Abstract: A process for the production and maintenance of pluripotent and/or totipotent progenitor cereal cells from undifferentiated callus cells is described. Production of the progenitor cells takes place via direct organogenesis on a medium containing at least one auxin and at least one cytokinin. For example, the auxin may be 2,4- dichlorophenoxyacetic acid, indole-3-acetic acid, picloran, naphthelenacetic acid, indole-3-propionic acid, indole-3-butyric acid, phenyl acetic acid, benzofuran-3-acetic acid or phenyl butyric acid, and the cytokinin may be benzyl amino purine, benzyladenine, thidiazuron, zeatin, isopentyladenine, trans-zeatin or dimethylallyladenine. Processes for transformation of the undifferentiated callus cells and/or the progenitor cereal cells are also described. Typical cereal cells are sorghum, maize, wheat, barley, millet, rye, canola, alfalfa, triticale and rice.

Abstract: The invention provides a tissue culture and rapid propagation method of blueberry. The method is characterized in that semi-lignified stems of the blueberry are used as explants to be inoculated to a culture medium containing improved READ culture medium, zeatin, naphthylacetic acid, saccharose and agar to perform connection culture; new tips are transplanted into a culture medium containing the improved READ culture medium, zeatin, saccharose and agar to perform continuous propagation culture; the regeneration seedlings are cut into small sections and then transplanted to a composite substrate, so as to achieve ex-vitro rooting. According to the method, zeatin is used as a plant growth modifier at the stages in a tissue culture vessel, including primary generation induction and subculture multiplication; zeatin has the effects of promoting cell division and cell growth, and is small in dosage and high in multiplication coefficients; the plants are strong and can be directly applied to the ex-vitro rooting process without strong sprout cultivation; meanwhile, the ex-vitro rooting technology is performed, which is high in rooting speed, high in efficiency, low in cost, and high in propagation speed compared with the test-tube rooting technology.

Abstract: This study was conducted to show the effect of transformation of potato (Solanum tuberosum) plants with the isopentenyltransferase (ipt) gene on plant morphology and cytokinin levels in leaves and tubers. The ipt gene, which catalyzes the rate-limiting step of the cytokinin synthesis pathway, was placed under the control of a tuber-specific patatin promoter and introduced into potato plants (cv. Norchip) via Agrobacterium-mediated transformation. The presence of the ipt gene in transgenic potato plants was confirmed by Southern hybridization. Shoot morphology was normal in nearly all transgenic plants prior to and after tuberization. Transgenic plants had extensive stolon growth and an increased number of tubers. Premature sprouting of tubers and an increase in the number of sprouts were also observed in transgenic plants. Total fresh and dry weight of tubers was similar in control and transgenic plants. Expression of the ipt gene was tuber specific, as determined by northern hybridization and cytokinin analysis. The concentration of zeatin and zeatin riboside was increased in tubers of transformed plants, but the level in source leaves was similar in transformed and control plants. This study supports a role for cytokinin in tuber initiation and release of dormancy.

Abstract: This experiment was carried out in the lath house of the department of Horticulture , landscaping / college of Agriculture / University of Baghdad during 10/04/2012 to 01/11/2013. Abstract: The establishment of an efficient adventitious shoot regeneration system from leaf discs and stem segments of one-year-old Russian olive(Elaeagnus angustifolia L.) was established.Shoot regeneration using leaf explants and stem segments was obtained on MS medium supplemented with different cytokinins viz.,6-benzyladenine(BA) and Zeatin(Zt),with combination of auxins viz.,naphthaleneacetic acid(NAA) andindole-3-butyric acid(IBA).MS + 0.5mg/L 6-BA + 0.2mg/L NAA was found to be a better combination for leaf regeneration,producing a maximum of 4.3 shoots per explant,whereas MS + 1.0mg/L Zt + 0.5mg/L NAAfor stem-segment explants produced the highest number of multiple shoots(3.6) per explant.The regenerated shoots produced 100% rooting on 1 /2 strength MS medium supplied with 0.5mg/L NAA.In vitro raised plantletswere successfully transferred to pots containing sterile soil∶ peat moss∶ sand(1∶ 1∶ 1) for acclimatization,and 77% of rooted plantlets survived under ex situ conditions.The currently employed regeneration protocol through leaf explants and stem segments facilitates germplasm conservation of this medicinally and ecologically importantspecies besides providing a standardized regeneration protocol for genetic transformation studies in the future.
