The number of mosquito-borne viruses (‘moboviruses’) occurring in Europe since the twentieth century now stands at ten; they belong to three families—Togaviridae (Sindbis, Chikungunya), Flaviviridae (West Nile, Usutu, Dengue), and Bunyaviridae (Batai, Ťahyňa, Snowshoe hare, Inkoo, Lednice). Several of them play a definite role in human or animal pathology (Sindbis, Chikungunya, Dengue, West Nile, Ťahyňa). Mobovirus outbreaks are strictly determined by the presence and/or import of particular competent vectors of the disease. Ecological variables affect moboviruses considerably; the main factors are population density of mosquito vectors and their vertebrate hosts, intense summer precipitations or floods, summer temperatures and drought, and presence of appropriate habitats, e.g., wetlands, small water pools, or intravillan sewage systems. A surveillance for moboviruses and the diseases they cause in Europe is recommendable, because the cases may often pass unnoticed or misdiagnosed not only in free-living vertebrates but also in domestic animals and even in humans.

Mosquito-borne viruses in Europe

Modern detection and identification tools can help to provide answers to urgent questions about the incidence, prevalence, and epidemiology of currently emerging diseases. We developed highly sensitive one-step TaqMan reverse transcription-PCR assays with sensitivities ranging from 10 4 to 10 1 molecules for 11 human pathogens of the orthobunyaviruses. We compared the performances of these assays on three currently available cyclers (ABI-PRISM 7700, LightCycler, and SmartCycler). The assay for Oropouche virus (OROV) was tested using sera collected from days 1 to 5 after onset of OROV disease and was found to be greatly superior to an established nested PCR system. A mean copy number of 1.31 10 7 OROV RNA/ml of serum was detected. Diagnostic RNA detection can be used as early as day 1 after onset of OROV disease. The use of a mobile SmartCycler and a hands-on time of less than 3 h could help to intensify outbreak surveillance and control, especially in field studies. The Bunyaviridae family of viruses is subdivided into five genera. Except for the plant pathogenic genus Tospovirus, the Orthobunyavirus, Hantavirus, Nairovirus, and Phlebovirus genera are composed of enzootic viruses, some of which cause zoonotic disease in humans. As is the case with many zoonoses, humans act as accidental dead-end hosts of a zoonotic transmission cycle oscillating between mammals (mostly rodents) and arthropods (Orthobunyavirus, Nairovirus, and Phlebovirus) or among rodents via aerosols and bite wounds (Hantavirus). There are 330 known viruses in these four genera, and 174 are listed as belonging to the genus Orthobunyavirus, including half of the approximately 60 Bunyaviridae viruses causing disease in humans (10, 11).

Rapid detection of human pathogenic orthobunyaviruses.

Three virus strains serologically related to the California encephalitis group (Bunyaviridae) of arboviruses were isolated from 7331 mosquitoes collected in Norway in June-August 1975. Two of the isolates (S 548 and S 618) seemed to be closely related and the third, S 568, more distantly related by serological techniques to Tahyna virus. Viruses were found in the mosquito species Aedes sticticus, A. diantaeus and A. hexodontus colllected (in order) from Oyern (59 degrees N, 11 degrees 12' E), Trandum (60 degrees 08' N, 11 degrees 10' E) and Masi (69 degrees 26' N, 23 degrees 39' E). The Masi isolate seems to be the northernmost arbovirus isolate in the world so far. Strain S 568 was from 16 male A. diantaeus, indicating transovarial transmission of the virus. An accidental infection demonstrated the potential human pathogenicity of one of the newly-isolated strains (S 568), and the ability of CE viruses to persistently infect suckling mouse brains was indicated by in vivo findings. The biological characteristics of the new strains so far investigated are consistent with those of the California encephalitis group. During this work the Aerosil absorption method for production of haemagglutinating antigens proved useful for Tahyna virus and the newly-isolated strains. Trypsinization of erythrocytes improved the haemagglutinating capacities of these viruses.

California encephalitis group viruses isolated from mosquitoes collected in southern and Arctic Norway.

Inkoo virus (INKV) and Chatanga virus (CHATV), which are circulating in Finland, are mosquitoborne California serogroup orthobunyaviruses that have a high seroprevalence among humans. Worldwide, INKV infection has been poorly described, and CHATV infection has been unknown. Using serum samples collected in Finland from 7,961 patients suspected of having viral neurologic disease or Puumala virus infection during the summers of 2001-2013, we analyzed the samples to detect California serogroup infections. IgM seropositivity revealed 17 acute infections, and cross-neutralization tests confirmed presence of INKV or CHATV infections. All children (<16 years of age) with INKV infection were hospitalized; adults were outpatients with mild disease, except for 1 who was hospitalized with CHATV infection. Symptoms included fever, influenza-like illness, nausea or vomiting, disorientation, nuchal rigidity, headache, drowsiness, and seizures. Although many INKV and CHATV infections appear to be subclinical, these viruses can cause more severe disease, especially in children.
