Inflammation has been advocated as a possible common central mechanism for developmental cognitive impairment. Rett syndrome (RTT) is a devastating neurodevelopmental disorder, mainly caused by de novo loss-of-function mutations in the gene encoding MeCP2. Here, we investigated plasma acute phase response (APR) in stage II (i.e., “pseudo-autistic”) RTT patients by routine haematology/clinical chemistry and proteomic 2-DE/MALDI-TOF analyses as a function of four major MECP2 gene mutation types (R306C, T158M, R168X, and large deletions). Elevated erythrocyte sedimentation rate values (median 33.0 mm/h versus 8.0 mm/h, ) were detectable in RTT, whereas C-reactive protein levels were unchanged (). The 2-DE analysis identified significant changes for a total of 17 proteins, the majority of which were categorized as APR proteins, either positive ( spots) or negative ( spots), and to a lesser extent as proteins involved in the immune system ( spots), with some proteins having overlapping functions on metabolism ( spots). The number of protein changes was proportional to the severity of the mutation. Our findings reveal for the first time the presence of a subclinical chronic inflammatory status related to the “pseudo-autistic” phase of RTT, which is related to the severity carried by the MECP2 gene mutation.

La maca (Lepidium meyenii) es una planta que crece sobre los 4000 metros de altitud en los Andes Centrales del Peru, presenta diferentes variedades de acuerdo al color de su hipocotilo. La presente revision resume los resultados de estudios sobre los efectos de la maca en la funcion sexual, la espermatogenesis, la funcion reproductiva femenina, la memoria, la depresion y la ansiedad, como energizante y contra la hiperplasia benigna de prostata, osteoporosis y sindrome metabolico. Se discute tambien su efecto antienvejecimiento y la seguridad en su consumo. Se han demostrado diferencias en el efecto de las variedades negra, amarilla y roja de maca. La maca negra es la que mejores resultados presenta sobre la espermatogenesis, la memoria y contra la fatiga, mientras que la maca roja es la variedad que mejor revierte la hiperplasia benigna de prostata y la osteoporosis inducida experimentalmente. Ademas, la maca reduce los niveles de glucosa, y su consumo se relaciona con la reduccion de la presion arterial y un mejor puntaje de salud. Estudios experimentales han demostrado que el consumo a corto como a largo plazo no muestra toxicidad tanto in vivo como in vitro. A pesar que los estudios experimentales han demostrado que la maca presenta diversos efectos beneficos, son necesarios mas estudios clinicos para confirmar estos resultados

This thesis argues for the need for a more comprehensive discussion of biodiversity use in relation to enhancing benefits of this use for biodiverse countries and promoting more equitable sharing of these benefits. The findings from this doctoral research reveal that biodiversity-based innovation is a social shaping process that has resulted in large benefits. The cumulative capability to use species from biodiversity gives meanings that contribute to the species shaping process, with organisations and institutional changes providing direction and increasing the rate of the shaping process. In showing how innovation takes place and how the appropriation of benefits occurs, this research contributes to studies on science policy and innovation in relation, especially, to biodiversity-based innovation.
The thesis introduces the Convention of Biological Diversity (CBD) and the Nagoya Protocol as representing change to the governance of biodiversity. The theoretical approach draws on evolutionary and institutional economics, both of which inform and extend a question that is central in the sociology of technology: That is how are technology (innovation understood as an output) and social practices shaped collectively? Three cases are used to trace what occurs in the shaping process of species from biodiversity: (i) The Jersey cow is a breed within the species Bos Taurus or modern taurine cattle. The isolated character of Jersey delimited the scope of the breed at a point in time when it was being bred locally and allow us to identify its shaping as a ‘technology’, and the broader diffusion of its use. The Jersey cow is used to introduce the theoretical framework and the analysis. (ii) Maca, originally from Peru, is a root crop with nutritional and, allegedly, fertility enhancing properties. It was domesticated in Peru and only a few world regions have conditions favouring its production. Maca is commercialised as flour and used as a raw material. (iii) Quinua has great potential as a staple food crop. The Food and Agriculture Organization (FAO) declared 2013 to be the International Year of Quinoa on the basis of its unique and nutritious character. Three Andean countries (Bolivia, Ecuador and Peru) report exports of quinua grain, although dozens of countries around the world are engaged in performing agronomic testing for its commercial production. A comparative analysis of the three cases helps to identify the science and technology policy issues related to implementation of the CBD and the Nagoya Protocol. The case studies demonstrate the innovation process of species from biodiversity. Benefits arise from the diffusion of the use of the species (via commercialisation), which accrued to individuals or groups. The characterisation of the innovation process highlights how the voices and agency of actors and organisations affected the shaping process. The governance over the goods that emerged from the use of the species defined the appropriation of benefits.

Benefitting from biodiversity-based innovation

Tyrosinase inhibitors are of far-ranging importance in cosmetics, medicinal products, and food industries. Peru is a diverse country with a wide variety of plants that may contain excellent anti-tyrosinase inhibitors. In the present study, the tyrosinase inhibitory properties of 50 medicinal plant extracts from Peru were investigated using tyrosinase assay. Among plant extracts, those that showed an inhibition rate >50% were Hypericum laricifolium Juss., Taraxacum officinaleF.H.Wigg., and Muehlenbeckia vulcanicaMeisn., with H. laricifolium Juss. showing the greatest anti-tyrosinase activity. Although H. laricifolium Juss. has been widely used as a medicinal plant by Peruvians, little is known regarding its bioactive components and effects on tyrosinase activity. For this reason, we attempted to discover tyrosinase inhibitors in H. laricifolium Juss. for the first time. The bioactive components were separated by Sephadex LH-20 chromatography and eluted with 100% methanol. Eight compounds were discovered and characterized by high-performance liquid chromatography coupled with diode array detection (HPLC-DAD): protocatechuic acid, p-hydroxybenzoic acid, chlorogenic acid, vanilic acid, caffeic acid, kaempferol 3-O-glucuronide, quercetin, and kaempferol. In addition, the concentration of these compounds required for 50% inhibition (IC50) of tyrosinase activity were evaluated. Quercetin exhibited the strongest tyrosinase inhibition (IC50 14.29 ± 0.3 μM). Therefore, the Peruvian plant H. laricifolium Juss. could be a novel source for anti-tyrosinase activity.

Screening of Peruvian Medicinal Plants for Tyrosinase Inhibitory Properties: Identification of Tyrosinase Inhibitors in Hypericum laricifolium Juss.

The present study attempts to investigate the cytotoxic activity of ethanol and ethyl acetate extracts of the Moroccan Berberis vulgaris and its major component berberine, together with exploring their antioxidant properties. It also consists of studying the combination effect of berberine and S-nitroso-N-acetylpenicillamine (SNAP), a nitric oxide (NO) donor, against the human breast adenocarcinoma cell line (MCF-7). Using the MTT assay, we report a differential cytotoxic effect of ethanol and ethyl acetate extracts since the ethanol extract is more cytotoxic than the ethyl acetate one, with IC50 = 3.54 μg/mL and 596.71 μg/mL, respectively. Interestingly, no cytotoxic effect was observed against normal cells. Furthermore, these extracts showed a remarkable antioxidant activity as measured by the DPPH free radicals scavenging assay. In fact, the IC50 values are 69.65 μg/mL and 77.75 μg/mL for the ethanol and ethyl acetate extracts, respectively. In addition, several concentrations of berberine, when combined with the NO donor used at IC30, induced a synergistic cytotoxic activity at concentrations ranging from 8.40 μM to 33.60 μM, as revealed by the combination index values, using the Chou–Talalay method. However, at the other concentrations tested, an antagonistic effect was observed. The observed cytotoxicity was related to apoptosis induction as demonstrated by the annexin-V-streptavidin FITC-staining analysis.

Studies on the Dual Cytotoxicity and Antioxidant Properties of Berberis vulgaris Extracts and Its Main Constituent Berberine.

There is a wide diversity of plants and seasonal crops in Peru, due to the presence of many climatic zones. Numerous plants are used to cure or prevent diseases. These plants are promising candidates for functional foods products. The most frequent form in which they are used is an aqueous infusion or decoction. In this study, we compared the antioxidant properties of ten Peruvian plants infusions and investigated their relation to the phenolic content. The studied plants were: Uncaria tomentosa (cat’s claw), Lepidium meyenii (maca), Berberis vulgaris L. (barberry, agracejo), Phyllantus niruri (chanca piedra), Annona muricata L. (graviola, soursop), Gentianella alborosea (hercampure), Geranium dielsianum (pasuchaca), Tabebuia ochracea (tahuari), Notholaena nivea (“cuti cuti”) and Tiquilia paronychioides (“flor de arena”). Infusions of all studied plants have shown antioxidant activity, though there was a large diversity between the results. The antioxidant properties, determined with DPPH and ABTS scavenging assays as well as FRAP test, were strongly correlated with total phenolic content, while there was no correlation with the carotenoid content.
