Abstract: When cultured in vitro, roots of four Japanese persimmon (Diospyros kaki L.) cultivars formed adventitious shoots on MS medium with 10 μM zeatin and 0.01 μM indole-3-acetic acid, although their organogenetic capacities varied. Histological study revealed that the origin of the adventitious shoots was the pericycle. The regenerated shoots grew well on the shoot proliferation medium (MS with 5 μM zeatin). Final rooting percentages of shoots regenerated from roots of three of the four cultivars were greater than those of shoots that originated from shoot tips and that had been subcultured >50 times. Shoots regenerated from Jiro' roots rooted 10 days earlier, had more roots than those from shoot tips, and maintained higher rooting ability over ten subcultures. Rooted 'Hiratanenashi' shoots regenerated from roots survived better after acclimatization than those from shoot tips. No obvious variants were observed either in vitro or in the field. The trees regenerated from roots flowered within 4 years. These findings suggest that partial rather than true rejuvenation was responsible for both the early flowering and the juvenile characteristics, i.e., the enhanced rooting ability, observed in the regenerated plants. Abstract: An efficient, genetically stable and rapid regeneration system via multiple shoot formation was developed for an endangered medicinal herb Solanum surattense . Leaves were cultured on MS medium supplemented with different concentration of cytokinins; 6-benzylaminopurine (BAP), kinetin (Kn), thiadiazuron (TDZ), zeatin and inorganic phosphate (iP) alone or in combinations. Among five cytokinins used, TDZ (0.1 mg/l) gave the maximum regeneration response (35 shoot buds per explant), with 100% regeneration efficiency. Comparative regeneration efficiency of cytokinins was in the following order: TDZ > zeatin > iP > BAP > Kn and the comparative ratio was 6:4:3:2:1. Highest frequency of adventitious shoot bud induction (54 shoot buds per explant) was achieved on combination of TDZ (0.1 mg/l) and BAP (1 mg/l). Combinations of cytokinins were more effective for shoot bud regeneration as compared to cytokinin alone. Best root formation was observed on Murashige and Skoog (MS) medium supplemented with 1 mg/l indole-3-acetic acid (IBA). The rooted plantlets were hardened gradually and resulted in a production of more than 80% plantlets. The efficiency of auxins for root organogenesis was in the following order: IBA > IAA > NAA > PAA. Genetic stability of regenerated plants was analyzed using random amplified polymorphic DNA (RAPD) markers. Abstract: The cultivars of barley and wheat resistant to mildew had a higher level of free zeatin and its derivatives during the whole ontogeny than the susceptible cultivars. The effect of exogenous application of cytokinins on the growth ofErysiphe graminis DC. was different. Kinetin and benzylaminopurine showed only a slight inhibitory effect. Zeatin and its derivatives completely inhibited growth of this fungus.

Abstract: Blueberries have a high nutritional value and strong antioxidant properties, qualities which account for their increasing request on the food and pharmaceutical markets. Cultivar 'Elliot' has a balanced content of active substances. It is also a late ripening cultivar suitable for long Transylvanian winters, which makes it a good candidate for large scale cultivation in this region. Because traditional propagation methods of highbush blueberry have several drawbacks, studies for creating an efficient micropropagation protocol were initiated. Dormant buds were used as initial explants for shoot regeneration while axillary and adventitious microshoots were further used for the proliferation stage. The influence of several plant growth regulators (2-iP, Zeatin, BAP) in relation with the explant positioning into media was investigated in order to ensure the highest proliferation rate. WPM supplemented with zeatin 2 mg l -1 and 2-iP 5 mg l -1 along with horizontal microshoot positioning gave the best proliferation rate, with an average of 17.9 shoots/explant. In vitro obtained leaves were tested for their regeneration capacity. The use of 2-iP 10 mg l -1 produced up to 90% regeneration rate. The best variant consisted in abaxial leaf positioning which resulted in 50% direct regeneration rate and an average of 6.5 shoots/initial explant. The maximum proliferation rate was recorded for adaxial positioning (26 shoots/initial explant). Direct ex vitro acclimatization and rooting in hydroculture were also tested with very good results.
