Following Sharpless' visionary characterization of several idealized reactions as click reactions, the materials science and synthetic chemistry communities have pursued numerous routes toward the identification and implementation of these click reactions. Herein, we review the radical-mediated thiol-ene reaction as one such click reaction. This reaction has all the desirable features of a click reaction, being highly efficient, simple to execute with no side products and proceeding rapidly to high yield. Further, the thiol-ene reaction is most frequently photoinitiated, particularly for photopolymerizations resulting in highly uniform polymer networks, promoting unique capabilities related to spatial and temporal control of the click reaction. The reaction mechanism and its implementation in various synthetic methodologies, biofunctionalization, surface and polymer modification, and polymerization are all reviewed.

Thiol-click chemistry: a multifaceted toolbox for small molecule and polymer synthesis

The photopolymerization of mixtures of multifunctional thiols and enes is an efficient method for the rapid production of films and thermoset plastics with unprecedented physical and mechanical properties. One of the major obstacles in traditional free-radical photopolymerization is essentially eliminated in thiol–ene polymerizations because the polymerization occurs in air almost as rapidly as in an inert atmosphere. Virtually any type of ene will participate in a free-radical polymerization process with a multifunctional thiol. Hence, it is possible to tailor materials with virtually any combination of properties required for a particular application. © 2004 Wiley Periodicals, Inc. Thiol–enes: Chemistry of the past with promise for the future

Polyethylene glycol (PEG) hydrogels are widely used in a variety of biomedical applications, including matrices for controlled release of biomolecules and scaffolds for regenerative medicine. The design, fabrication, and characterization of PEG hydrogels rely on the understanding of fundamental gelation kinetics as well as the purpose of the application. This review article will focus on different polymerization mechanisms of PEG-based hydrogels and the importance of these biocompatible hydrogels in regenerative medicine applications. Furthermore, the design criteria that are important in maintaining the availability and stability of the biomolecules as well as the mechanisms for loading of biomolecules within PEG hydrogels will also be discussed. Finally, we overview and provide a perspective on some of the emerging novel design and applications of PEG hydrogel systems, including the spatiotemporal-controlled delivery of biomolecules, hybrid hydrogels, and PEG hydrogels designed for controlled stem cell differentiation.

PEG hydrogels for the controlled release of biomolecules in regenerative medicine.

This review, with over 600 references, summarizes the recent applications of photoredox catalysis for organic transformation and polymer synthesis. Photoredox catalysts are metallo- or organo-compounds capable of absorbing visible light, resulting in an excited state species. This excited state species can donate or accept an electron from other substrates to mediate redox reactions at ambient temperature with high atom efficiency. These catalysts have been successfully implemented for the discovery of novel organic reactions and synthesis of added-value chemicals with an excellent control of selectivity and stereo-regularity. More recently, such catalysts have been implemented by polymer chemists to post-modify polymers in high yields, as well as to effectively catalyze reversible deactivation radical polymerizations and living polymerizations. These catalysts create new approaches for advanced organic transformation and polymer synthesis. The objective of this review is to give an overview of this emerging field to organic and polymer chemists as well as materials scientists.

Photocatalysis in organic and polymer synthesis

A thiol monomer is shown to copolymerize with vinyl ether, allyl, acrylate, methacrylate, and vinylbenzene monomers. These thiol−ene polymerizations are photoinitiated without the use of photoinitiator molecules. It is seen that the polymerization proceeds more readily when initiatorless samples are irradiated with light centered around 254 nm as compared to 365 nm light. To demonstrate resistance to oxygen inhibition, thin polymer films of 3−15 μm are polymerized while exposed to ambient air. Without photoinitiator molecules present, light is attenuated only by the monomer and polymer. This feature leads to greater penetration of ultraviolet light and allows for the polymerization of extremely thick polymers. Thick cures of up to 25 in. are obtained using a thiol−vinyl ether system.
