Abstract: Cytokinin-like substances were produced in higher amounts by soil isolates than by those isolated from the rhizosphere and mycorrhizosphere. However more bacterial strains isolated from the rhizosphere and mycorrhizosphere were capable of synthesizing cytokinin-like compounds. A distinct effect of pH on the production of these substances was found. It is suggested that zeatin and zeatin riboside are likely to be among the cytokinin-like substances detected.

Abstract: Introduction of the gene for cytokinin synthesis into potato genome lead to a manifold increase in the level of cytokinins (zeatin, zeatin riboside, isbpentenyl-adenine, isopentenyladenosine) in plantlets grownin vitro.The increasing cytokinin level was associated with increasing tendency to teratoma formation, to decreasing leaf net photosynthetic rate and to increasing dark and light respiration rates and CO2 compensation concentration. During plantlet (or teratoma) ontogeny, net photosynthetic rate increased simultaneously with the decrease in cytokinin level. High level of endogenous cytokinins was associated also with lower photochemical activities of both photosystems in isolated chloroplasts.

Abstract: Leaf protoplasts of axenic shoot cultures of Lonicera nitida cv Maigrun underwent sustained division to give multicellular colonies (microcalli) on a modified, ammonium-free MS (Murashige & Skoog) medium containing 0.5 mg l-1 NAA (1-naphthaleneacetic acid), 1.0 mg l-1 BAP (6-benzylaminopurine) and 150 mg l-1 casein enzymatic hydrolysate. Callus was produced upon transfer of cell colonies to MS medium with 2.0 mg l-1 NAA and 0.2 mg l-1 BAP. About 110 days from isolation protoplast-derived shoots were regenerated on a half-strength MS medium with 0.01 mg l-1 NAA, 5.0 mg l-1 BAP, 0.5 mg l-1 zeatin and a complex mixture of group B vitamins. The replacement of such mixture by 250 mg l-1 casein enzymatic hydrolysate promoted rhizogenesis in calli, with shoot buds being subsequently regenerated from the protoplast-derived roots. Micropropagation of protoplast-derived shoots (of either origin) was difficult, due to a strong apical dominance, but could be accomplished by transferring single-node explants to half-strength MS medium with 1.5 mg l-1 BAP. Such shoots were, in turn, successfully rooted and transferred to the glasshouse where they completed acclimatization.

Abstract: Gagaminine, a steroidal alkaloid isolated from the roots of Cynanchum wilfordii, exhibited potent inhibitory effects on the aldehyde oxidase activity and lipid peroxidation. To find a possible means of mass production of this active component, which will be useful for animal tests, we synthesized it by an in vitro culture method using various growth conditions. Calli were induced from the explants of this medicinal plant and cultivated under culture conditions which varied in light, and the kinds and concentration of plant growth regulators. The production of gagaminine was found to be significantly higher in the dark than in the light. The best gagaminine content (0.960%) was obtained after cultivation of stems on the medium containing 2,4-dichlorophenoxyacetic acid (2,4-D, 2.0 mg/l) only. However, gagaminine was not detected by the mixture of 2,4-D and kinetin, while the mixtures of 2,4-D/zeatin and 2,4-D/6-benzylaminopurine produced a low content of gagaminine (<0.4%). In addition, suspension medium was much better for the formation of gagaminine than solid medium with an increase from 0.960 to 2.227% yield. These results suggest that gagaminine can be produced massively by in vitro culture using stems under the conditions of dark and 2,4-D on liquid medium.
