Abstract: Dendrobium heterocarpum Lindl. is a native orchid in Indonesia. Its population continue to decline due to unsustainable exploitation. One solution to increase its population is via in vitro technique propagation. This research aimed to determine effect of hormone addition (Zeatin and NAA) to induce shoot multiplication of D. heterocarpum Lindl. grown in Murashige and Skoog (MS) basic medium. The research was conducted in Tissue Culture Laboratory, Department of Biology, Faculty of Mathematics and Science, Udayana University from November 2015 until March 2016. The experiment employed Completely Randomized Design (CRD) with 20 combinations of treatments, with three replicates each treatment. The treatment werer addition of Zeatin and NAA, with five concentrations of Zeatin (0, 0.1, 0,5, sweet corn and hybrids corn as the natural Zeatin); four NAA concentrations (0, 0.05, 0.1 and 0.5). Variable recorded include such as the number of shoots, roots and explant’s vigor. The quantitative data which include number of leaves and shoots after 12 weeks, were analyzed using Analysis of Variance (ANOVA); and if significantly different, tested further using Duncan Multiple Range Test (DMRT) at 5% level. Z 5 N 3 medium revealed significantly different on amount of leaves and shoot height and also the best medium on amount of roots. Best hormone combination that can induce multiplication of new shoots are medium Z 4 N 3 and Z 5 N 1 .

Abstract: PURPOSE:To increase the callus formation rate from plant tissues and the plant body redifferentiation rate from callus by treating the plant tissue with an aqueous solution containing natural or synthetic auxin, before inoculation of the plant tissue in the culture medium. CONSTITUTION:A plant tissue such as pollen, seed embryo, young leaf is disinfected with ethanol or sodium hypochlorite and treated with an aqueous solution containing natural auxin such as indole-acetic acid or synthetic auxin such as 2,4-dichlorophenoxyacetic acid. The aqueous solution can additionally contain a natural cytokinin such as zeatin or a synthetic cytokinin such as kinetin. Then, the treated plant tissue is inoculated in a culture medium to form callus and the callus is continuously cultured to obtain the redifferentiated plant bodies efficiently.

Abstract: Nodal regments of tropical fruit Averrhoa carambola (Oxalidaceae) were used as explants to investigate optimum conditions for rapid propagation. Among cytokinins (1.0 mg L-1), zeatin (1.0 mg L-1) was best for culture, and kinetin (1.0 mg L-1) the worst. MS medium supplemented with 0.5 mg L-1 BA, 0.2 mg L-1 NAA and 0.2 mg L-1 GA was optimal for the plantlet growth, the proliferation coefficient being as high as 3.7, and the rate of abnormal plantlets was only 23.3%. As for rooting, half-strengh MS medium containing 0.2 mg L-1 IBA, 0.1 mg L-1 IAA and 2%-3% sucrose was the best. The rooting rate was 87.1%.
