Abstract: A gene, qid74, of mycoparasitic filamentous fungus Trichoderma harzianum and its allies encodes a cell wall protein that is induced by replacing glucose in the culture medium with chitin (simulated mycoparasitism conditions). Because no trace of this gene can be detected in related species such as Gibberella fujikuroi and Saccharomyces cerevisiae, the qid74 gene appears to have arisen de novo within the genus Trichoderma. Qid74 protein, 687 residues long, is now seen as highly conserved tandem repeats of the 59-residue-long unit. This unit itself, however, may have arisen as tandem repeats of the shorter 13-residue-long basic unit. Within the genus Trichoderma, the amino acid sequence of Qid74 proteins has been conserved in toto. The most striking is the fact that Qid74 shares 25.3% sequence identity with the carboxyl-terminal half of the 1,572-residue-long BR3 protein of the dipteran insect Chironomus tentans. BR3 protein is secreted by the salivary gland of each aquatic larva of Chironomus to form a tube to house itself. Furthermore, the consensus sequence derived from these 59-residue-long repeating units resembles those of epidermal growth factor-like domains found in divergent invertebrate and vertebrate proteins as to the positions of critical cysteine residues and homology of residues surrounding these cysteines.

Abstract: The effect of aeration on the production of xylanase by Trichoderma longibrachiatum on wheat bran in a solid-state fermentation (SSF) system has not been investigated. This study was conducted to investigate the interactive effects of aeration, initial moisture content of the substrate, and depth of the substrate on xylanase activity produced in a tray fermenter. The experiment was conducted as a split plot experiment with factorial treatments and three replications of each treatment combination. The whole plot treatment was aeration rate (0, 2.9, 5.7 L/min/kg bran). Initial moisture content (45, 55, 65% w.b.) and depth of substrate (1.0 and 2.5 cm) were investigated factorially. Trays of wheat bran were assayed after fermentation by Trichoderma longibrachiatum to determine the production of xylanase activity. Aeration rate had a significant nonlinear effect on enzyme activity with highest yields obtained at an aeration rate of 2.9 L/min/kg bran (738 U/g, averaged over all initial moisture contents and depth of substrates). Initial moisture content of the substrate also had a significant nonlinear effect on enzyme activity with the highest yields at 55% (556 U/g, averaged over all airflow rates and depths of substrate). Depth of substrate had no significant statistical effect on enzyme activity. The treatment combination of 2.9 L/min/kg airflow rate and 55% moisture content resulted in the highest yields (948 U/g, averaged over depth of substrate).

Abstract: Trichoderma longibrachiatum is an emerging pathogen in immunocompromised patients. We report a case of Trichoderma post-operative mediastinitis and peritonitis in a child with complex congenital cardiac disease and functional asplenia. The patient was treated unsuccessfully, initially with caspofungin alone followed by a combination of voriconazole (systemic and topical), caspofungin and intraperitoneal amphotericin B.

Abstract: Cellulase is one of the most widely distributed enzymes with wide application. They are involved in conversion of biomass into simpler sugars. Cellulase of Trichoderma longibrachiatum, a known cellulolytic fungus was compared with Clostridium thermocellum [AAA23226.1] cellulase. Blastp was performed with AAA23226.1 as query sequence to obtain nine similar sequences from NCBI protein data bank. The physicochemical properties of cellulase were analyzed using ExPASy’s ProtParam tool namely ProtParam, SOPMA and GOR IV. Homology modeling was done using SWISS MODEL and checked quality by RMSD values using VMD1.9.1. Active sites of each model were predicted using automated active site prediction server of SCFBio. Study revealed instability of cellulase of two eukaryotic strains namely Trichoderma longibrachiatum [CAA43059.1] and Melanocarpus albomyces [CAD56665.1]. The negative GRAVY score value of cellulases ensured better interaction and activity in aqueous phase. It was found that molecular weight (M. Wt) ranges between 25-127.56 kDa. Iso-electric point (pI) of cellulases was found to be acidic in nature. GOR IV and SOPMA were used to predict secondary structure of cellulase, which showed that random coil, was dominated. Neighbor joining tree with C. thermocellum [AAA23226.1] cellulase as root showed that cellulases of Thermoaerobacter subterraneus [ZP_07835928] and C. thermocellum [CAA4305.1] were more similar to eukaryotic cellulases supported by least boot strap values. Pseudoalteromonas haloplanktis cellulase was found to be the ideal model supported by least RMSD score among the predicted structures. Trichoderma longibrachiatum cellulase was found to be the best compared to other cellulases, which possess high number of active sites with ASN and THR rich active sites. CYS residues were also present ensuring stable interaction and better bonding. Hydrophilic residues were found high in active sites of all analyzed models and template.

Abstract: The present work profiled the fungal community structure found in Brazilian energy transmission tower with signs of corrosion and/or biofilm formation using cloning (ITS-rRNA gene libraries) and culture-dependent technique. A total of 31 isolates comprising 10 filamentous fungi and 21 yeasts were recovered from enrichment cultures. As determined by polyphasic taxonomy 9 genera and 13 species were identified including Aspergillus fumigatus, Aspergillus niger, Candida albicans, Candida pseudointermedia, Candida tropicalis, Cryptococcus laurentii, Debaryomyces nepalensis, Exophiala dermatitidis, Fusarium sp., Fusarium solani, Paecilomyces lilacinus, Trichoderma citrinoviride, Trichoderma longibrachiatum, and Pichia guilliermondii. Metagenomic analyses based on 160 clone sequences revealed 30 operational taxonomic units (OTUs) comprising 20 OTUs of filamentous fungi and 10 OTUs of yeasts. The majority of OTUs were related to the genera Capnobotryella, Cryptococcus, Devriesia, Fellomyces, Fusarium, Kockovaella, Panaeolus, Rhodotorula, Sirobasidium, Sporobolomyces, Strelitziana and Teratosphaeria. Although members of the fungal community from transmission tower samples are ubiquitous fungi commonly found in other environments, some have been related to microbiologically-influenced corrosion of metals. Comparisons between fungal community composition obtained by both culture-dependent and independent methods highlighted the different aspects of the mycobiota, emphazising the need of complementary approaches to assess the microbial assemblage of unusual environments.
