cab can be a devastating disease affecting all classes of wheat and other small grains. This fungal disease, also called Fusarium head blight (FHB), has the ability to completely destroy a potentially high-yielding crop within a few weeks of harvest. Lush, green fields become blighted seemingly overnight (Figs. 1 and 2). Frequent rainfalls, high humidities, and/or heavy dews that coincide with the flowering and early kernel-fill period of the crop favor infection and development of the disease. Damage from head scab is multifold: reduced yields, discolored, shriveled “tombstone” kernels (Figs. 3 to 5), contamination with mycotoxins, and reduction in seed quality. The disease also reduces test weight and lowers market grade. Difficulties in marketing, exporting, processing, and feeding scabby grain are experienced. In North America, Fusarium graminearum Schwabe (teleomorph Gibberella zeae (Schwein.) Petch; synonym = G. saubinetti) predominates among several Fusarium species that can cause scab (4,5,8,40,48,60). Other species may predominate in cooler climates or where crops other than wheat and corn are dominant (8,40,48,60). F. graminearum also is associated with stalk and ear rot of corn and may cause a root rot of cereals. The fungus persists and multiplies on infected crop residues of small grains and corn. The chaff, light-weight kernels, and other infected head debris of wheat and barley, returned to the soil surface during harvest, serve as important sites of overwintering of the fungus. Continued moist weather during the crop growing season favors development of the fungus, and spores are windblown or water-splashed onto heads of cereal crops. Wheat and barley are susceptible to head infection from the flowering (pollination) period up through the soft dough stage of kernel development. Spores of the causal fungus may land on the exposed anthers of the flower and then grow into the kernels, glumes, or other head parts. Excellent descriptions of the disease cycle and spore stages of the causal fungi have been published (4,8,21,40,48). Mycotoxins are frequently associated with the growth and invasion of cereal grains by scab fungi. The most common toxin associated with F. graminearum– infected grain is vomitoxin (deoxynivalenol). Vomitoxin is known to cause vomiting and feed refusal in nonruminant animals and poses a threat to other animals and humans if exposure levels are high (45). The presence of mycotoxins in infected grain further exacerbates the losses that scab can cause. Recent articles have reviewed the epidemiology, management, and history of scab outbreaks in the United States, Canada, Europe, Asia, and South America (5,40,45,48). As these papers indicate, numerous research and survey reports have described the worldwide occurrence and epidemic levels of scab during the past century. Yield loss reports have not always been based on replicated research trials, but extensive surveys of producers’ fields have provided assessments of head blighting severity, which were translated into yield loss estimates. In the United States, scab was found in 31 of 40 states surveyed in 1917, with losses estimated at 288,000 metric tons (10.6 million bushels), primarily in Ohio, Indiana, and Illinois (4). Fusarium head blight (FHB) is a devastating disease of wheat and barley worldwide. Resistant cultivars could reduce damage from FHB. Chinese wheat cultivar Sumai 3 and its derivatives represent the greatest degree of resistance to FHB known. A major quantitative trait locus (QTL) on chromosome 3BS and other minor QTL for FHB resistance have been identified in these cultivars and used in wheat-breeding programs worldwide. Many breeding lines with the 3BS resistance QTL and improved agronomic traits have been developed. In barley, only limited sources of FHB resistance are available, especially in six-rowed barley, and none of them contains a DON level low enough to meet the safety requirement of the brewing industry. Several QTL have been identified for lower FHB severity, DON content, and kernel discoloration and used to enhance FHB resistance in barley. Marker-assisted selection for FHB resistance QTL on 3BS of wheat and on 2H of barley is in progress.

Management and resistance in wheat and barley to fusarium head blight.

During the past decade, numerous studies have been published on molecular mapping of Fusarium head blight (FHB) resistance in wheat. We summarize the relevant findings from 52 quantitative trait loci (QTL) mapping studies, nine research articles on marker-assisted selection and seven on marker-assisted germplasm evaluation. QTL for FHB resistance were found on all wheat chromosomes except chromosome 7D. Some QTL were found in several independent mapping studies indicating that such QTL are stable and therefore useful in breeding programmes. We summarize and update current knowledge on the genetics of FHB resistance in wheat resulting from QTL mapping investigations and review and suggest FHB breeding strategies based on the available information and DNA markers.

QTL mapping and marker-assisted selection for Fusarium head blight resistance in wheat: A review

Wheat and barley are critical food and feed crops around the world. Wheat is grown on more land area worldwide than any other crop. In the United States, production of wheat and barley contributes to domestic food and feed use, and contributes to the export market and balance of trade. Fifteen years ago, Plant Disease published a feature article titled “Scab of wheat and barley: A re-emerging disease of devastating impact”. That article described the series of severe Fusarium head blight (FHB) epidemics that occurred in the United States and Canada, primarily from 1991 through 1996, with emphasis on the unparalleled economic and sociological impacts caused by the 1993 FHB epidemic in spring grains in the Northern Great Plains region. A Unified Effort to Fight an Enemy of Wheat and Barley: Fusarium Head Blight.

Fusarium head blight (FHB, scab) is a fungal disease of wheat and other small cereals that is found in both temperate and semi-tropical regions. FHB causes severe yield and quality losses, but the most-serious concern is the possible mycotoxin contamination of cereal food and feed. Breeding for FHB resistance by conventional selection is feasible, but tedious and expensive. This study was conducted to identify and map DNA markers associated with FHB resistance genes in wheat. A population of 364 F1-derived doubled-haploid (DH) lines from the cross ’CM-82036’ (resistant)/’Remus’ (susceptible) was evaluated for Type II resistance (spread within the spike) during 2 years under field conditions. Marker analysis was performed on 239 randomly chosen DH lines. Different marker types were applied, with an emphasis on AFLP and SSR markers. Analysis of variance, as well as simple and composite interval mapping, were applied. Three genomic regions were found significantly associated with FHB resistance. The most-prominent effect was detected on the short arm of chromosome 3B, explaining up to 60% of the phenotypic variance for Type II FHB resistance. A further QTL was located on chromosome 5A and a third one on 1B. The QTL regions on 3B and 5A were tagged with flanking SSR markers, the 1B QTL was found associated with the high-molecular-weight glutenin locus. These results indicate that FHB resistance is under control of a few major QTLs operating together with unknown numbers of minor genes. Marker-assisted selection for these major QTLs involved in FHB resistance appears feasible and should accelerate the development of resistant and agronomically improved wheat cultivars.

Molecular mapping of QTLs for Fusarium head blight resistance in spring wheat. I. Resistance to fungal spread (Type II resistance)

Resistance of wheat to Fusarium head blight caused by Fusarium graminearum and F. culmorum was identified in natural epidemics in 1985 and 1987 as well after artificial inoculations (1983–1988 and 1984–1987). Out of 25 genotypes tested, five were identified with no significant difference in head blight scores, but differing significantly in yield after artificial inoculation, i.e. tolerance differences were detected at different resistance levels. Some genotypes that were similar in yield or head blight scores differed in seed infection severity. Genotypes with awns were more susceptible to head blight when tested under natural epidemic condition in the field; but this trait did not influence head blight severity in artificial inoculations. Dwarf genotypes were more severely infected by head blight than tall genotypes under natural conditions, but genotypes of different plant height classes were similarly susceptible after artificial inoculations. In the early generations of a breeding programme resistance measured by visual evaluation of artificial inoculation is the most important way to screen. If selection of dwarf and awned genotypes cannot be avoided, the higher susceptibility caused by awns and dwarfness under natural epidemic conditions can be decreased by a higher level of physiological resistance, as variability in physiological resistance is available. In later generations, traits like percentage of seed infection or tolerance can be identified by additionally measuring yield reduction. Stability of disease reaction appears to be connected with resistance level, the most resistant genotypes are the most stable, and the most susceptible ones tend to have more unstable reactions in different epidemic conditions.
