Abstract: Endogenous cytokinins were quantified and identified in germinating achenes and developing seedlings of Tagetes minuta L. incubated at 25 °C over a 144 h period. The process of germination (radicle emergence) was completed 38 h after commencement of imbibition. Subsequent growth was considered to cover seedling establishment. Eighteen isoprenoid cytokinins, belonging to the zeatin (9), dihyrozeatin (5) and isopentenyladenine (4) groups and one aromatic cytokinin, benzyladenine, were identified. The total isoprenoid cytokinin concentration increased upon imbibition, reached a peak by 48 h and subsequently decreased with seedling development. The individual cytokinin groups and the respective derivatives within each group did, however, not follow such a consistent trend. During the course of the experiment, the ribotides and ribosides were present in the highest concentrations, reaching a peak at 48 h and decreasing thereafter. The free bases and O-glucoside remained at low levels throughout the experiment. Isopentenyladenine-9-glucoside increased dramatically in the developing seedlings and after 144 h was the predominate cytokinin. Benzyladenine was the only aromatic cytokinin detected throughout the experiment. It was present in high concentrations in the dry achenes and declined rapidly upon imbibition.

Abstract: High-frequency somatic embryogenesis was achieved from an embryogenic cell suspension culture of Acanthopanax koreanum Nakai. Stem segments were cultured on Murashige and Skoog (MS) medium containing auxins and cytokinins. Opaque and friable embryogenic callus formed on MS medium with 4.5 µm 2,4-dichlorophenoxyacetic acid (2,4-D) and 2.0 µm kinetin or zeatin, but was highest on medium containing 4.5 µm 2,4-D alone. Embryogenic calli were transferred to MS liquid medium containing 4.5 µm 2,4-D and maintained by subculture at 2-week intervals. Initiation of somatic embryogenesis and development up to the globular stage from embryogenic cell clumps occurred in medium containing 0.45 µm 2,4-D, whereas maturation and germination of somatic embryos occurred in MS medium lacking 2,4-D. Cytokinin treatment suppressed the normal growth of embryos, but stimulated secondary somatic embryogenesis from the surfaces of primary embryos. Plants from somatic embryos were acclimatized in a greenhouse.

Abstract: Node-derived shoot cultures of Eucalyptus regnans were established from in vitro grown seedlings on Murashige and Skoog basal medium supplemented with 0.5 mg L-1 (2 μm) zeatin and 0.05 mg L-1 (0.3 μm) napthaleneacetic acid. A double sterilisation method was essential to obtain clean material from seed. Microcuttings from established cultures were used to develop an efficient method for in vitro rooting. Rooting was best after a 7 day pulse on 20 mg L-1 (98 μm) indolebutyric acid. Hoagland's or Woody Plant Medium supported better rooting than MS basal medium and rooting was significantly enhanced by subculture to activated charcoal after the auxin pulse. Carbohydrate (sucrose or glucose) was essential for rooting while high light intensity was inhibitory. Optimal light conditions were a 12 h day (17 W m-2). In all, 90% of plantlets established in the nursery survived the winter.
