Abstract: In vitro shoot multiplication of cucumber (Cucumis sativus) was examined from the nodal explants of 10-day-old aseptic plantlets using Murashige and Skoog (MS) media supplemented with different concentration (0, 0.5, 1, 2, 3 mg L-1) of cytokinins (6-Benzylaminopurine-BAP, Kinetin-Kn, Thidiazuron-TDZ, and Zeatin). Nodal explants of cucumber showed shoot induction and multiplication in response to all cytokinins tried. MS medium containing Kn was the most effective for inducing shoots from nodal explants of cucumber. The maximum rate of regeneration (83%), the highest number of obtained shoots (7.93 shoots/explant) and the longest shoots (3.61 cm) were obtained on MS medium fortified with 1 mg L-1 Kn. The lowest culture responses were recorded for media supplemented with either BAP or Zeatin. In conclusion, using Kn is strongly recommended than using Zeatin, TDZ and BAP to obtain the highest percentage of regeneration, the highest number of shoot/explant, and the highest shoot length for nodal explants of cucumber.

Abstract: Zeatin and zeatin-9, beta-ribonucleoside enhance the germination of dormant ash embryos. While the first macroscopic signs of germination appear only after about 72 hours, 12 hours of exposure to 50 mum zeatin is as effective as continuous incubation. There must be barriers against transport out of the embryos since 8-(14)C-zeatin and its metabolites, zeatin-9, beta-ribonucleoside, the 5'-mono and the suspected di- and triphosphates, accumulate against a concentration gradient. Zeatin ribonucleoside is about as effective as zeatin in enhancing embryo germination, yet the internal 8-(14)C-zeatin level is lower by a factor of about 50 when the ribonucleoside is fed. The physiological effects of zeatin and abscisic acid on the germination of ash embryos are antagonistic. There is, however, no evidence that abscisic acid has a significant effect on 8-(14)C-zeatin uptake or conversions.

Abstract: Antisera have been raised in rabbits against dihydrozeatin riboside and isopentenyladenosine, and their cross-reactivity characteristics have been examined in detail. These antisera, together with an antiserum previously raised against zeatin riboside, have been employed in radioimmunoassays. Separative procedures that enable a wide range of naturally occurring cytokinins to be separated prior to analysis by radioimmunoassay have been developed. The accuracy with which the following cytokinins can be quantified by our methods, which employ tritiated cytokinin recovery markers, has been estimated: zeatin riboside, zeatin, dihydrozeatin riboside, dihydrozeatin, O-glucosyl zeatin riboside, O-glucosyl zeatin, O-glucosyl dihydrozeatin riboside, O-glucosyl dihydrozeatin, zeatin-9-glucoside, zeatin-7-glucoside, lupinic acid, isopentenyladenosine, and isopentenyladenine.
