Abstract: The dynamic changes of the indole acetic acid(IAA), gibberellin(GA-3),zeatin(ZT) and abscisic acid(ABA) contents in flower buds of tobacco cytoplasmic male sterility(CMS) and homolog maintainer lines were determined by high-performance liquid chromatogram.The results showed that from small bud → middle bud → big bud,the content of IAA and ZT in CMS buds were higher than in maintainer line buds.While the content of GA-3 and ABA in CMS buds were lower than in maintainer line buds.It was suggested that the changes of endogenous hormone contents were closely related with the occurrence of cytoplasmic male sterility.

Abstract: Several inbred lines from a radish (Raphanus sativus var. Radicula Pers.) genetic collection spontaneously form tumors on the crop roots during flowering. Here we studied the influence of the Agrobacterium tumefaciens ipt gene, which controls cytokinin biosynthesis, and the Agrobacterium rhizogenes rolC gene, which influences cytokinin metabolism, on tumor formation in the radish lines. Two in planta transformation methods – transformation of ovaries via pollen-tube pathway and transformation of seedling apexes – were firstly used to obtain transgenic radish plants. Both methods showed high efficiency for radish. Transformation of radish lines by both ipt and rolC genes induced tumor formation in non-tumorous lines. We noted a several-fold increase of free zeatin concentration in ipt-transgenic radish plants while rolC-transgenic radish plants showed an increase in zeatin riboside concentration comparing with the untransformed line. We suppose that the tumorigenic effect of ipt and rolC genes on radish lines is likely to be due to change of phytohormonal balance in the tissues of transgenic plants.

Abstract: Effect of cytokinins (BA, TDZ, zeatin, 2iP, and kinetin) applied either singly or in combination on in vitro growth of two grape cultivars (`Cabernet Sauvignon` and `Campbell Early`) was investigated as a serial work for mass production of grapevine nursery stocks. In single treatment, shoot growth of two cultivars was most favorable in control. Shoot proliferation was satisfactory with 10 M BA regardless of cultivars and cytokinin combinations, followed by TDZ. Other treatments resulted in very poor or no branching. Total explants ready for subculture produced by 10 M BA outnumbered those by other treatments. TDZ was also effective. TDZ significantly increased the fresh weight and callus formation while shoot growth was unsatisfactory. Shoot growth response of two cultivars in combined treatments was also most favorable in control as was in single treatments. When TDZ was combined with zeatin, 2iP, and kinetin which failed to induce branching, proliferous branching was induced though the shoot number was behind that of single treatments of BA and TDZ. TDZ was very effective for total number of explants and fresh weight, showing 10-fold increase.

Abstract: Zeatin belongs to the derivation of purine, and the C=C is one of the functional groups in the zeatin structure. So, there are trans- and cis- structures for the zeatin molecule. Interestingly, the trans-zeatin has biological activities, but the cis-zeatin has no obvious biological activity. The trans-zeatin is the first separated cell cytokinins from the plant tissue. And the zeatin, especially trans-zeatin has important and extensive applications in many research fields. In this study, the Geometries of the trans-zeatin and cis-zeatin were optimized by using the density function Becke, three-parameter, Lee-Yang-Parr, DFT/B3LYP/6-31G method. Additionally, FT-IR experiments were conducted for the trans-zeatin. The geometry structures and molecular vibrational property of the zeatin were simply discussed.
