Journal ArticleDOI
Development of a synthetic live bacterial therapeutic for the human metabolic disease phenylketonuria
Vincent M. Isabella,Binh Ha,Mary Castillo,David Lubkowicz,Sarah Rowe,Yves Millet,Cami Leigh Anderson,Ning Li,Adam B. Fisher,Kip A. West,Philippa J. Reeder,Munira Momin,Christopher Bergeron,Sarah E. Guilmain,Paul F. Miller,Caroline B. Kurtz,Dean Falb +16 more
TL;DR: In mice and primates, Phe was converted to trans-cinnamate by SYNB1618, quantitatively metabolized by the host to hippurate and excreted in the urine, acting as a predictive biomarker for strain activity and defining a strategy for translation of live bacterial therapeutics to treat metabolic disorders.
Journal ArticleDOI
Insulin demand regulates β cell number via the unfolded protein response
Rohit Sharma,Amy C. O’Donnell,Rachel E. Stamateris,Binh Ha,Karen M. McCloskey,Paul R. Reynolds,Peter Arvan,Laura C. Alonso +7 more
TL;DR: A stem cell-independent model of tissue homeostasis is defined, in which differentiated secretory cells use the UPR sensor to adapt organ size to meet demand, suggesting that therapeutic UPR modulation has potential to expand β cell mass in people at risk for diabetes.
