Abstract: An Agrobacterium tumefaciens-mediated transformation method was developed for Salix matsudana Koidz. using mature seeds as starting material. Multiple shoots were induced directly from embryonic shoot apices of germinating seeds. Although thidiazuron, 6-benzylaminopurine and zeatin induced multiple shoot induction with high frequency, zeatin (4.5 µM) was more effective for elongation of shoots and roots. The binary vector pCAMBIA1303, which contained neomycin phosphotransferase as a selectable marker gene and β-glucuronidase as a reporter gene, was used for transformation. Factors affecting transformation efficiency were examined for optimization of the procedure. Up to 35 of 180 seeds regenerated kanamycin-resistant shoots under optimal transformation conditions as follows: seeds were precultured for 4 days, apices of embryonic shoots were removed and infected with A. tumefaciens strain LBA4404 grown to a cell density equivalent (OD600) of 0.6, and then the infected explants were cultivated at 21 ° C for 4 days. Storage of seeds at –20 ° C for as long as 3 years had no significant effect on the induction of kanamycin-resistant shoots. Using this method, transgenic plants were obtained within ~5 months with a transformation frequency of 7.2%. Analysis by polymerase chain reaction (PCR) showed that 36.4–93.8% of plants from all 13 tested kanamycin-resistant lines were PCR positive. Several ‘escapes’ were eliminated by a second round of selection. PCR, Southern blot and reverse transcriptase–PCR analyses of selected transgenic individuals 2 years after cutting propagation confirmed the successful generation of stable transformants. Our method, which minimizes the duration of axenic culture, may provide an alternative procedure for transformation of other recalcitrant Salix species.

Abstract: The endogenous levels of isopentenyladenine, isopentenyladenosine, zeatin and zeatin riboside and the ability forin vitro axillary shoot organogenesis and rhizogenesis were compared between mature and rejuvenated clones ofHevea brasiliensis (Mull. Arg.). Enhancement of thein vitro organogenesis ability of rubber-tree clones following somatic embryogenesis or repeated grafting onto juvenile rootstocks was accompanied by an increase of zeatin riboside levels in shoots used as starting material forin vitro micropropagation. Furthermore, the zeatin level, inin vitro shoots of clones treated byin vitro micrografting, and consequently capable of axillary shoot and root organogenesis, was higher than inin vitro shoots of non treated mature material incapable of in vitro organogenesis. We conclude that the endogenous zeatin-like cytokinin level (free and ribosylated forms) can be considered as a reliable marker for the recovery ofin vitro shoot and root organogenesis after rejuvenating treatments in rubber-tree clones.

Abstract: Kinetin has been shown to act synergistically with indole-3-acetic acid (IAA) or calcium ion (Ca2+) to stimulate ethylene production. Several commercially available cytokinins (kinetin, kinetin-riboside, benzyladenine, benzyladenine-riboside, isopentenyladenine, isopentenyladenine-riboside, and zeatin) as well as noncytokinin bases (adenine and xanthine) were administered to mung bean (Phaseolus aureus Roxb.) hypocotyls to study their effects, alone or in combination with IAA or Ca2+, on ethylene production. In the presence of IAA or Ca2+, all cytokinins tested synergistically stimulated ethylene production and were as effective or nearly as effective as kinetin. Noncytokinin bases (adenine and xanthine) were, however, inactive in this system.

Abstract: Endogenous cytokinins (CK) were measured in Cucurbita pepo L. (zucchini) cotyledons after spraying 7-day-old seedlings with methyl jasmonate (MeJA) to examine whether the senescence-promoting action of MeJA is associated with changes in metabolic regulation of CK levels. MeJA promoted senescence as estimated by the loss of chlorophyll and injury to chloroplast ultrastructure. This was already detected one day after treatment. The contents of physiologically active CKs determined by high-performance liquid chromatography-mass spectrometry (CK bases: trans-zeatin, dihydrozeatin and isopentenyladenine and their ribosides), especially trans-zeatin and its riboside declined considerably. CK nucleotides and physiologically inactive CK 7- and 9-glucosides were also markedly decreased whereas the content of storage CK O-glucosides as well as cis-isomers of zeatin declined only slightly. Chlorophyll content and the normal chloroplast structure recovered 5 days after the MeJa treatment. The process of photosynthetic apparatus recovery correlated with an increased concentration of physiologically active CKs, especially Z. In contrast, the content of CK nucleotides and storage CK O-glucosides decreased further. We suggest that the ability of MeJA to promote some aspects of senescence in intact zucchini cotyledons is at least partially due to down-regulation of endogenous CK levels and their interconversion between active and inactive forms.
