Abstract: Micropropagation of Coffea arabica cvs. Oudayni, Hammady and Dawaeiry from Yemen were initiated from seeds. Seeds were surface sterilized and inoculated into media supplemented with different salt strengths and germinated under dark. Seeds germinated on agar medium gave high hypocotyl length, high root length and full cotyledonary leaves expansion after 120 days of culture. Proliferation of these cultivars was experimented on MS media supplemented with different levels (0.0, 2.0, 4.0. 6.0 or 8.0 mg l ) of 1 N6-Benzyladenine (BA), Thidiazuron (TDZ), 6-furfurylaminopurine (Kinetin), 6-(4-hydroxy-3-methyl-2- butenylamino) purine (Zeatin) or 6-( , -Dimethylallylamino) purine (2ip). Highest proliferation for all cultivars was obtained when BA was used at the highest level (8.0 mg l ). Satisfactory proliferation rate in the three 1 cultivars was achieved at 8.0 mg l kinetin and 6.0 mg l TDZ. Zeatin and 2ip were both failed to promote 11 proliferation at any used level. Rooting was experimented on half-strength MS media supplemented with different levels (0.0, 2.0, or 3.0 mg l ) of indole-3- butyric acid (IBA), indole-3- acetic acid (IAA) or 1

Abstract: An antiserum against the cytokinincis-zeatin riboside was raised in rabbits and characterized for use in radioimmunoassays. Cross-reactivity studies demonstrated the specificity of the selected antiserum forcis-zeatin riboside andcis-zeatin in preference to a range of cytokinins and other purines. HPLC systems were developed that separatedcis-zeatin andcis-zeatin riboside from zeatin/dihydrozeatin and zeatin riboside/dihydrozeatin riboside, respectively. These systems enabled the separation of these compounds in xylem sap samples of wheat and oats and their quantification using radioimmunoassay. A TLC system for the separation ofcis-zeatin andcis-zeatin riboside from zeatin/dihydrozeatin and zeatin riboside/dihydrozeatin riboside, respectively, is also described.

Abstract: A method for propagation of tulip in vitro is reported. Shoots were regenerated from stem explants placed on medium containing 5/iM zeatin and 5ftM a-naphthaleneacetic acid. However, about 90% of the structures formed under these conditions did not contain a meristem and could not be used for propagation. Meristem formation on stem explants was slightly improved by silverthiosulphate and strongly by paclobutrazol or methyljasmonate. The propagation rate under standard conditions was about 1.5 in ten weeks and increased after preculture of shoots in liquid medium.

Abstract: Using chromatographic, chemical, and enzymic techniques, 11 compounds capable of stimulating the division of soybean callus cells were tentatively identified in the root sap of Bougainvillea `San Diego Red.9 These cytokinin-like compounds included phosphorylated and glucosylated forms of zeatin, ribosylzeatin, and their dihydro derivatives. In addition, isopentenyl adenosine and isopentenyl adenine were apparently also present. The occurrence of glucosylated derivatives in the root sap of plants has not been substantiated previously.

Abstract: Studies were performed on the growth and the alkaloid content of stem callus cultures of Cinchona pubescens Vahl concerning the influence of plant growth regulators in the medium. After two months the increase of fresh weight of cultures grown in the dark was less than those incubated with illumination. A number of quinoline and indole alkaloids could be detected by means of TLC. Media F (zeatin: 1 microM, indole-3-butyric acid: 1 microM) and H (zeatin: 1 microM, 2,4-dichlorophenoxy acetic acid: 1 microM) showed the best alkaloid production and tissue growth.
