Abstract: An XIP (xylanase inhibiting protein) type xylanase inhibitor was purified from Algerian pearl millet ( Pennisetum glaucum L.) grains and characterized for the first time. Cation exchange and affinity chromatography with immobilized Trichoderma longibrachiatum glycoside hydrolase (GH) family 11 xylanase resulted in electrophoretically pure protein with a molecular mass of 27-29 kDa and a pI value of 6.7. The experimentally determined N-terminal amino acid sequence of the purified XIP protein is 87.5%, identical to that of sorghum ( Sorghum bicolor L.) XIP and 79.2% identical to that of wheat ( Triticum aestivum L.) XIP-I. The biochemical properties of pearl millet XIP are comparable to those described earlier for sorghum XIP, except for the higher specific activity toward a T. longibrachiatum GH family 11 xylanase. On the basis of immunoblot neither TAXI nor TLXI type xylanase inhibitors were detected in pearl millet grains.

Abstract: Different inocula of Trichoderma longibrachiatum were tested in a murine model, and only the highest one (1 × 10(7) CFU/animal) killed all of the mice at day 15 postinfection, with spleen and liver the most affected organs. The efficacies of amphotericin B deoxycholate, liposomal amphotericin B, voriconazole, and micafungin were evaluated in the same model, with very poor results. Our study demonstrated the low virulence but high resistance to antifungal compounds of this fungus.

Abstract: FIELD: biotechnology, mycology, biochemistry, enzymology. SUBSTANCE: strain of mycelioid fungus Trichoderma longibrachiatum TW-1 VKM F-3634D is obtained by method of multiple classical mutagenesis and selection from the parent culture Trichoderma reesei QM 6a using ultraviolet irradiation. Strain shows ability to produce complex of highly active carbohydrases that makes a possibility of full complex of enzymes and also if necessary individual enzymes (components) of complex. For achievement of high productivity of the strain the use of complex and expensive nutrient media is not required. Nutrient media conventionally using in industrial technologies for preparing similar enzyme preparations are used for culturing. Invention can be used in food industry and agriculture. Invention allows to expanse effectiveness and to broad spectrum of enzyme preparations using in different fields of biotechnology and, especially, as fodder additions. Abstract: Xylanases catalyse the cleavage of various forms of xylan. A new crystal form of xylanase II from the fungus Trichoderma longibrachiatum that diffracts to better than 1 A resolution was grown from 12% PEG 8K, 0.1 M Tris pH 8.5, 0.2 M CaCl(2) with the addition of 2% glycerol to overcome crystal twinning. The crystals grow in a body-centered orthorhombic Bravais lattice, with unit-cell parameters a = 66.78, b = 67.94, c = 79.18 A. The solvent content is 42% with one molecule per asymmetric unit. Molecular-replacement analysis reveals the space group to be I222. This atomic resolution structure will provide important insights that will lead to a better understanding of the enzymatic mechanism of the family 11 xylanases.

Abstract: Corn (Zea mays) is one of the important cereals crops as the source of food, animals feed or energy. The corn produces has decreased by year to year. One of the factors that decreased the corn produces is downy mildew. Downy mildew desease caused by Peronosclerospora maydis fungi that decreased the production until 90%. Application of antagonistic fungi is one of the alternative control. Trichoderma spp. is one of the antagonistic fungi that can suppress growth of another fungus. Trichoderma spp. is saprofit soil microorganism that naturally attacks pathogen fungi and has benefits for plant growth. This research was aimed to know the effect of some isolates of Trichoderma spp. for diseases of downy mildew and the effect for the plant growth. Treatments were prepared with Randomized Complete Block (RCB) with 7 treatments and 4 repetitions. The treatments consist are P0 (control), P1 (application Trichoderma viride isolate GDR), P2 (application Trichoderma harzianum isolate TRJ), P3 (application Trichoderma harzianum isolate TGN), P4 (application Trichoderma viride isolate NTF), P5 (application Trichoderma longibrachiatum isolate GRP) and P6 (application Trichoderma viride isolate KLN). The results of this research were Trichoderma viride isolate GDR, Trichoderma harzianum isolate TGN, Trichoderma viride isolate NTF and Trichoderma viride isolate KLN can be suppressed the disease incidence of downy mildew, but unaffected for the plants growth.
