to study the antimicrobial activity of active eluent of thin layer chromatography (TLC) from Streptomyces isolated from marine sample. Methods: To determine the antimicrobial activity by agar well diffusion method. Active compounds were extracted by solvent extraction method and purified using TLC. The presence of active compounds was confirmed by gas chromatography-mass spectrometry (GC-MS) analysis. Identification of active Streptomyces by 16S rRNA partial gene sequencing method. Results: The active strain was identified as Streptomyces cacaoi strain SU2 (JF730119). The GC-MS analysis revealed that the presence of dodecane, eicosane, cetene, diethyl phthalate, phthalic acid isobutyl nonyl ester, thieno[3,2-e] benzofuran and bis (2-ethylhexyl) phthalate. Conclusion: The methods adopted and active metabolites extracted and purified can be fruitfully employed for obtaining novel antibiotic compounds to treat human pathogenic bacterial and fungal infections. Abstract: Changes in the fatty acid composition of egg-yolk fat of hens fed diets with increasing fish meal content are studied by total fatty acid analysis. The fatty acid composition of the major lipid fractions in egg-yolk fat after feeding a high-level fish meal diet was determined by a combination of thin layer chromatography (t.l.c.) and gas-liquid chromatography (g.l.c.) The changes produced show a fatty acid pattern similar to those of the diets themselves. Long-chain polyunsaturated fatty acids are deposited preferentially in the phospholipids, reaching the highest concentration in cephalin and lecithin.
