Surface plasmon resonance: towards an understanding of the mechanisms of biological molecular recognition.

Interest in the development of surface plasmon resonance (SPR) based immunosensors for detection and monitoring of low-molecular-weight analytes of biomedical, food and environmental fields has been rapidly increasing over the last 10 years. By combining the advantages of the specific antigen–antibody immunoreaction and the high sensitivity and reliability of SPR signal transduction, SPR immunoassays offer exceptional performance capabilities with respect to sensitivity, specificity, speed and multianalyte detection in complex analytical matrices. Advancements in the technology of antibody production and the signal transduction provide a promising scope for SPR immunosensors to lead in the next generation biosensors. This review highlights the current state-of-the-art in SPR immunosensors and outlines briefly the important issues with regard to the development of SPR immmunosensors, such as preparation of the biomolecules, sensor fabrication, non-specific adsorption, surface regeneration and detection principles. Particular emphasis is given to the indirect competitive immunoassay principle which is compatible and highly promising for detection of small analytes with enhanced sensitivity. In addition, recent advancements and trends in the application of SPR immunosensors in biomedical, environmental and food-related analyses are discussed.

Recent advancements in surface plasmon resonance immunosensors for detection of small molecules of biomedical, food and environmental interest

In the year 2003 there was a 17% increase in the number of publications citing work performed using optical biosensor technology compared with the previous year. We collated the 962 total papers for 2003, identified the geographical regions where the work was performed, highlighted the instrument types on which it was carried out, and segregated the papers by biological system. In this overview, we spotlight 13 papers that should be on everyone's 'must read' list for 2003 and provide examples of how to identify and interpret high-quality biosensor data. Although we still find that the literature is replete with poorly performed experiments, over-interpreted results and a general lack of understanding of data analysis, we are optimistic that these shortcomings will be addressed as biosensor technology continues to mature.

Survey of the year 2003 commercial optical biosensor literature

During last decade there has been significant progress in the development of analytical techniques for evaluation of receptor-ligand interaction. Surface plasmon resonance (SPR)-based optical biosensors are now being used extensively to define the kinetics of wide variety of macromolecular interactions and high- and low-affinity small molecule interactions. The experimental design data analysis methods are evolving along with widespread applications in ligand fishing, microbiology, virology, host-pathogen interaction, epitope mapping, and protein-, cell-, membrane-, nucleic acid-protein interactions. SPR-based biosensors have strong impact on basic and applied research significantly. This brief review describes the SPR technology and few of its applications in relation to receptor-ligand interaction that has brought significant change in the methodology, analysis, interpretation, and application of the SPR technology.

Integrated optics is becoming one of the most widely researched areas in optical physics. In this paper we describe a simple technique for fabrication of integrated optic units which contain planar waveguides and embossed surface relief grating couplers which may be used as optical chemical sensor platforms. The entire IO unit is fabricated via the sol-gel process. Fabrication of sol gel-based planar waveguide/grating coupler platforms for use as optical chemical sensors

A novel, generic configuration for absorption-based optical sensors is presented. This technique utilizes embossed sol- gel glass derived gratings to couple LED light into and out of a planar waveguide. The power of the guided light is monitored as its evanescent field interrogates a dye-doped sol-gel sensor layer. This waveguiding arrangement is easy to manufacture and shows great potential for use in disposable, mass-produced sensor chips for a range of absorption-based sensing tasks.
