Abstract: We studied the effects of various cytokinins and urea derivatives on germination of aged seeds of in Lotus corniculatus L. The following substances were applied: N6-isoprenoid cytokinins (isopentenyl adenine and zeatin), adenine sulfate, N6-aromatic cytokinins (kinetin, benzyladenine) and their N9-ribosides, N-benzyl-9-(2- tetrahydropyranyl)adenine, and urea derivatives (diphenylurea, thidiazuron, and chloro-pyridyl phenylurea). With the exception of adenine sulfate, all cytokinins increased the percentage of seed germination up to twofold, depending on their kind and concentration. It is concluded that cytokinins may be among the missing factors in aged seeds of L. corniculatus contributing to the implementation of their full germination potential. They could be used to improve germination of both freshly harvested and aged seed samples, if necessary. .

Abstract: Premise of the Study Vasconcellea pubescens is an important Caricaceae species cultivated in several countries of South America. The objective of this study was to investigate different media compositions and plant growth regulators to induce plant regeneration. Methods Anthers were cultured in Murashige and Skoog medium with varying concentrations of naphthalene acetic acid (NAA) and 2,4-dichlorophenoxyacetic acid (2,4-D) plus a cytokinin (N-(2-chloro-4-pyridyl)-N'-phenylurea). The effect of the basal medium supplemented with auxins and cytokinins on shoot regeneration from the induced calli was also evaluated. Addition of maltose to the basal medium was also tested. Results The combination of 0.54 μM NAA and 22.66 μM 2,4-D induced the highest rate of calli formation. Regeneration via organogenesis was obtained in Murashige and Skoog and Woody Plant Medium supplemented with maltose and containing 8.88 μM 6-benzylaminopurine, 5.71 μM indoleacetic acid, and 2.28 μM zeatin. Discussion The plant regeneration protocol reported here permits the development of haploid and double haploid plants that can be useful for propagation purposes, allow a better molecular understanding of the species, and facilitate the production of new cultivars.

Abstract: A procedure for isolation and culture with high viable protoplast yield from axenic shoot culture of potato (Solanum tuberosum L. cv May Queen) has been developed. The isolated protoplasts, cultured in a suitable medium, regenerated cell-wall within 48hr, initiated DNA synthesis in 76-96hr, and divided to produce cell colonies. For regeneration of whole plants, cell colonies cultured with IAA and zeatin gave the highest frequency of shoot-bud formation at 20°C under 4, 000 lux of irradiance.

Abstract: Organogenesis in thin cell layers of Nicotiana tabacum L. was studied in relation to the effects of natural and synthetic auxins in combination with various cytokinins. All cytokinins tested, benzyladenine (BA), kinetin, zeatin (Z), zeatin riboside (ZR), N6-Δ2-isopentenyl) adenine (IPA), dihydrozeatin [(diH)Z] and dihydrozeatin riboside [(diH)ZR], seem to be active in flower bud formation. In addition to the initiation of flower buds, vegetative buds or roots were also formed on the explants in the presence of BA, Z or IPA as exogenous cytokinins. Only dihydrozeatin and its riboside stimulated the initation of flower buds alone (as is known for kinetin), especially if supplemented with indole-3-acetic acid (IAA) as exogenous auxin. A high number of explants with flower buds was also found with high cytokinin/2,4-D ratios. In these conditions the presence of (diH)Z yielded the higest number of flower buds per explant.
