Tomato (Solanum lycopersicum) is an important dietary source of bioactive phytochemicals and active breeding programs constantly produce new cultivars possessing superior and desirable traits. The phytopathogenic Ralstonia solanacearum, the causal agent of bacterial wilt, is a highly destructive bacterial disease with a high economic impact on tomato production. This study followed an untargeted metabolomic approach involving four tomato cultivars and aimed at the identification of secondary metabolites involved in plant defense after infection with R. solanacearum. Liquid chromatography coupled to mass spectrometry (LC-MS) in combination with multivariate data analysis and chemometric modelling were utilized for the identification of discriminant secondary metabolites. The total of 81 statistically selected features were annotated belonging to the metabolite classes of amino acids, organic acids, fatty acids, various derivatives of cinnamic acid and benzoic acids, flavonoids and steroidal glycoalkaloids. The results indicate that the phenylpropanoid pathway, represented by flavonoids and hydroxycinnamic acids, is of prime importance in the tomato defense response. The hydroxycinnamic acids esters of quinic acid, hexoses and glucaric acids were identified as signatory biomarkers, as well as the hydroxycinnamic acid amides to polyamines and tyramine. Interestingly, the rapid and differential accumulation of putrescine, dopamine, and tyramine derivatives, along with the presence of a newly documented metabolite, feruloyl serotonin, were documented in the infected plants. Metabolite concentration variability in the different cultivar tissues point to cultivar-specific variation in the speed and manner of resource redistribution between the host tissues. These metabolic phenotypes provide insights into the differential metabolic signatures underlying the defense metabolism of the four cultivars, defining their defensive capabilities to R. solanacearum.

Metabolomic Profiling of the Host Response of Tomato (Solanum lycopersicum) Following Infection by Ralstonia solanacearum.

Severe Acute Respiratory Syndrome Corona Virus 2 (SARS-CoV-2) being a causative agent for global pandemic disease nCOVID'19, has acquired much scientific attention for the development of effective vaccines and drugs. Several attempts have been made to explore repurposing existing drugs known for their anti-viral activities, and test the traditional herbal medicines known for their health benefiting and immune-boosting activity against SARS-CoV-2. In this study, efforts were made to examine the potential of 605 phytochemicals from 37 plant species (of which 14 plants were endemic to India) and 139 antiviral molecules (Pubchem and Drug bank) in inhibiting SARS-CoV-2 multiple protein targets through a virtual screening approach. Results of our experiments revealed that SARS-CoV-2 MPro shared significant disimilarities against SARS-CoV MPro and MERS-CoV MPro indicating the need for discovering novel drugs. This study has screened the phytochemical cyanin (Zingiber officinale) which may exhibit broad-spectrum inhibitory activity against main proteases of SARS-CoV-2, SARS-CoV and MERS-CoV with binding energies of (-) 8.3 kcal/mol (-) 8.2 kcal/mol and (-) 7.7 kcal/mol respectively. Amentoflavone, agathisflavone, catechin-7-o-gallate and chlorogenin were shown to exhibit multi-target inhibitory activity. Further, Mangifera indica, Anacardium occidentale, Vitex negundo, Solanum nigrum, Pedalium murex, Terminalia chebula, Azadirachta indica, Cissus quadrangularis, Clerodendrum serratum and Ocimum basilicumaree reported as potential sources of phytochemicals for combating nCOVID'19. More interestingly, this study has highlighted the anti-viral properties of the traditional herbal formulation "Kabasura kudineer" recommended by AYUSH, a unit of Government of India. Short listed phytochemicals could be used as leads for future drug design and development. Genomic analysis of identified herbal plants will help in unraveling molecular complexity of therapeutic and anti-viral properties which proffer lot of chance in the pharmaceutical field for researchers to scout new drugs in drug discovery.

Exploring Phytochemicals of Traditional Medicinal Plants Exhibiting Inhibitory Activity Against Main Protease, Spike Glycoprotein, RNA-dependent RNA Polymerase and Non-Structural Proteins of SARS-CoV-2 Through Virtual Screening

Lycium shawii Roem. & Schult.: A new bioactive antimicrobial and antioxidant agent to combat multi-drug/pan-drug resistant pathogens of wound burn infections.

Medicinal plants and plant remedies have been in use in Ethiopia for centuries. Studies on ethnobotany, ethnomedicine, and ethnoveterinary estimate that nearly 80% of Ethiopians use some type of medicinal plants and plant remedies. Medicinal plants are regarded as the most important and sometimes the only source of therapeutics in the country. Some 800 plant species are used as sources of medicine to treat about 300 physical and mental disorders. However, because these plant species are not adequately studied, there is a big limitation in their documentation, profiling, and management. Moreover, there is a continuous loss of knowledge about medicinal plants because the communities and people are adopting new lifestyles. Hence, this article reports the finding of a study aimed at providing the gross phytochemical characteristics and antimicrobial activities of ethanol and aqueous extracts of fruit, leaf, and stem of Solanum incanum L. against two Gram-negative (Escherichia coli and Salmonella typhi) and two Gram-positive (Bacillus subtilis and Staphylococcus aureus) bacteria for developing gross antimicrobial profile of the plant. Phytochemical screening of fruit, leaf, and stem extracts of S. incanum has shown that it is the source of alkaloids, saponins, flavonoids, glycosides, terpenoids, and steroids. According to agar disc-diffusion tests, 100 mg/mL extracts of the plant produced bacterial growth inhibition zones of 0.00 to 16.06 mm. Ethanol and aqueous leaf extracts produced inhibition zones ranging from 11.34 to 16.06 mm against all bacterial species. The greatest inhibition zone of 16.06 mm was recorded in E. coli subjected to ethanol leaf extract. The same extract resulted in a growth inhibition zone of 16.04 mm in S. aureus. The greatest growth inhibition zones in B. subtilis (13.34 mm) and S. typhi (11.56 mm) were observed with ethanol leaf and fruit extracts, respectively. Aqueous leaf extracts produced growth inhibition zones ranging from 10.45 mm (for S. typhi) to 14.02 mm (for E. coli). Ethanol leaf extracts resulted in the lowest Minimum Inhibition Concentration (MIC) of 1.56 mg/mL in E. coli and S. aureus. Therefore, fruits, leaves, and stems of S. incanum can be regarded as good sources of some bioactive compounds. The findings are important for taking measures for conservation and sustainable use of the plant as well as for further elucidation of its phytochemistry and antimicrobial efficacy of its constituents.

Preliminary Antimicrobial Profile of Solanum incanum L.: A Common Medicinal Plant.

Cyclocarya paliurus (Batal.) Iljinskaja is an indigenous and multifunction tree species in China, but it is mainly used in pharmaceutical and nutraceutical ingredients. To make a comprehensive evaluation on its bioactive metabolites, antioxidant and antitumor potentials of C. paliurus leaves, the leaf samples were collected from 15 geographic locations (natural populations) throughout its distribution areas. High-performance liquid chromatography (HPLC) and colorimetric methods were used to detect the contents of bioactive metabolites. The antioxidant activity was evaluated by 2,2′-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and reducing power assays. The antiproliferative activity on different cancer cell types was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Contents of bioactive metabolites, and antioxidant and antiproliferative activities in the extracts were significantly affected by solvent and population. In most cases, the contents of flavonoids and triterpenoids, and the antioxidant and antiproliferative activities in the ethanol extracts were higher than the water extracts. The best scavenging capacity of DPPH (IC50 = 0.34 mg/mL) and ABTS (IC50 = 0.50 mg/mL) radical occurred in the ethanol extracts of S15 and S7 population respectively, while the strongest reducing power (EC50 = 0.71 mg/mL) was achieved in the ethanol extracts of S14 population. The antiproliferation effects of C. paliurus extracts on cancer cells varied with different cell types. The HeLa cell was the most sensitive to C. paliurus extracts, and their IC50 values of the ethanol extracts varied from 0.13 to 0.42 mg/mL among C. paliurus populations. Redundancy analysis showed that total polyphenol had the greatest contribution to the antioxidant activity, but total flavonoid was mostly responsible for the antiproliferation effects. These results would provide important scientific evidences not only for developing C. paliurus as a potent antioxidant and antitumor reagent, but also for obtaining the higher yield of bioactive compounds in the C. paliurus plantation.
