Marine natural products.

Marine sponges often contain diverse and abundant microbial communities, including bacteria, archaea, microalgae, and fungi. In some cases, these microbial associates comprise as much as 40% of the sponge volume and can contribute significantly to host metabolism (e.g., via photosynthesis or nitrogen fixation). We review in detail the diversity of microbes associated with sponges, including extensive 16S rRNA-based phylogenetic analyses which support the previously suggested existence of a sponge-specific microbiota. These analyses provide a suitable vantage point from which to consider the potential evolutionary and ecological ramifications of these widespread, sponge-specific microorganisms. Subsequently, we examine the ecology of sponge-microbe associations, including the establishment and maintenance of these sometimes intimate partnerships, the varied nature of the interactions (ranging from mutualism to host-pathogen relationships), and the broad-scale patterns of symbiont distribution. The ecological and evolutionary importance of sponge-microbe associations is mirrored by their enormous biotechnological potential: marine sponges are among the animal kingdom's most prolific producers of bioactive metabolites, and in at least some cases, the compounds are of microbial rather than sponge origin. We review the status of this important field, outlining the various approaches (e.g., cultivation, cell separation, and metagenomics) which have been employed to access the chemical wealth of sponge-microbe associations.

Secondary metabolites of fungi from marine habitats.

The marine environment is distinguished by unique groups of organisms being the source of a wide array of fascinating structures. The enormous biodiversity of marine habitats is mirrored by the molecular diversity of secondary metabolites found in marine animals, plants and microbes. The recognition that many marine invertebrates contain endo- and epibiotic microorganisms and that some invertebrate-derived natural products are structurally related to bacterial metabolites suggests a microbial origin for some of these compounds. Other marine natural products, however, are clearly located in invertebrate tissue and microbial involvement in the biosynthetic process seems unlikely. The complexity of associations in marine organisms, especially in sponges, bryozoans and tunicates, makes it extremely difficult to definitively state the biosynthetic source of many marine natural products or to deduce their ecological significance. Whereas many symbiotic marine microorganisms cannot be isolated and cultured, numerous epi- and endobiotic marine fungi produce novel secondary metabolites in laboratory cultures. The potent biological activity of many marine natural products is of relevance for their ecological function but is also the basis of their biomedical importance.

A marine fungal isolate, identified as Wardomyces anomalus, was cultivated and found to produce two new xanthone derivatives, 2,3,6,8-tetrahydroxy-1-methylxanthone (1) and 2,3,4,6,8-pentahydroxy-1-methylxanthone (2), in addition to the known xanthone derivative 3,6,8-trihydroxy-1-methylxanthone (3) and the known fungal metabolite 5-(hydroxymethyl)-2-furanocarboxylic acid (4). The structures of all compounds were determined on the basis of extensive spectroscopic measurements (1D and 2D NMR, MS, UV, and IR). Compounds 1 and 4 showed significant antioxidant activities. The total extract and 1, 3, and 4 were shown to be inhibitors of p56lck tyrosine kinase.

Two new xanthone derivatives from the algicolous marine fungus Wardomyces anomalus

A marine fungal isolate, identified as Acremonium sp., was mass cultivated and found to produce two novel hydroquinone derivatives, 7-isopropenylbicyclo[4.2.0]octa-1,3,5-triene-2,5-diol (1) and 7-isopropenylbicyclo[4.2.0]octa-1,3,5-triene-2,5-diol-5-beta-D-glucopyranoside (2). Compound 1 and its glucoside 2 possess a most unusual ring system. The new natural products (3R,4S)-3,4-dihydroxy-7-methyl-3,4-dihydro-1(2H)naphthalenone (3) and (3S,4S)-3,4-dihydroxy-7-methyl-3,4-dihydro-1(2H)-naphthalenone (4) were obtained as a 1:0.8 mixture. 2-(1-Methylethylidene)pentanedioic acid (5) was isolated for the first time as a natural product and its structure proven by X-ray analysis. In addition to these compounds an inseparable mixture of three new isomeric compounds, pentanedioic acid 2-(1-methylethylidene)-5-methyl ester (6), pentanedioic acid 2-(1-methylethylidene)-1-methyl ester (7), and pentanedioic acid 2-(1-methylethenyl)-5-methyl ester (8), was also obtained. Isolated together with the new compounds were three known hydroquinone derivatives, 9, 10, and 11. The structures of all compounds were determined by interpretation of their spectroscopic data (1D and 2D NMR, MS, UV, and IR). Each isolate was tested for its antioxidant properties, and compounds 1 and 9-11 were found to have significant activity.

New antioxidant hydroquinone derivatives from the algicolous marine fungus Acremonium sp.

The fungus Epicoccum sp., was isolated from the marine brown alga Fucus vesiculosus. After cultivation the fungus was investigated for its secondary metabolite content, and found to contain the new natural product 4,5,6-trihydroxy-7-methylphthalide (1, epicoccone), together with 5-(acetoxymethyl)-furan-2-carboxylic acid (2), furan-2-carboxylic acid (3), 5-(hydroxymethyl)-furan-2-carboxylic acid (4), (-)-(3 R,4 S)-4-hydroxymellein (5), and (-)-(3 R)-5-hydroxymellein (6). The structures of all compounds were determined by interpretation of their spectroscopic data (1D and 2D NMR, MS, UV, optical rotation and IR). Each isolate was tested for its antioxidative properties. Compound 1 was found to be potently active, showing 95 % DPPH radical scavenging effects at 25 microg/mL. Compound 1 also inhibited the peroxidation of linolenic acid in the TBARS assay (62 % inhibition at 37 microg/mL).
