Objective This study intends to explore the potential of galangal extract as a co-chemotherapeutic agent through the analysis of its cytotoxic and migratory effects on metastatic breast cancer cells and as an anti-ageing agent through its senescence inhibitory effect on normal fibroblast cells. Methods Galangal ethanolic extract (GE) was subjected to a cytotoxicity test with the 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay alone or in combination with doxorubicin (Dox) against 4T1 cells but not in NIH-3T3 cells. Evidence of senescent cells was detected using a SA-β galactosidase based assay. In addition, the level of reactive oxygen species (ROS), apoptosis, and cell cycle were measured with a flow cytometry-based assay. Meanwhile, cell migration and matrix metalloproteinase (MMP)-9 expression after GE treatment on 4T1 cells were measured using the scratch wound healing assay and gelatin zymography assay, respectively. The metabolomic profiles of GE were traced using gas chromatography-mass spectrometry (GC-MS) analysis. Results GE effectively inhibited the growth of 4T1 cells with an IC50 value of 135 µg/mL and increased the cytotoxic effect of Dox at concentrations of 50 and 100 µg/mL. GE increased the number of senescent cells arrested in the G2/M phase but did not cause apoptosis. This effect is compounded by increasing intracellular levels of ROS. However, GE reduced senescence to normal in fibroblast cells (NIH 3T3 cells) under oxidative stress by Dox without any changes in the ROS level. Moreover, GE also inhibited the migration of 4T1 cells and suppressed the expression of MMP-9 induced by Dox. Conclusion Galangal has the potential for use as a co-chemotherapeutic agent by inducing senescence in correlation with increasing intracellular ROS toward metastatic breast cancer. However, the effect of GE in decreasing the senescence phenomena toward normal fibroblast cells illustrates its potential as a promising anti-ageing agent. .

Revealing the Reversal Effect of Galangal (Alpinia galanga L.) Extract Against Oxidative Stress in Metastatic Breast Cancer Cells and Normal Fibroblast Cells Intended as a Co- Chemotherapeutic and Anti-Ageing Agent

The use of plant products as immunomodulator has a long history. Forms eternal era, plant, mineral and animal products are used as drugs for the treatment of various diseases. The present-day synthetic compounds find their leads in natural products. The process of immunomodulation retunes the immune system of an individual by restoring routine functions. Research on immunomodulators from natural sources has been extensively studied for modulation of immune system along with protection and prevention of diseases. Rutin, a flavonoid has been investigated for its potential immunomodulatory effects. Increased particle clearance, decreased inflammation, increased leucocyte and antibody production was observed with administration of rutin in rats. The present study is focused on exploring in silico interaction of rutin with some chemokines and inflammatory targets. In this study, rutin was docked with TNF-α, IL-1β, IL-6, and NOs. Docking studies revealed the excellent interaction of rutin with these targets. The result of present study provides insight for the discovery of novel molecules for immunomodulation and treatment of inflammatory disorders. Findings from the present study show that rutin may interact with several chemokines and inflammatory mediators. Further studies on rutin and associated flavonoids are necessary to develop and establish QSAR and QSPR studies which may serve a stepping stone for the development of novel and safe immunomodulator. Rutin therefore, can be considered for development of an immunomodulatory agent.

Phytochemical and Pharmacological Support for the Traditional Uses of Zingiberacea Species in Suriname - A Review of the Literature

Notwithstanding scientific advances, a significant number of the treatments in male infertility remained stayed vague. This study was aimed to study the impact of AlpiniaOfficinarum on sex hormones, serum antioxidant and biochemical markers in rats. Forty adult male rats, of (220 ± 10g) were partitioned into five groups (each consists of eight rats). The primary group was negative control group (-ve) and fed on basal diet only. The other four groups were subcutaneously injected with a single dose of lead acetate (200 mg/kg b.w) to reduce fertility, then were divided into 4 subgroups: including control positive group, and 2nd, 3rd and 4th subgroup were fed on basal diet with supplementation of dried A. officinarum at (5, 7.5 and 10%) respectively for two month. The results revealed that, supplementation with A. officinarum caused a significant positive effectson testes which due to a significant increase in the levels of serum total testosterones, Follicular stimulating Hormone (FSH), Luteinizing Hormone (LH) and Superoxide Dismutase (SOD) levels while Malondialdehyde (MDA) level was diminished. In addition, liver functions and serum lipid profiles were significantly improved compared to the positive control group. In conclusion: Our findings provide a scientific evidence to substantiate A. officinarum in improving fertility in human which may be due to its potent antioxidant properties and androgenic activities.

Effect of (Alpinia officinarum) Hance on Sex Hormones and Certain Biochemical Parameters of Adult Male Experimental Rats

Alpinia galanga is a well-known medicinal plant in Southeast Asia and has been used for a long time as food and medicine. A large number of flavonoid phytochemical compounds have been identified in various parts of this medicinal herb. Flavonoids are commonly known as attractive compounds that can be applied to cosmetic or cosmeceutical product development because of their antioxidant, anti-aging and many other potential biological activities. This recent review aims to illustrate and update the taxonomic status as well as the species description that will be helpful for a rigorous identification and authenticate the raw material or living specimen from A. galanga. The flavonoid phytochemical compounds and the bioactivity of this medicinal plant are also provided. The future perspectives and research directions of A. galanga and its flavonoids are pointed out in this study as well.

GC-MS Analysis and Screening of Anti-Proliferative Potential of Methanolic Extract of Garcinia cowa on Different Cancer Cell Lines

Azadirachta indica , popularly known as ‘ Neem ’, is a very important plant in the Ayurveda system of medicine. It is known toprevent about 40 types of diseases in home-practice. A previous report showed that the methanolic extract of this plant can effectively control the proliferation of leukemia cells. Therefore, this research focuses on searching for a new molecule with potent anti-leukemic property. Ethanolic extract was prepared from the dried leaves. Four bio-molecules were isolated viz. rutin, isoquercetin, quercetrin and β-sitosterol-3-O-β-D-glucoside from the ethanolic extract by repeated column chromatography and HPLC. Quercetrin structures of the isolated molecules were confirmed by Mass, 1HNMR, 13C NMR spectra analysis. MTT assay revealed that β-sitosterol-3-O-β-D-glucosideeffectively reduced the proliferation of MOLT 4 leukemic cells in a dose and time dependent manner. DAPI staining with confocal microscopy indicated that β-sitosterol-3-O-β-D-glucoside efficiently induced nuclear DNA fragmentation in MOLT 4 cells. Finally, flow cytometry after PI staining showed that the compound has potential to check the cell cycle progression at sub G1 phase. In summary, we can conclude that β-sitosterol-3-O-β-D-glucoside has potential to be good therapeutic drug in leukemia treatment in future.

Garcinia cowa of the Clusiaceae family, native to South‐East Asia used in traditional medicine. It has antipyretic, antimicrobial, and many other biological activities. In this review, a thorough study of this plant's chemical constituents and pharmacological and therapeutic effects was conducted from the research articles from PubMed, Science Direct, Google Scholar, and Scopus from 1977 to 2022. Reported secondary metabolites are enriched with xanthones, phloroglucinols, depsidones, steroids, etc. α‐mangostin, β‐mangostin, cowaxanthone, rubraxanthone, cowanin, norcowanin, etc. represent the major xanthones. This article discusses the relationship between the different functional groups in xanthone compounds and their bioactivity against cancer, diabetes, bacteria, leishmania, malaria, and inflammation. This review is a comprehensive compendium of major bioactive molecules and its implication for human disease.
