Abstract: Tacca leontopetaloides is a tuberous species with nutritional, medicinal, and industrial potential. The aim of the present study was to develop a simplified micropropagation protocol for this species, combining phases of in vitro propagation and rooting, and to assess the genetic stability of in vitro regenerants using ISSR and flow cytometric analysis. In the first step, the effect of four cytokinins (BA, zeatin, kinetin, thidiazuron) at concentrations of 0.1-0.7 mg l-1 on shoot organogenesis was tested. The highest number of shoots was achieved on MS medium supplemented with 0.1 mg l-1 zeatin (4.11 ± 0.35 shoots/explant). Thereafter, this treatment was combined with auxins (NAA, IAA) at concentrations of 0.01-0.07 mg l-1 to study shoot organogenesis and in vitro rooting. A combination of plant growth regulators provided unchanged shoot organogenesis, while the root number per explant increased. The highest number of roots of all tested treatments was produced on MS medium supplemented with zeatin (0.1 mg l-1) in combination with NAA (0.05 mg l-1) (5.00 ± 0.38 roots/explant), suggesting that this medium may be used for both propagation and rooting. Well-rooted plants were transferred ex vitro, with a 97.78% survival rate. Ten randomly chosen in vitro regenerants were subjected to ISSR and flow cytometric analysis. Eleven ISSR primers produced 40 clear and reproducible bands per sample. All amplified products were monomorphic; no polymorphism was detected. Similarly, the results of flow cytometric analysis showed no variation in ploidy level. Thus, the micropropagation method optimized here can be used for highly effective production of true-to-type plants of T. leontopetaloides.

Abstract: A competitive hyper-immune yolk Immunoglobulin Y - Enzyme Linked Immune Sorbent Assay (CIgY-ELISA), was developed as an alternative method to detect zeatin and 2ip in plantlets of gerbera. The endogenous level of hormones in the plantlets in vitro of gerbera with one or six weeks after replication was determined with competitive IgY-ELISA set to detect between 1 and 100 pmoles of plant hormone for each 1.0 g tissue. The plantlets of six weeks presented sprouts and root, while the plantlets of one week presented only sprouts. The CIgY-ELISA was set with high independent variables values of sensitivity/specificity of 96/89 % for zeatin and 94/78 % for 2ip, with high values of reproducibility (up to 90 %) for both the cytokinins. Zeatin content varied from 2.2 to 2.8 pmoles.g-1 and from 2.7 to 3.3 pmoles.g-1 on the plantlet with one and six weeks, respectively. The 2ip content did not vary and was detected near the detection limit in all the assays. It was concluded that the observed capabilities of CIgY-ELISA were putative and the competitive assay was a highly robust and stable method, which could be used for the studies on plant physiology for endogenous cytokinins.

Abstract: The optimal condition of in vitro regeneration and transformation were investigated for newly bred potato varieties in Korea. Leaf and internodal stem tissues of `Chubak`, `Namsuh`, `Jasim`, `Jopung` and `Jowon` were used to investigate the influence of growth regulator on regeneration efficiency The effect of phenolic compound acetosyringone on gene transformation efficiency was also investigated. Leaf tissue of `Jowon` and internodal stem tissues of `Jopung` were showed high regeneration efficiency on M5 medium supplemented with 0.1 mg/L GA₃, 2.0 mg/L Zeatin and 0.01 mg/L NAA. The other three potato cultivars were showed low regeneration efficiency less than 25%. The effect of acetosyringone on Agrobacterium-mediated gene transformation with leaf and internodal stem tissues were noticeable. By adding the 75 μM of acetosyringone during the Agrobacterium innoculation, the transformation efficiency was increased up to 1.5∼4.0 fold compare to non-treated control. In case of `Jowon` the transformation efficiency was 87.9% in leaf tissue and `Jopung` was 68.4% in internodal stem tissues.
