Estimation of the molecular weights of proteins by Sephadex gel-filtration.

Inhibition of root elongation and modification of membrane properties are sensitive responses of plants to aluminium. The present paper reports on the effect of AI on lipid peroxidation and activities of enzymes related to production of activated oxygen species. Soybean seedlings (Glycine max L. cv. Sito) were precultured in solution culture for 3–5 days and then treated for 1–72 h with Al (AICI3) concentrations ranging from 10 to 75 μM at a constant pH of 4.1. In response to Al supply, lipid peroxidation in the root tips (< 2 cm) was enhanced only after longer durations of treatment. Aluminium-dependent increase in lipid peroxidation was intensified by Fe2+ (FeSO4). A close relationship existed between lipid peroxidation and inhibition of root-elongation rate induced by Al and/or Fe toxicity and/or Ca deficiency. Besides enhancement of lipid peroxidation in the crude extracts of root tips due to Al, the activities of superoxide dismutase (EC 1.15.1.1) and peroxidase (EC 1.11.1.7) increased, whereas catalase (EC 1.11.1.6) activity decreased. This indicates a greater generation of oxygen free radicals and related tissue damage. The results suggest that lipid peroxidation is part of the overall expression of Al toxicity in roots and that enhanced lipid peroxidation by oxygen free radicals is a consequence of primary effects of Al on membrane structure.

Effect of aluminium on lipid peroxidation, superoxide dismutase, catalase, and peroxidase activities in root tips of soybean (Glycine max)

F. Ginseng (Panax ginseng) is planted in the forest to enhance the natural ginseng resources, which have an immense medicinal and economic value. The morphology of the cultivated plants becomes similar to that of wild growing ginseng (W. Ginseng) over the years. So far, there have been no studies highlighting the physiological or functional changes in F. Ginseng and its wild counterparts. In the present study, we used proteomic technologies (2DE and iTRAQ) coupled to mass spectrometry to compare W. Ginseng and F. Ginseng at various growth stages. Hierarchical cluster analysis based on protein abundance revealed that the protein expression profile of 25-year-old F. Ginseng was more like W. Ginseng than less 20-year-old F. Ginseng. We identified 192 differentially expressed protein spots in F. Ginseng. These protein spots increased with increase in growth years of F. Ginseng and were associated with proteins involved in energy metabolism, ginsenosides biosynthesis, and stress response. The mRNA, physiological, and metabolic analysis showed that the external morphology, protein expression profile, and ginsenoside synthesis ability of the F. Ginseng increased just like that of W. Ginseng with the increase in age. Our study represents the first characterization of the proteome of F. Ginseng during development and provides new insights into the metabolism and accumulation of ginsenosides.

Publisher Summary This chapter discusses the assay of catalases and peroxidases are: (1) catalase assay by disappearance of peroxide; (2) method for crude cell extracts; (3) direct spectrophotometric assay of catalase and peroxidase in cells and tissues; and (4) peroxidase assay by spectrophotometric measurements of the disappearance of hydrogen donor or the appearance of their colored oxidation products. Two methods are described for the catalase assay by disappearance of peroxide are: ultraviolet spectrophotometry and permanganate titration. Ultraviolet spectrophotometryis a method devised, on the basis of the absorption curves for peroxide solutions, for determining the activity of catalase by direct measurements of the decrease of light absorption in the region 230 to 250 mμ caused by the decomposition of hydrogen peroxide by catalase. In the case of method for crude cell extracts, oxygen evolution caused by the decomposition of hydrogen peroxide is measured with the conventional manometric technique. Peroxidase assay by spectrophotometric measurements of the disappearance of hydrogen donor or the appearance of their colored oxidation products includes the guaiacol test and the pyrogallol test.
