Abstract: In vitro micropropagation of Carob tree (Ceratonia siliqua L.) by cotyledonary buds taken from 13 day old
seedlings was attempted on Woody Plant Medium (WPM) supplemented with microelements and Murashige and
Skoog (MS) vitamins. Comparison of the effect of four cytokinines (benzylaminopurine, zeatin, kinetinand 2-iP)
has shown that seedling growth was stimulated by zeatin at 1 mg/L, while organogenesis was better under BAP or
zeatin at 1 mg/L. The effect of four auxins (AIA, AIB, ANA and 2,4-D) at low concentrations (0.1 mg/L)
combined with BAP at 0.5 mg/L has shown that Naphthalene Acetic Acid (NAA) stimulates better stem
formation and growth. Moreover, combination of Gibberellic Acid (GA3) at different concentrations with BAP at
0.5 mg/L did not show a significant effect on growth. Shoot multiplication was stimulated by medium
concentrations of BAP (0.5 - 1 mg/L) alone or with GA3 at 0.2 mg/L. Among the auxins tested during rooting,
Indol-3-Butyric Acid (IBA) (2 mg/L) was found to be the most efficient for root formation.

Abstract: (E)-4-Hydroxy-3-methylbut-2-enyl diphosphate (1) is a key intermediate of the deoxyxylulose phosphate pathway of isoprenoid biosynthesis and a precursor of the plant hormone zeatin. The availability of this intermediate with various labeling patterns is pivotal for its use in biosynthetic studies. The number of positions, however, that can be easily labeled by chemical synthesis is limited, and the synthesis by means of recombinant enzymes is laborious and time consuming. We demonstrated that chromoplasts from Capsicum annuum, whose enzyme activity was impaired by freeze-thawing, accumulate 1. This observation built the basis for the development of a cell-free system allowing the synthesis of this intermediate with labels in various positions. With 2C-methyl-D-erythritol 2,4-cyclodiphosphate (5) as substrate, yields were in the range of 50%.

Abstract: Calli were induced by culturing the leaf segment of Actinidia deliciosa A. arguta clone 118 on MS medium supplemented with 0.5 mg/L 2,4-D, 0.1 mg/L NAA and 0.05 mg/L BA for 8 weeks in light condition. The induced calli were inoculated in liquid MS medium containing 0.5 mg/L 2,4-D, 0.1 mg/L NAA, 0.05 mg/L BA and 3% sucrose to establish cell suspension culture. The cells at the exponential stage and the stationary stage could be observed between 5-11 days and after that 12 days in culture, respectively. The fresh weight of callus induced from the suspended cells did not vary much among the media containing eight different combinations of plant growth regulators tested. The highest frequency of shoot induction (88.3%) was observed in MS medium containing 2.0 mg/L zeatin. Either BA or zeatin mixed with thidiazuron (TDZ) seemed to be effective in shoot induction. The induced shoots were transferred to MS medium containing 0.2 mg/L zeatin for further shoot growth. And then the shoots were transferred to Standardi (ST) medium containing 1.0 mg/L indolebutyric acid (IBA) for rooting. Plantlets could be obtained through cell suspension culture of Actinidia deliciosa A. arguta clone 118.
