Abstract: Three tomato cultivars were used to examine the influence of the genetic background on the regeneration efficiency. White embryonic calli were formed within two weeks of culture. Shoots emerged either directly from the explant or indirectly from the embryonic callus. Multiple adventitious shoots were formed by clonal propagation of somatic embryos in the presence of 2 mg zeatin L-1. The meristematic end of the hypocotyl of the cultivar Pontaroza showed a high regeneration frequency (70.2%) compared with the cotyledonary leaf explant (35.3%). The plants grown in the green house and the regenerants obtained showed a similar peroxidase banding pattern. The combined analysis of variance indicated that the difference in shoot induction between cultivars was highly significant. Shoot induction frequency was 57.2%, 43.5%, and 35.5% for the cultivars UC-97, Pontaroza, and Zuishi, respectively. Abstract: We describe a protocol for somatic embryogenesis (SE) induction from an adult wild olive tree (Olea europaea ssp. europaea var. sylvestris. The protocol used confirms for the first time that there is no need to use juvenile or rejuvenated material for SE induction. For SE induction, petiole and leaf (proximal, intermediary and distal zones) explants were grown on Murashige and Skoog (MS) or Olive Medium (OM) media with different combinations of plant growth regulators (PGR): α- naphthaleneacetic acid (NAA), Zeatin (Zea), indole-3-butyric acid (IBA), 2-isopentyl adenine (2iP), thidiazuron (TDZ) and 6-benzylaminopurine (BAP). All media had 30 g/l sucrose and 7 g/l agar, and the pH was adjusted to 5.8. Cultures were incubated in the dark and, after 3 months, they were transferred to MS medium without PGR for expression. Petiole explants gave the highest callus production, while for SE induction and expression distal blade leaf and petiole explants gave the highest rates. The best medium for SE induction was MS with 12.25 μM IBA plus 4.56 μM Zea. Histological analyses confirmed the individuality of globular somatic embryos. This is the first report of SE expression in explants without rejuvenation in Olea genus, and opens perspectives for using this strategy in SE protocols both for this wild genotype and for commercial genotypes.

Abstract: The change in the levels of endogenous cytokinins in the shoot and grain of rice (Oryza sativa L. japonica cultivar Nihonbare) during its growth and development was followed by selected ion monitoring using deuterium-labelled internal standards. Five cytokinins, zeatin (Z), cis-Z, ribosyl-zeatin (RZ), cis-RZ and 6-(3-methyl-but-2-enylamino)purine riboside were detected in the shoot, root and ear of rice. The maximum contents of RZ and Z in the shoot and root were found at the maximum tillering stage. In the ear, the levels of Z and RZ changed markedly. It is therefore suspected that RZ and Z play some roles in the tillering and seed growth in rice.

Abstract: Regeneration of adventitious shoots of pothos (Epipremnum aureum Linden and Andre) ‘Jade’ was obtained using leaf and petiole explants preprated from shoot tips of 3-yr-old greenhouse-grown plants. Explants were cultured on Murashige and Skoog (MS) basal medium supplemented with thidiazuron (TDZ), 6-(4-hydroxy-3-methy-trans-2-butenyl-amino)purine (zeatin) or N-isopentenylaminopurine (2iP) individually with α-naphthaleneacetic acid (NAA) in 18 combinations. Callus was initiated from cut surface and along the midrib or major vein of leaf sections. Shoot regeneration from leaf and petoole explants occurred in 30d on medium containing 1, 5 or 10μM TDZ with 0.5 or 1.0μM NAA except petioles on medium with 10 μM TDZ and 1.0 μM NAA where regeneration failed. More time (50d) was needed for shoot regeneration when explants were cultured on medium containing either 2iP or zeatin with NAA. Regeneration frequencies were up to 20% and 50% for leaf and petiole explants, respectively. Shoot numbers per responding explant attained 30 for leaf and petiole explants on medium containing TDZ but only one to four on medium containing either 2iP or zeatin. These results indicate that TDZ is a more effective cytokinin for in vitro regeneration of pothos than either zeatin or 2iP.. Shoots elongated readily and rooted well on MS basal medium, without plant growth regulators. Plantlets acclimatized rapidly and grew vigorously in the greenhouse after transfer to pots containing a commerecial potting medium.
