Abstract: The β2 adrenergic receptor (β2AR) plays a key role in the control of smooth muscle relaxation in airways, the therapy of asthma, and a series of other basic physiological functions. Recently, the crystal structure of the β2AR-Gs protein complex was reported, which facilitates study of the activation mechanism of the β2AR and G-protein-coupled receptors (GPCRs). In this work, we perform 20 ns molecular dynamics (MD) simulations of the β2AR-Gs protein complex with its agonist in an explicit lipid and water environment to investigate the activation mechanism of β2AR. We find that during 20 ns MD simulation with a nanobody bound the interaction between the β2AR and the Gs protein is stable and the whole system is equilibrated within 6 ns. However, without a nanobody stabilizing the complex, the agonist triggers conformational changes of β2AR sequentially from the extracellular region to the intracellular region, especially the intracellular parts of TM3, TM5, TM6, and TM7, which directly interact with the Gs protein. Our results show that the β2AR-Gs protein complex makes conformational changes in the following sequence: (1) an agonist-bound part of β2AR, (2) the intracellular region of β2AR, and (3) the Gs protein.

Abstract: The Pearl River Delta Economic Zone is the most dynamic economic area in South China. One of the major problems in the region is the sustainable utilization of the water resources. On the basis of analysis of the water environment status and pollution sources, it is suggested that domestic sewage is the primary cause of pollution. Two new concepts "degradation coefficient" and "degradation volume" of water resources, due to pollution, which may be used to assess macroscopically the carrying capacity of the water resources and sustainability of the water environment, are proposed by the authors. The results calculated indicate that the volumes of degraded water resources will be up to 204, 352, and 537 million m3 in 2002, 2010, and 2020. It is suggested that water for daily consumption and domestic sewage must be controlled more effectively and there should be cross-regional coordination in tackling problems of water environment.

Abstract: Enteroviruses can be introduced into the water environment as a result of human activity. Contaminations within hot tubs, spas and public baths are also possible. We investigated the distribution of enteroviruses at six hot spring recreation areas throughout Taiwan. Spring water was collected from 34 sites and enteroviruses were detected in 13 (38.2%). The most frequently detected was coxsackievirus A2, followed by echovirus 11. Enterovirus 71 (EV 71) and porcine enterovirus 9 were detected once. Water quality indicators were not statistically associated with the occurrence of enteroviruses, although the enterovirus-positive samples were positive for a greater number of microbiological indicators and showed a link to pH and water temperature. The results confirm the ubiquity of enteroviruses in Taiwan spring recreation areas. Coxsackievirus A2, echovirus 11 and EV 71, the enteroviruses responsible for disease outbreaks identified at these sites, should be considered a potential public health threat in spring recreation areas of Taiwan.

Abstract: As one of the largest copper–molybdenum (Cu–Mo) mines in the world, the Erdenet Mine in Mongolia has been active since 1978 and is expected to continue operations for at least another 30 years. In this study, the potential impacts of mining activities on the soil and water environments have been evaluated. Water samples showed high concentrations of sulfate, calcium, magnesium, Mo, and arsenic, and high pH values in the order of high to low as follows: tailing water > Khangal River > groundwater. Statistical analysis and the δ2H and δ18O values of water samples indicate that the tailing water directly affects the stream water and indirectly affects groundwater through recharge processes. Soil and stream sediments are highly contaminated with Cu and Mo, which are major elements of ore minerals. Based on the contamination factor (CF), the pollution load index (PLI), and the degree of contamination (Cd), soil appears to be less contaminated than stream sediments. The soil particle size is similar to that of tailing materials, but stream sediments have much coarser particles, implying that the materials have different origins. Contamination levels in stream sediments display a tendency to decrease with distance from the mine, but no such changes are found in soil. Consequently, soil contamination by metals is attributable to wind-blown dusts from the tailing materials, and stream sediment contamination is caused by discharges from uncontained subgrade ore stock materials. Considering the evident impact on the soil and water environment, and the human health risk from the Erdenet Mine, measures to mitigate its environmental impact should be taken immediately including source control, the establishment of a systematic and continuous monitoring system, and a comprehensive risk assessment.
