Abstract: The explants with higher regeneration potential raised through in vitro germination of seeds of two genotypes of Withania somnifera namely: WS-100 and WS-134 were cultured on MS basal media fortified with 0.6mg/ml BAP and 0.4 mg/ml IAA. Culture medium MS + 0.2 mg/l BAP + 30.0 gm/l sucrose + 7.5 gm/l agar, induced callus in higher frequencies in both the varieties. While in multiple shooting, maximum shoots were observed in WS 100 with MS+0.2 mg/lit BAP+0.2 mg/lit IAA and in WS 134 with MS+0.3mg/l BAP + 0.2 mg/l IAA. Higher in vitro rooting response was achieved on rooting medium MS + 5.0 mg/1 IBA + 25.0 gm/1 sucrose + 7.5 gm/1 agar in WS 100 and in WS 134 with MS +0.6 mg/l BAP+ 2.5 mg/1 IBA + 25.0 gm/1 sucrose + 7.5 gm/1 agar. In terms of in vitro genotypic response, genotype WS-100 was found significantly superior to WS-134 for the most of the attributes investigated. Regenerated plantlets were established successfully in the field after primary and secondary hardening. The present investigation brought out possibility of mass-scale in vitro (micropropagation) production of Indian Ashwagandha cultivars.

Abstract: Ashwagandha (Withania somnifera Dunal) is a popular Indian medicinal plant and has been used for over 3000 years in Ayurvedic medicine to treat diverse range of diseases. Ashwagandha is a member of the class of herbs called rasayana, long prized for rejuvenating effects on health. It is believed that it assists body to respond appropriately to stressors, both acute and chronic [1]. The plant was first mentioned in English language text by Van Rheede in 1868, who described the use of its leavesin home ointments. Other parts of the Ashwagandha plant, including roots, shoots, seeds, and berries have also been used in daily tonics and various home remedy recipes to increase health and longevity [2]. Abstract: Withania somnifera (Ashwagandha) root extract is an important herbal medicine. The current study was designed to investigate the impact of Consciousness Energy Healing Treatment (The Trivedi Effect®) on the characteristic properties of the phytoconstituents present in the ashwagandha root extract using LC-MS, GC-MS, and NMR spectroscopy. Ashwagandha root extract was divided into two parts – one part was control (without treatment), while another part was treated with the Consciousness Energy Healing Treatment remotely by seven renowned Biofield Energy Healers and defined as the Biofield Energy Treated sample. The LC-MS analysis revealed that the retention time (Rt) of the phytoconstituents remained same in both the control and treated samples, whereas the peak area% was significantly altered. The peak area% of the treated sample at Rt of 5.06, 5.24, 5.49, 5.68, 6.78, 6.87, 8.12, 8.22, 8.36, 8.55, 9.02, and 10.13 minutes were significantly increased in the range of 0.27% to 146.94% compared to the control sample. In addition, the peak area% of the treated sample at Rt of 5.29, 5.36, 6.40, 6.49, 6.56, 6.67, 7.08, 7.37, 7.76, 7.87, 8.02, 8.43, 8.85, 8.97, and 9.12 minutes were significantly decreased in the range of 1.83% to 23.53% compared with the control sample. A total of 18 withanolides such as sitoindoside IX, viscosa lactone B, 24, 25-dihydrowithanolide D, withanolide A, withanone, withaferin A, withanolide D, ixocarpalactone A, withanolide S, withanolide sulfoxide, etc. were proposed with their structure from the molecular mass at m/z 650, 489, 473, 471, 505, and 992 at retention times of 6.87, 7.10, 7.87, 8.22, 8.54, and 9.11 minutes with the help of GC-MS and NMR data of both the control and Biofield Energy Treated samples. The mass peak intensity values of the Biofield Energy Treated sample at the same retention time in was significantly decreased in the range of 3.99% to 52.28% compared with the control sample. These findings suggest that Energy of Consciousness Healing Treatment could be beneficial for altering the concentration of the phytoconstituents in the ashwagandha root extract by modifying their intrinsic physicochemical properties, which might be helpful to improve the bioavailability of active constituents of ashwagandha root extract that might provide better therapeutic response against inflammatory diseases, immunological disorders, arthritis, cancer, diabetes, sexual disorders, aging and other chronic infections.

Abstract: Objective: To investigate the protective effect of methanolic extract of Withania somnifera (Ashwagandha) on Mitomycin C induced damage in mouse bone marrow and antioxidant enzymes in liver. Materials and Methods: Protective effect of methanolic extract of Ashwagandha (250 mg/kg b.w. p.o, for 7 days) was assessed against formation of micronuclei (MN) in polychromatic cells (PCEs) and normochromatic cells (NCEs) induced by Mitomycin C (4 mg/kg b.w., i.p) using mouse bone marrow micronucleus test. The effect of Mitomycin C and Ashwagandha extract on liver enzymes, superoxide desmutase (SOD), and glutathione (GSH) was also evaluated. Results: The frequency of occurrence of micronucleated cells in bone marrow was reduced significantly when methanolic extract of Ashwagandha was administered before Mitomycin C. There was a reversal of decrease in P/N ratio. The level of SOD and GSH in liver was significantly depressed by Mitomycin C treatment and the Ashwagandha treatment enhanced level of these enzymes significantly. Conclusions: These findings suggest that Ashwagandha extract is effective in preventing DNA damage, and one of the mechanisms of action might involve scavenging of active oxygen radicals generated in reactions initiated by the mutagens.
