The eect of dierent growth regulators on in vitro growth and plant regeneration of tomato (Lycopersicon esculentum Mill.) explants, derived from hypocotyls and cotyledons of aseptically grown seedlings, was studied. With regard to the regeneration frequency, number of shoot primordia and shoots per explant, the best regeneration medium was the Murashige-Skoog (MS) medium supplemented with 1 mg L 1 of zeatin and 0.1 mg L 1 of indole3-acetic acid. In all genotypes studied, 100% frequency of regeneration was observed when hypocotyl explants were used.

Abstract: Micropropagation methods were developed for the three French varieties of olive (Olea europaea L.), Aglandau and Tanche, that have the Appelation d’Origine Controlee (AOC) status and one ecotype (05300, Laragne, France). Explants consisting of partially lignified nodal segments were collected from rejuvenated glasshouse-grown plant material. Nodal explants with axillary buds were cultured on different media. For AOC varieties, olive medium modified (OM mod) to contain half the concentration of macroelements was the most efficient in inducing bud break and growth when supplemented with 30 g l−1sucrose and 4 mg l−1 zeatin. The resulting shoot buds were further multiplied and maintained on OM mod medium. Rooting was best achieved on OM supplemented with 4 mg l−1 indole-3-butyric acid (IBA). For the Laragne ecotype, maximal shoot proliferation occurred when explants were cultured on woody plant medium supplemented with 15 g l−1 sucrose and 0.1 mg l−1 zeatin. Efficient rooting was achieved with 1 mg l−1 IBA combined with 0.75 mg l−1 naphthaleneacetic acid. After acclimatization in the glasshouse, survival rates ranged from 57 to 92%, depending on the genotype. Abstract: DNA breakdown (Miller et al., 1955). Kinetin was significantly more active than adenine in the tobacco tissue culture bioassay. The use of the plant cell culture bioassay was a key to the eventual isolation of zeatin from corn by Letham (1963). ETHYLENE. The growth regulating properties of ethylene were first recognized by the Russian scientist Nejebulov in 1901 (Beyer et al., 1984). His experiments showed that illuminating gas could cause leaf abscission and epinasty. The fruit physiologist Crocker developed the Alaskan pea bioassay that used the triple response of shortening and thickening of the hypocotyl, agravitropic growth, and maintained hypocotyl hook (Reid and Howell, 1995) to assess ethylene levels. He was the first to suggest that ethylene was an endogenous plant hormone. However, few scientist at the time agreed with Crocker since it was difficult to visualize that a gas could act as an endogenous regulating substance. It was not until the 1960s when gas chromatography was used to quantify endogenous ethylene that the significance of ethylene was recognized and acknowledged as an endogenous hormone. ABSCISIC ACID.While researchers had long speculated that a hormone which affected abscission, dormancy, and stress reduction existed, it was not until the 1950s that work on the isolation and identification of this compound began. In 1953 Bennet-Clark and Kefford discovered an acid fraction of plant extracts that was a potent growth inhibitor. In 1955 Osborne reported on the diffusion of a senescence factor from older leaves that could speed the abscission of bean petioles. By 1963, Addicott s group studying cotton fruit abscission in California had identified and characterized abscisin II. At the same time, Wareing s group in Wales identified the dormin as a compound that promoted dormancy in woody plants. In 1968, these groups formally redesignated abscisin II and dormin as abscisic acid (Milborrow, 1984).

Abstract: Excised shoot apices (0.6 or 1.0 mm long) of Rosa multiflora were grown in axenic culture on defined media including auxins, gibberellic acid and cytokinins. In the presence of zeatin (10(-8)-10(-7)M) and 6-benzylaminopurine (10(-7)-10(-6) M) apices grew into plantlets but two other cytokinins, kinetin and 6-(3-methylbut-2-enyl)aminopurien, were ineffective. Auxins or gibberellic acid were either inhibitory or had no effect on differentiation.
