Phosphatidylinositol 4,5-Bisphosphate Functions as a Second Messenger that Regulates Cytoskeleton–Plasma Membrane Adhesion

Nuclear pore complexes have been isolated in association with a 150 A thick lamina by detergent and salt fractionation of nuclear envelopes from rat liver. The pore complexes exhibit characteristic morphology and appear to be attached in a highly specific orientation to the lamina, which extends over relatively large areas. The pore complex-lamina fraction is composed of three major and several minor polypeptides with little or no DNA, RNA, or phospholipid. It is suggested that the association of the pore complexes and the lamina reflects the structural arrangement of the nuclear periphery in vivo.

Membrane flow and interconversions among endomembranes

The ultrastructure of the apical zone of lactating rat mammary epithelial cells was studied with emphasis on vesicle coat structures. Typical 40-60 nm ID "coated vesicles" were abundant, frequently associated with the internal filamentous plasma membrane coat or in direct continuity with secretory vesicles (SV) or plasma membrane proper. Bristle coats partially or totally covered membranes of secretory vesicles identified by their casein micelle content. This coat survived SV isolation. Exocytotic fusion of SV membranes and release of the casein micelles was observed. Frequently, regularly arranged bristle coat structures were identified in those regions of the plasma membrane that were involved in exocytotic processes. Both coated and uncoated surfaces of the casein-containing vesicles, as well as typical "coated vesicles", were frequently associated with microtubules and/or microfilaments. We suggest that coat materials of vesicles are related or identical to components of the internal coat of the surface membrane and that new plasma membrane and associated internal coat is produced concomitantly by fusion and integration of bristle coat moieties. Postexocytotic association of secreted casein micelles with the cell surface, mediated by finely filamentous extensions, provided a marker for the integrated vesicle membrane. An arrangement of SV with the inner surface of the plasma membrane is described which is characterized by regularly spaced, heabily stained membrane to membrane cross-bridges (pre-exocytotic attachment plaques). Such membrane-interconnecting elements may represent a form of coat structure important to recognition and interaction of membrane surfaces.

Involvement of vesicle coat material in casein secretion and surface regeneration.

Plasma membranes were isolated from rat and mouse livers, one rat hepatoma (and its subline) and two mouse hepatomas, and their lipid class compositions were determined. Lipids accounted for 30 to 35% of the dry weight of the membranes of livers and mouse hepatomas, and for 45% in the case of rat hepatoma-subline. Of the total lipids of rat-liver plasma membranes, 60% consisted of phospholipids, the corresponding values for mouse-liver and rat-hepatoma plasma membranes amounting to 55% and for both mouse-hepatoma plasma membranes to about 50%. The free cholesterol and cholesteryl ester contents of all hepatoma plasma membranes were significantly increased as compared with normal. Evidence is presented that the increase of free cholesterol was not a preparative artefact. The major phospholipid classes in all plasma membranes were phosphatidyl choline, sphingomyelin, phosphatidyl ethanolamine and phosphatidyl serine. The relative proportions in each plasma membrane species could differ appreciably, the mouse- and rat-liver membranes showing the closest resemblance. Possible reasons for (a) the higher level of phosphatidyl serine as compared with published values, and (b) the wide divergencies which may be found among the phospholipid profiles of rat-liver plasma membranes reported by other authors, are presented. Cardiolipin was absent from liver plasma membranes, but some could be found in the hepatoma membranes due to mitochondrial contamination. No consistent phospholipid profile characterized hepatoma as distinct from liver plasma membranes, nor did the hepatoma data-including plasmalogens-resemble the few available data on other hepatomas.

Studies on plasma membranes : XVIII. lipid class composition of plasma membranes isolated from rat and mouse liver and hepatomas.

Nuclear membranes were isolated from rat and pig liver by sonication of highly purified nuclear fractions and subsequent removal of adhering nucleoproteins in a high salt medium. The fractions were examined in the electron microscope by both negative staining and thin sectioning techniques and were found to consist of nuclear envelope fragments of widely varying sizes. Nuclear pore complex constituents still could frequently be recognized. The chemical composition of the nuclear membrane fractions was determined and compared with those of microsomal fractions prepared in parallel. For total nuclei as well as for nuclear membranes and microsomes, various enzyme activities were studied. The results indicate that a similarity exists between both fractions of cytomembranes, nuclear envelope, and endoplasmic reticulum, with respect to their RNA:protein ratio and their content of polar and nonpolar lipids. Both membranous fractions had many proteins in common including some membrane-bound enzymes. Activities in Mg-ATPase and the two examined cytochrome reductases were of the same order of magnitude. The content of cytochrome b5 as well as of P-450 was markedly lower in the nuclear membranes. The nuclear membranes were found to have a higher buoyant density and to be richer in protein. The glucose-6-phosphatase and Na-K-ATPase activities in the nuclear membrane fraction were very low. In the gel electrophoresis, in addition to many common protein bands, some characteristic ones for either microsomal or nuclear membranous material were detected. Significant small amounts of DNA and RNA were found to remain closely associated with the nuclear envelope fragments. Our findings indicate that nuclear and endoplasmic reticulum membranes which are known to be in morphological continuity have, besides a far-reaching similarity, some characteristic differences.
