The structural and mechanical properties of gels formed from biopolymers are discussed both in terms of the techniques used to characterise these systems, and in terms of the systems themselves The techniques included are spectroscopic, chiroptical and scattering methods, optical and electron microscopy, thermodynamic and kinetic methods and rheological characterisation The systems considered are presented in order of increasing complexity of secondary, tertiary and quaternary structure, starting with gels which arise from essentially ‘disordered’ biopolymers via formation of ‘quasicrystalline’ junction zones (eg gelatin, carrageenans, agarose, alginates etc), and extending to networks derived from globular and rod-like species (fibrin, globular proteins, caseins, myosin) by a variety of crosslinking mechanisms Throughout the text, efforts are made to pursue the link (both from experiment and from theory) between the structural methods and mechanical measurements As far as we are aware this is the first major Review of this area since that of J D Ferry in 1948 — The interest shown by polymer physicists in more complex biochemical systems, and the multi-disciplinary approaches now being applied in this area, make the format adopted here, in our opinion, the most logical and appropriate

Structural and mechanical properties of biopolymer gels

Degradation of plant cell wall polysaccharides is of major importance in the food and feed, beverage, textile, and paper and pulp industries, as well as in several other industrial production processes. Enzymatic degradation of these polymers has received attention for many years and is becoming a more and more attractive alternative to chemical and mechanical processes. Over the past 15 years, much progress has been made in elucidating the structural characteristics of these polysaccharides and in characterizing the enzymes involved in their degradation and the genes of biotechnologically relevant microorganisms encoding these enzymes. The members of the fungal genus Aspergillus are commonly used for the production of polysaccharide-degrading enzymes. This genus produces a wide spectrum of cell wall-degrading enzymes, allowing not only complete degradation of the polysaccharides but also tailored modifications by using specific enzymes purified from these fungi. This review summarizes our current knowledge of the cell wall polysaccharide-degrading enzymes from aspergilli and the genes by which they are encoded.

Aspergillus Enzymes Involved in Degradation of Plant Cell Wall Polysaccharides

Hemicellulose is a complex group of heterogeneous polymers and represents one of the major sources of renewable organic matter. Mannan is one of the major constituent groups of hemicellulose in the wall of higher plants. It comprises linear or branched polymers derived from sugars such as d-mannose, d-galactose, and d-glucose. The principal component of softwood hemicellulose is glucomannan. Structural studies revealed that the galactosyl side chain hydrogen interacts to the mannan backbone intramolecularly and provides structural stability. Differences in the distribution of d-galactosyl units along the mannan structure are found in galactomannans from different sources. Acetyl groups were identified and distributed irregularly in glucomannan. Some of the mannosyl units of galactoglucomannan are partially substituted by O-acetyl groups. Some unusual structures are found in the mannan family from seaweed, showing a complex system of sulfated structure. Endohydrolases and exohydrolases are involved in the breakdown of the mannan backbone to oligosaccharides or fermentable sugars. The main-chain mannan-degrading enzymes include β-mannanase, β-glucosidase, and β-mannosidase. Additional enzymes such as acetyl mannan esterase and α-galactosidase are required to remove side-chain substituents that are attached at various points on mannan, creating more sites for subsequent enzymatic hydrolysis. Mannan-degrading enzymes have found applications in the pharmaceutical, food, feed, and pulp and paper industries. This review reports the structure of mannans and some biochemical properties and applications of mannan-degrading enzymes.

Publisher Summary This chapter discusses the interaction of galactomannans with other polysaccharides. The general chemistry of the galactomannans has been reviewed by Whistler and Smart and others. The chapter discusses the uses of guar and locust-bean gums in various industrial applications and investigates the structural chemistry of galactomannans, especially as revealed by enzymic studies. Mixtures of locust-bean gum with the non-gelling polysaccharide from Xanthomonas campestris have been shown to interact synergistically to give firm, rubbery gels, whereas the use of the galactomannan from C yamopsis tetragonolobus (guar gum) results only in viscosity enhancement. These interactions are important in many of the industrial applications of galactomannans, and a study of them may also help provide an understanding of the associations among polysaccharide chains, contributing to biological cohesion and texture. The two main groups of galactomannan polysaccharides are those derived from (1) the endosperm of plant seeds, the vast majority of which originate in the Leguminoseae, and (2) microbial sources, in particular, the yeasts and other fungi. D-Mannose and D galactose are also found in numerous other plant polysaccharides—for example, glucomannansm, mannans, and galactans.
